Description:
CBF binds to the core site, 5'-PYGPYGGT-3', of a number of enhancers and promoters, including murine leukemia virus, polyomavirus enhancer, T-cell receptor enhancers, LCK, IL-3 and GM-CSF promoters. The alpha subunit binds DNA and appears to have a role in the development of normal hematopoiesis. Isoform AML-1L interferes with the transactivation activity of RUNX1. Acts synergistically with ELF4 to transactivate the IL-3 promoter and with ELF2 to transactivate the mouse BLK promoter. Inhibits KAT6B-dependent transcriptional activation. Controls the anergy and suppressive function of regulatory T-cells (Treg) by associating with FOXP3. Neuronal migration of the central nervous system is a specialized form of cell motility that takes place in the laminar structure of the cortical regions of brain. Astrotactin is a neuronal cell surface protein expressed on postmitotic neuronal precursors in the cerebellum, hippocampus, cerebrum, and olfactory bulb. Astrotactin 2 is a paralog of Astrotactin. Astrotactin mediates neuron-astroglial interactions and is also implicated in synaptic development as well as many other neuronal activities. Astrotactin has three epidermal growth factor repeat domains and two fibronectin type III repeat domains. The human Astrotactin gene shows extensive homology to the mouse Astrotactin gene. Mutations in the Astrotactin gene are linked to neuronal migration defects in both species.

Aladin (Adracalin) belongs to a family of WD repeat-containing proteins. These proteins have a wide variety of functions, including signal transduction regulation, RNA processing and transcription. Aladin plays a role in peripheral and central nervous system development. It is widely expressed, with the highest expression seen in pituitary gland, corpus callosum, cerebellum, adrenal gland and gastrointestinal structures. Defects in Aladin cause the autosomal recessive disorder achalasia-addisonianism-alacrima (triple A) syndrome. Triple A syndrome is characterized by achalasia, alacrima and adrenocortico-tropin-resistant adrenal insufficiency. Robust expression in neural systems associated with cognitive, motor and sensory functions is consistent with the myriad of symptoms experienced by patients with triple A syndrome.

This protein bridges B cell receptor-associated kinase activation with downstream signaling pathways, thereby affecting various biological functions. The phosphorylation of five tyrosine residues is necessary for this protein to nucleate distinct signaling effectors following B cell receptor activation. Mutations in this gene cause hypoglobulinemia and absent B cells, a disease in which the pro- to pre-B-cell transition is developmentally blocked. Deficiency in this protein has also been shown in some cases of pre-B acute lymphoblastic leukemia. Alternatively spliced transcript variants have been found for this gene. Human Pecanex proteins are homologs of the Drosophila Pecanex protein, a maternal-effect neurogenic protein that is involved in normal development of the fly nervous system. Three human Pecanex homologs exist, designated Pecanex, Pecanex 2, also known as PCNXL2 (pecanex-like 2), and Pecanex 3. Pecanex 2 is a 2,137 amino acid multi-pass membrane protein that exists as five alternatively spliced isoforms characterized by high mutational frequencies and biallelic mutations in colorectal tumors, thereby likely functioning as a target gene in these tumors. Pecanex 2 is encoded by a gene that maps to human chromosome 1q42.2, which is linked to an inherited microduplication. This microduplication that includes Pecanex 2 may play a role in autism and mild mental retardation.
