Description:   NOSTRIN (nitric oxide synthase trafficker isoform 1), also known as endothelial nitric oxide synthase traffic inducer, is a member of the Pombe Cdc15 homology (PCH) family of proteins. NOSTRIN is expressed in the vascular endothelial cells of highly vascularized tissues such as placenta, lung, kidney and heart. It consists of an N-terminal Cdc15 domain with an FCH (Fes/CIP homology) region, two coiled coil domains and a C-terminal SH3 domain. NOSTRIN typically exists as a trimer. It functions as an adaptor protein binding to caveolin-1 via an internal domain and NOS3 via its SH3 domain, forming a ternary complex which facilitates caveolar transport of NOS3. The NOS3 protein is responsible for the production of nitric oxide (NO), a potent mediator in various biological processes. The translocation of NOS3 from the plasma membrane to intracellular vesicle-like structures diminishes NO production. NOSTRIN also interacts with Dynamin and N-WASP via its SH3 domain.

Description:   C17orf57 is a 973 amino acid protein that is encoded by a gene mapping to human chromosome 17. Chromosome 17 makes up over 2.5% of the human genome with about 81 million bases encoding over 1200 genes. Two key tumour suppressor genes are associated with chromosome 17, namely, p53 and BRCA1. tumour suppressor p53 is necessary for maintenance of cellular genetic integrity by moderating cell fate through DNA repair versus cell death. Malfunction or loss of p53 expression is associated with malignant cell growth and Li-Fraumeni syndrome. Like p53, BRCA1 is directly involved in DNA repair, specifically it is recognised as a genetic determinant of early onset breast cancer and predisposition to cancers of the ovary, colon, prostate gland and fallopian tubes. Chromosome 17 is also linked to neurofibromatosis, a condition characterised by neural and epidermal lesions, and dysregulated Schwann cell growth. Alexander disease, Birt-Hogg-Dube syndrome and Canavan disease are also associated with chromosome 17.
