Description:   The protein encoded by this gene is a member of the MAP kinase family. MAP kinases act as an integration point for multiple biochemical signals, and are involved in a wide variety of cellular processes such as proliferation, differentiation, transcription regulation and development. This kinase is activated by various environmental stresses and proinflammatory cytokines. The activation requires its phosphorylation by MAP kinase kinases (MKKs), or its autophosphorylation triggered by the interaction of MAP3K7IP1/TAB1 protein with this kinase. The substrates of this kinase include transcription regulator ATF2, MEF2C, and MAX, cell cycle regulator CDC25B, and tumor suppressor p53, which suggest the roles of this kinase in stress related transcription and cell cycle regulation, as well as in genotoxic stress response. Four alternatively spliced transcript variants of this gene encoding distinct isoforms have been reported. [provided by RefSeq, Jul 2008]

Description:   The RING-type zinc finger motif is present in a number of viral and eukaryotic proteins and is made of a conserved cysteine-rich domain that is able to bind two zinc atoms. Proteins that contain this conserved domain are generally involved in the ubiquitination pathway of protein degradation. RNF185 (ring finger protein 185), also known as FLJ38628, is a 192 amino acid multi-pass membrane protein containing one RING-type zinc finger. Two RNF185 isoforms exist as a result of alternative splicing, and the gene encoding RNF185 maps to human chromosome 22, which houses over 500 genes and is the second smallest human chromosome. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, Neurofibromatosis type 2, autism and schizophrenia.

Description:   The MAGUK (membrane-associated guanylate kinase homologs) family of proteins contain multiple protein-binding domains and are involved in cell junction organization, tumor suppression, and signaling. CASK (also designated LIN-2) belongs to a MAGUK subfamily which is characterized by a novel domain structure that consists of a calcium/calmodulin- dependent protein kinase domain followed by PDZ, SH3 and guanylate kinase-like (GUK) domains. CASK is expressed in rat brain where it binds to cell-surface proteins, such as neurexin and syndecan, and is thought to be involved in signaling at neuronal synapses. CASK translocates to the nucleus and interacts with Tbr-1 to form a complex, which binds to a specific DNA sequence (the T-element), and induces the expression of specific genes, including Reelin. CASK displays a transcription regulation function, which appears crucial for cerebrocortical development.

Description:   The protein encoded by this gene is a member of the keratin family. The keratins are intermediate filament proteins responsible for the structural integrity of epithelial cells and are subdivided into cytokeratins and hair keratins. The type I cytokeratins consist of acidic proteins which are arranged in pairs of heterotypic keratin chains. Unlike its related family members, this smallest known acidic cytokeratin is not paired with a basic cytokeratin in epithelial cells. It is specifically expressed in the periderm, the transiently superficial layer that envelopes the developing epidermis. The type I cytokeratins are clustered in a region of chromosome 17q12-q21.<be>This gene encodes the type I intermediate filament chain keratin 17, expressed in nail bed, hair follicle, sebaceous glands, and other epidermal appendages. Mutations in this gene lead to Jackson-Lawler type pachyonychia congenita and steatocystoma multiplex. [provided by RefSeq, Aug 2008].

Description:   Cell adhesion molecules (CAMs) influence cell growth, differentiation, embryogenesis, immune response and cancer metastasis by networking information from the extracellular matrix to the cell. The four major families of cell adhesion molecules are immunoglobulin (Ig) superfamily (calcium-independent transmembrane glycoproteins), integrins (transmembrane non-covalently linked heterodimers of Alpha and Beta subunits), calcium-dependent cadherins and divalent cation-dependent selectins. Regulation of neuronal synaptic adhesion by CAMs has proven important for learning and memory. Proper embryonic morphogenic development is also heavily dependent on the regulation of cell adhesion molecules. Neurotrimin (hNT) is a neural cell adhesion molecule localizing to the cell membrane, where it acts as a lipid-anchor. Neurotrimin belongs to the IgLON family of proteins, a member of the larger immunoglobulin superfamily.

Description:   The cystatin superfamily is a well-established family of cysteine protease inhibitors. All true cystatins inhibit cysteine peptidases of the papain family, such as cathepsins, while some also inhibit legumain family enzymes. The CRES (cystatin-related epididymal spermatogenic) protein defines a new subgroup in the family 2 cystatins of the cystatin superfamily. CRES proteins lack two of the three consensus sites necessary for the cystatin inhibition of C1 cysteine proteases. They are also preferentially expressed in postmeiotic germ cells, the proximal caput epididymidis, and anterior pituitary gonadotrophs. Therefore, CRES proteins may perform unique and tissue-specific functions in the reproductive and neuroendocrine systems. As a member of the CRES subfamily, Cystatin 8 is a 142 amino acid protein that is expressed in the proximal caput region of the epididymis, where it performs a specialized role during sperm development and maturation.

Description:   Semaphorins are a family of cell surface and secreted proteins involved in neural development that are conserved from insects to humans. Members of this family are approximately 750 amino acids in length (including signal sequences) and are defined by a conserved extracellular “semaphorin” domain of approximately 500 amino acids containing 14-16 cysteines, blocks of conserved sequences and no obvious repeats. The transmembrane semaphorins are characterized by an additional 80 amino acid transmembrane domain and an 80-110 amino acid cytoplasmic domain. SEMA6C, also known as SEMA Y, is a transmembrane protein expressed in fetal brain and adult skeletal muscle. Three isoforms of this semaphorin exist due to alternative splicing: SEMA6C 1, SEMA6C 2 and SEMA6C 3. The extracellular domain of SEMA6C induces growth cone collapse of dorsal root ganglion and plays a role in generation or stability of entorhino-hippocampal synapses.

Description:   Apoptosis, or programmed cell death, occurs during normal cellular differentiation and development of multicellular organisms. Apoptosis is induced by certain cytokines including TNF and Fas ligand of the TNF family through their death domain containing receptors, TNFR1 and Fas. Cell death signals are transduced by death domain (DD) containing adapter molecules and members of the ICE/CED3 protease family. A novel DD containing molecule was recently cloned from mouse, human and monkey and designated Daxx. Daxx is a death domain containing important intermediate in the Fas mediated apoptosis. Daxx binds specifically to the Fas death domain and enhances Fas induced apoptosis and activates the Jun N terminal kinase (JNK) pathway. It is widely expressed in fetal and adult human and mouse tissue, indicating its important function in Fas signaling pathways.

Description:   Cysteine-rich secretory proteins (CRISPs) represent a family of evolutionarily conserved proteins that may play a role in the innate immune system and are transcriptionally regulated by androgens in several tissues. GAPR-1 (Golgi-associated plant pathogenesis-related protein 1), also known as GLIPR2, is a 154 amino acid lipid anchor protein belonging to the CRISP family. GAPR-1 also shares similarity with the pathogenesis-related protein (PR) superfamily, and may play an important role in the immune system. Existing as a homodimer, GAPR-1 is highly expressed in lung and peripheral leukocytes with minor expression in liver and kidney. Containing a conserved sperm-coating protein (SCP) domain, GAPR-1 binds to negatively charged lipids and may be involved in the differentiation of epithelial cells into mesenchymal cells. Increased expression of GAPR-1 in kidney may contribute to the development of fibrosis.

Description:   The 145-2C11 antibody is specific for mouse CD3e, also known as CD3 epsilon, a 20 kDa subunit of the T cell receptor complex, along with CD3 gamma and CD3 delta. These integral membrane protein chains assemble with additional chains of the T cell receptor (TCR), as well as CD3 zeta chain, to form the T cell receptor – CD3 complex. Together with co-receptors CD4 or CD8, the complex serves to recognize antigens bound to MHC molecules on antigen-presenting cells. Such interactions promote T cell receptor signaling (T cell activation) and can result in a number of cellular responses including proliferation, differentiation, production of cytokines or activation-induced cell death. CD3 is differentially expressed during thymocyte-to-T cell development and on all mature T cells.

Description:   The Hit8a antibody is specific for the 32-34 kDa alpha chain of human CD8, known as CD8a or CD8 alpha. CD8a can form a homodimer (CD8 alpha-alpha), but is more commonly expressed as a heterodimer with a second chain known as CD8b or CD8 beta. CD8 acts as a co-receptor for antigen recognition and subsequent T cell activation that is initiated upon binding of the T cell receptor (TCR) to antigen-bearing MHC Class I molecules. The cytoplasmic domains of CD8 provide binding sites for the tyrosine kinase lck, facilitating intracellular signaling events that lead to T cell activation, development, and cytotoxic effector functions. CD8+ cytotoxic T cells (CTLs) play an important role in inducing cell death of tumor cells, as well as cells infected by virus, bacteria or parasites.
