GARNL1 localizes to the cytoplasm where it may play a role regulating GTP hydrolysis of proteins such as Ran and Rap. GARNL1 is imported to the nucleus via dimerization with E12. GARNL1 interacts with the HLH region of E12 and may function to negatively regulate the transcription of E12-dependent downstream target genes. This suggests that at least a portion of the function of GARNL1 is dependent upon its association with E12. GARNL1 may also associate with other HLH proteins and influence a variety of HLH signaling cascades. In adult brain, GARNL1 activity does not involve E12 and therefore it may serve a different function in developed neural tissue.

Description:   Lissencephaly (smooth brain) is an abnormality of brain development characterized by incomplete neuronal migration and a smooth cerebral surface, manifesting as severe mental retardation. Genetic analysis has identified two proteins that are mutated in some cases of lissencephaly, designated lissencephaly-1 protein (LIS1) and doublecortin. LIS1 displays sequence homology to ?subunits of heterotrimeric G proteins, and doublecortin contains a consensus Abl phosphorylation site. In addition, the DCAMKL1 (doublecortin-like and CAM kinase-like 1) protein shows homology to doublecortin. All three proteins are highly expressed in developing brain and may function together to regulate microtubules involved in neuronal migration. The DCAMKL1 protein encodes a functional kinase that is capable of phosphorylating myelin basic protein and itself, but its kinase activity does not appear to affect its microtubule polymerization activity.

Description:   Three highly similar proteins termed NOMO1, NOMO2 and NOMO3, are encoded by a gene mapping to a region of duplication on the p arm of human chromosome 16. All three NOMO proteins share similar functions and have been difficult to characterize individually. NOMO1 (Nodal modulator 1), also known as PM5, is a 1,222 amino acid highly conserved single-pass type I membrane protein expressed in colon tumor tissue and normal colonic mucosa. NOMO proteins are novel antagonists of Nodal signaling which interact with Nicalin to form a Nicalin-NOMO complex, and are rapidly degraded or stabilized by Nicalin. NOMO proteins were once considered candidates for the development of pseudoxanthoma elasticum (PXE), a heritable disorder of connective tissue, as the NOMO genes are located in close proximity to the gene responsible for PXE development (MRP6).

Description:   Calcium-dependent secretion activators (CAPS-1 and CAPS-2) are calcium-binding proteins that direct neurotransmitter and neuropeptide-filled vesicles to the cell membrane for secretory granule exocytosis. Both CAPS-1 and CAPS-2 are expressed primarily in the brain where they regulate the secretion of various substances. The CAPS proteins contain a PH domain that is essential for regulation of exocytosis, as well as regulation of phospholipid binding. Through their regulation of neurotrophin release from granule cells, CAPS proteins help to regulate cell fate during neuronal development. CAPS-1 is thought to regulate catecholamine release from neuronal cells, while CAPS-2 is thought to regulate release of both brain-derived neurotrophic factor and neurotrophin-3 from granule cells.
