Description:
Clusterin shares homology with the small heat shock protein family of molecular chaperones. The mature secreted form of the protein is a glycosylated, 80-kDa disulphide-linked heterodimer of alpha and beta subunits (produced by internal cleavage). Clusterin is expressed in virtually all tissues and found in all human fluids. It is involved in numerous physiological processes important for carcinogenesis and tumour growth, including apoptotic cell death, cell cycle regulation, DNA repair, cell adhesion, tissue remodelling, lipid transportation, membrane recycling, and immune system regulation. Clusterin also exists as a nuclear protein. The secreted form of Clusterin has extracellular chaperone and anti-apoptotic activities while the nuclear form acts as a proapoptotic factor.

Retinol binding protein 4 (RBP4; RBP) is a 21kDa secreted protein, a member of the lipocalin family and is known as the primary transporter of retinol (vitamin A) to tissues. A recent report revealed RBP4 as an adipokine linking glucose transporter 4 (GLUT4) suppression in adipose tissue to insulin. Elevated human and mouse serum RBP4 levels are associated with insulin resistance and its severity, obesity, and certain components of metabolic syndrome. Furthermore, human serum RBP4 levels are closely related to renal function.

RRM2 supplies the precursors essential for DNA synthesis. RRM2 catalyzes the biosynthesis of deoxyribonucleotides from the corresponding ribonucleotides. Phosphorylation on Ser-20 relieves the inhibitory effect on Wnt signaling.

Human Fractalkine (CX3CL1) is a member of the CX3C family of chemokines. Human Fractalkine contains both chemokine and mucin domain. The soluble form of Fractalkine is chemotactic for T-cells and monocytes, but not for neutrophils. The membrane bound form of Fractalkine promotes leukocytes adhesion to endothelial cells. Fractalkine regulates leukocyte adhesion and migration processes at the endothelium and binds to CX3CR1. Natural Human Fractalkine is produced as a long protein (373-amino acid). The mucin-like stalk permits it to bind to the cell surface.

Dipeptidyl peptidase-IV (DPPIV) is also known as adenosine deaminase complexing protein 2, DPPIV or CD26 is antigenic enzyme expressed on the surface of most cell types and is associated with immune regulation, signal transduction and apoptosis. It is an intrinsic membrane glycoprotein and a serine exopeptidase that cleaves X-proline dipeptides from the N-terminus of polypeptides. The substrates of DPPIV are proline (or alanine)-containing peptides and include growth factors, chemokines, neuropeptides, and vasoactive peptides. DPPIV plays a major role in glucose metabolism. It is responsible for the degradation of incretins such as GLP-1. DPPIV plays an important role in tumor biology, and is useful as a marker for various cancers, with its levels either on the cell surface or in the serum increased in some neoplasms and decreased in others.[4] DPPIV also binds the enzyme adenosine deaminase specifically and with high affinity. The significance of this interaction has yet to be established.

Glial Cell Line-Derived Neurotrophic Factor Family Receptor alpha-1 (GDNFR alpha1) is a glycosylphosphatidylinositol (GPI) linked cell surface protein belonging to GDNF-family receptor alpha subtype which consists of at least four members. GFR alpha1and GFR alpha2 are the cognate co-receptor for the neurotrophic factor neurturin mediating the NRTN-induced autophosphorylation and activation of the RET tyrosine kinase receptor. Soluble GFR alphas released enzymatically from the cell surface by phosphatidylinositol phospholipase C, as well as recombinantly produced soluble GFR alpha1, can also bind with high affinity to GDNF and trigger the activation of Ret tyrosine kinase. Human GFR alpha1 shares 93% amino acid identity with mouse GFR alpha1.The expression of the various GFR alphas are differentially regulated in the central and peripheral nervous system, suggesting complementary roles for the GFR alphas in mediating the activities of the GDNF family of neurotrophic factors.

TNFRSF11B is a secreted protein, containing 2 death domains and 4 TNFR-Cys repeats. TNFRSF11B is a decoy receptor for the receptor activator of nuclear factor kappa B ligand (RANKL). By binding RANKL, TNFRSF11B inhibits nuclear kappa B (NF-κB) which is a central and rapid acting transcription factor for immune-related genes, and a key regulator of inflammation, innate immunity, and cell survival and differentiation. TNFRSF11B levels are influenced by voltage-dependent calcium channelsCav1.2. TNFRSF11B can reduce the production of osteoclasts by inhibiting the differentiation of osteoclast precursors (osteoclasts are related to monocytes/macrophages and are derived from granulocyte/macrophage-forming colony units (CFU-GM)) into osteoclasts and also regulates the resorption of osteoclasts in vitroand in vivo. TNFRSF11B binding to RANKL on osteoblast/stromal cells, blocks the RANKL-RANK ligand interaction between osteoblast/stromal cells and osteoclast precursors. This has the effect of inhibiting the differentiation of the osteoclast precursor into a mature osteoclast.

TNFRSF10B is a member of the TNF-receptor superfamily, and contains an intracellular death domain. This receptor can be activated by tumor necrosis factor-related apoptosis inducing ligand (TNFSF10/TRAIL/APO-2L), and transduces apoptosis signal. The adapter molecule FADD recruits caspase-8 to the activated receptor and is required for the apoptosis mediated by TNFRSF10B. TNFRSF10B is expressed in a number of cell types, and to particularly high levels in lymphocytes and spleen. This single-pass transmembrane protein contains two cysteine-rich repeat units in its extracellular region, followed by a transmembrane segment and a cytoplasmic tail containing a typical “death domain”. TNFRSF10B expression is regulated by the tumor suppressor p53. It is also indicated that the activation of NF-kappa-B can be promoted by TNFRSF10B.

It binds to ligand CD70, and plays a key role in regulating B-cell activation and immunoglobulin synthesis. Mouse Ifng is a secreted protein which belongs to the type I I (or gamma) interferon family. IFNG is produced by lymphocytes and activated by specific antigens or mitogens. In addition to having antiviral activity, IFNG also has important immunoregulatory functions. It is a potent activator of macrophages and has antiproliferative effects on transformed cells. It can potentiate the antiviral and antitumor effects of the type I interferons. Genetic variation in IFNG is associated with the risk of aplastic anemia (AA) which is a rare disease in which the reduction of the circulating blood cells results from damage to the stem cell pool in bone marrow. In most patients, the stem cell lesion is caused by an autoimmune attack. T-lymphocytes, activated by an endogenous or exogenous, and most often unknown antigenic stimulus, secrete cytokines, including IFN-gamma, which would in turn be able to suppress hematopoiesis.

UBE2T accepts the ATP-dependent ubiquitin from the E1 complex and catalyzes its covalent attachment to other proteins. In vitro, UBE2T is able to catalyze polyubiquitination using all 7 ubiquitin Lys residues, but may prefer 'Lys-11'-, 'Lys-27'-, 'Lys-48'- and 'Lys-63'-linked polyubiquitination. They catalyze the hydrolysis of amides, thioesters and esters, peptide and isopeptide bonds formed by the C-terminal Gly of ubiquitin. Up regulation of UCHL3 is associated with uterine cervical neoplasms. UCHL3 is implicated in age related cognitive disorders. UCHL3 also promotes adipogenesis and insulin signaling. In mice, UCHL3 knockout have been shown to be resistant to diet-induced obesity.
