SATA (and SATP) are reagents for introducing protected sulfhydryls into proteins, peptides and other molecules. for covalent modification of primary amines and addition of a protected sulphhydryl group.
- Adds a protected sulfhydryl that can be deprotected by hydroxylamine
- Allows long-term storage of the sulfhydryl-modified molecule
- Offers more steric freedom than SATA for the unmasked sulfhydryl group
- Forms cleavable disulfide bonds with other sulfhydryl-containing molecules
- Reacts with primary amines (eg. lysine residues proteins) to form stable amide bonds
- Preserves protein activity with its mild, non-denaturing reaction cdonditions
SATA (and SATP) are the N-hydroxysuccinimide (NHS) esters of S-acetylthioacetic and propionic acid. A stable, covalent amide bond is formed from the reaction of the NHS ester with primary amines. The amine reacts with the NHS ester by nucleophilic attack, with N-hydroxysuccinimide being released as a by-product. Deprotection (deacylation) to generate a sulfhydryl for use in cross-linking and other applications is accomplished using hydroxylamine·HCl.
