Enables the low-temperature cleavage of fusion proteins containing the PreScission Protease recognition sequence.
- Cleavage buffer 50 mM Tris-HCl, 150 mM NaCl, 1 mM EDTA, 1 mM dithiothreitol (DTT), pH 7.0
- Elution buffer 50 mM Tris-HCl, 10 mM reduced glutathione, pH 8.0
- Inhibitor 100 mM ZnCl2 (< 50% inhibition); 100 µM chymostatin; 4 mM Pefabloc
Can be used either following affinity purification or while fusion proteins are bound to Glutathione Sepharose 4B Glutathione Sepharose High Performance, or Glutathione Sepharose 4 Fast Flow.
PreScission Protease is a genetically engineered fusion protein consisting of human rhinovirus 3C protease and GST. This protease was designed to facilitate removal of the protease by allowing simultaneous protease immobilization and cleavage of GST fusion proteins produced from the pGEX-6P vectors pGEX-6P-1, pGEX-6P-2, and pGEX-6P-3; see pGEX Vectors (GST Gene Fusion System). PreScission Protease specifically cleaves between the Gln and Gly residues of the recognition sequence of LeuGluValLeuPheGln/GlyPro.
One unit will cleave > 90% of 100 µg of a test GST-fusion protein at 5°C for 16 hours.
