SSEA-4 (Stage-specific embryonic antigen 4) is a glycoprotein expressed early in embryonic development and in pluripotent stem cells. SSEA-4 can be used as a marker of Human Embryonic Stem Cells, Human Embryonic Carcinoma Cells and Human Embryonic Germ Cells. Monoclonal antibodies to this target have been widely used in the characterization of pluripotent stem cells. Mouse pluripotent stem cells are not recognised by anti-SSEA-4 antibodies but do express the antigen upon differentiation.

The human Mel-CAM gene maps to chromosome 11q23 and encodes a trans-membrane glycoprotein, also designated MCAM, MUC 18 or CD146, that belongs to the immunoglobulin superfamily and functions as a Ca2 -independent cell adhesion molecule. Mel-CAM expression is restricted to advanced primary and metastatic melanomas and to cell lines of the neuroectodermal lineage, but not normal melanocytes. Mel-CAM is found on 80% of advanced primary human melanomas and correlates well with development of metastatic disease.

It may be involved in the regulation of mitogenic signalling in vascular smooth muscles cells. Several alternatively spliced transcript variants of this gene have been described, but the full length nature of some of these variants has not been determined. Expressed only in brain and heart. Isoform 1 and isoform 2 are only expressed in brain. Isoform 3 is expressed in both heart and brain. Belongs to the NDRG family.
