End-point PCR Enzymes and Kits
Direct PCR Kits save time and cost by allowing amplification of DNA directly from the source material. Direct PCR is based on Thermo Scientific Phire® Hot Start II DNA Polymerase and Phusion® Hot Start II High-Fidelity DNA Polymerase. These fusion enzymes are exceptionally fast, robust and highly tolerant of many PCR inhibitors present in unpurified sample material. This allows reliable amplification in challenging conditions where conventional DNA polymerases are completely inhibited.

PCR directly from plants
Phire® Plant Direct PCR Kit is designed to amplify DNA directly from plant samples. It has been tested with leaves and seeds from a wide variety of plant species. The kit includes a complete set of optimised reagents, sampling tools and detailed protocols, making it an easy and convenient choice for amplification of plant DNA.
Each kit includes Phire® Hot Start II DNA Polymerase, 200 µl; 2X Phire® Plant PCR Buffer (includes dNTPs and MgCl₂), 5×1 ml; control primer mix (25 µM each), 40 µl; and dilution buffer, 5 ml.
PCR directly from animal tissues
Phire® Animal Tissue Direct PCR Kit has been developed for amplification of DNA directly from a wide variety of animal tissues including mice, fish, birds and insects. The kit contains all the necessary components for amplification of DNA directly from animal tissues: optimised reagents, sampling tools and Thermo Scientific DNARelease™ additive, which can be used to improve the release of DNA from animal tissues. In addition, the kit includes detailed instructions and control primers that can be used with numerous animal species.
Each kit includes Phire® Hot Start II DNA Polymerase, 200 µl; 2X Phire® Animal Tissue PCR Buffer (includes dNTPs and MgCl₂), 5×1 ml; universal control primer mix (25 µM each), 40 µl; dilution buffer, 5 ml; DNARelease™ additive, 3×100 µl; and gel loading dye, 3×1 ml.
PCR directly from human sample types
Phusion® Human Specimen Direct PCR Kit amplifies DNA directly from unpurified human samples. The kit is compatible with fresh, frozen, and even formalin-fixed, paraffin embedded (FFPE) human tissue samples. It includes optimised reagents and detailed protocols for several sample materials. Modified Phusion® Hot Start II High-Fidelity DNA Polymerase utilised in the kit is a highly accurate proofreading DNA polymerase. PCR products obtained are perfectly suitable even for cloning purposes. The kit is for research use only.
Each kit includes Phusion® Human Specimen DNA Polymerase, 80 μl; 2X Phusion® Human Specimen PCR Buffer (includes dNTPs and MgCl₂), 2×1 ml; universal control primer mix (25 μM each), 40 μl; dilution buffer, 2×5 ml; and DNARelease™ additive, 3×100 μl.
PCR directly from whole blood
Phusion® Blood Direct PCR Kit is designed for amplification of DNA from whole blood. The Phusion® Hot Start II High-Fidelity DNA Polymerase is resistant to PCR inhibitors present in blood and retains its activity even with samples containing 40% whole blood. The kit tolerates heparin, citrate and EDTA, and is compatible with frozen blood or blood stored on various types of filter papers. The Phusion® Blood Direct PCR Kit an ideal choice even for demanding applications, such as cloning.
Each F-747S kit includes Phusion® Blood II DNA Polymerase, 40 μl; 2X Phusion® Blood PCR Buffer (includes dNTPs and MgCl₂), 1 ml; universal control primer mix (25 mM each), 40 μl; 50 mM EDTA (pH 8.0), 0.5 ml; 50 mM MgCl₂ solution, 1.5 ml; and 100% DMSO, 0.5 ml.
