Neurexins comprise a family of neuronal cell surface proteins, which include neurexin I (NRXN1), neurexin II (NRXN2), neurexin III (NRXN3) and Caspr (neurexin IV). Neurexins I-III are expressed as a and b isoforms. The a isoforms are made of three cassettes, which contain two LNS (Laminin A, Neurexins, Sex hormone-binding)-domains separated by EGF domains, followed by a transmembrane region and a 55 amino acid cytoplasmic C-terminal. The a isoforms bind to neurexophilins at the second LNS site and to the excitatory neurotoxin a-latrotoxin. The b isoforms have only one LNS-domain, bind to neuroligins, and play a role in the formation and remodeling of synapes. Caspr (for Contactin-Associated Protein 1, also designated Paranodin in mouse), contains an extracellular domain similar to the other three neurexins, and binds to the surface glycoprotein Contactin. Caspr and the closely related Caspr2, a mammalian homolog of Drosophila Neurexin IV (Nrx-IV), demarcate distinct subdomains in myelinated axons. Specifically, Caspr exists at the paranodal junctions, while Caspr2 colocalizes with Shaker-like K+ channels in the juxtaparanodal region. Caspr may play a role in the communication of glial cells and neurons during development.

Chromosome 9 consists of about 145 million bases and 4% of the human genome and encodes nearly 900 genes. Considered to play a role in gender determination, deletion of the distal portion of 9p can lead to development of male to female sex reversal, the phenotype of a female with a male X,Y genotype. Hereditary hemorrhagic telangiectasia, which is characterised by harmful vascular defects, is associated with the chromosome 9 gene encoding endoglin protein, ENG. Familial dysautonomia is also associated with chromosome 9 though through the gene IKBKAP. Notably, chromosome 9 encompasses the largest interferon family gene cluster. Chromosome 9 is partnered with chromosome 22 in the translocation leading to the aberrant production of BCR-ABL fusion protein often found in leukemias. The C9orf78 gene product has been provisionally designated C9orf78 pending further characterisation.

Glutamate receptors mediate most excitatory neurotransmission in the brain and play an important role in neural plasticity, neural development and neurodegeneration. Ionotropic glutamate receptors are categorized into NMDA receptors and kainate/AMPA receptors, both of which contain glutamate-gated, cation-specific ion channels. Kainate/AMPA receptors are co-localized with NMDA receptors in many synapses and consist of the structurally related subunits GluR-1 to -7, KA1 and KA2. KA1 (also designated EEA1) and KA2 (also designated EEA2) form heteromeric receptors with GluR subunits when coexpressed, forming ion channels with various properties. The kainate/AMPA receptors are primarily responsible for the fast excitatory neuro-transmission by glutamate.

Annexins bind to phospholipids and may be involved in regulation of membrane transport, membrane channel activity, and interaction of the cell membrane with the extracellular matrix. Annexin A7 is a member of the annexin family of calcium dependent phospholipid binding proteins. The Annexin A7 gene contains 14 exons and spans approximately 34 kb of DNA. Structural analysis of the protein suggests that Annexin A7 is a membrane binding protein with diverse properties including voltage sensitive calcium channel activity, ion selectivity and membrane fusion.

The Hox proteins play a role in patterns of embryonic development and cellular differentiation by regulating downstream target genes. In vivo, the HoxD9 protein interacts with the autoregulatory and cross-regulatory enhancers of the murine HoxB1 and human HoxD9 genes. Specifically, the HoxD9 protein interacts with the human control region (HCR) of the HoxD9 gene, thus inducing transcription of the HoxD9 promoter. HoxD9 may be a multifunctional transcriptional regulator, as it contains different activation domains. Activation of HoxD9 depends on the structure of the target regulatory element, and results in differential cofactor interaction. The HoxD9 protein is expressed in the early stages of mouse joint development, primarily in the articular cartilage. HoxD9 transcripts are also detected in the synovial tissue of arthritic mice, but not in that of normal mice, suggesting that HoxD9 may have a role in the pathology of arthritis. Furthermore, the HoxD9 protein is highly expressed in the synoviocytes of patients with rheumatoid arthritis (RA), but not in osteoarthritis patients. The human HoxD9 protein is also differentially expressed in the human cervical cancer cell line HeLa, but is not expressed in the normal cervix and may thus play a role in tumorigenesis.
