CD8 is a cell surface receptor expressed either as a heterodimer with the CD8 β chain (CD8 α/β) or as a homodimer (CD8 α/α). A majority of thymocytes and a subpopulation of mature T cells and NK cell express CD8a. CD8 binds to MHC class 1 and through its association with protein tyrosine kinase p56lck plays a role in T cell development and activation of mature T cells. For mature T-cells, CD4 and CD8 are mutually exclusive, so anti-CD8, generally used in conjunction with anti-CD4. It is a useful marker for distinguishing helper/inducer T-lymphocytes, and most peripheral T-cell lymphomas are CD4 /CD8-. Anaplastic large cell lymphoma is usually CD4 and CD8-, and in T-lymphoblastic lymphoma/leukemia, CD4 and CD8 are often co-expressed. CD8 is also found in littoral cell angioma of the spleen.

It is expressed in epithelial cells of the thyroid gland and the lung. Nuclei from liver, stomach, pancreas, small intestine, colon, kidney, breast, skin, testes, pituitary, prostate, and adrenal glands are unreactive. Anti-TTF-1 is useful in differentiating primary adenocarcinoma of the lung from metastatic carcinomas originating in the breast, mediastinal germ cell tumors, and malignant mesothelioma. It can also be used to differentiate small cell lung carcinoma from lymphoid infiltrates.Loss of TTF-1 expression in non-small cell lung carcinoma has been associated with aggressive behavior of such neoplasms. TTF-1 reactivity is also seen in thyroid malignancies.

This MAb reacts with the rod domain of human cytokeratin-19 (CK19), a polypeptide of 40 kDa. Its epitope maps between amino acid 312-335. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Anti-CK19 is a useful marker for detection of tumor cells in lymph nodes, peripheral blood, bone marrow and breast cancer.

This MAb reacts with MUC1. The dominant epitope of this MAb has not yet been determined. MUC1 is a large cell surface mucin glycoprotein expressed by most glandular and ductal epithelial cells and some hematopoietic cell lineages. It is expressed on most secretory epithelium, including mammary gland and some hematopoietic cells. It is expressed abundantly in lactating mammary glands and over expressed abundantly in >90% breast carcinomas and metastases. Transgenic MUC1 has been shown to associate with all four c-erbB receptors and localize with c-erbB1 (EGFR) in lactating glands. The MUC1 gene contains seven exons and produces several different alternatively spliced variants. The major expressed form of MUC1 uses all seven exons and is a type 1 transmembrane protein with a large extracellular tandem repeat domain. The tandem repeat domain is highly O glycosylated and alterations in glycosylation have been shown in epithelial cancer cells. Antibody to EMA is useful as a pan-epithelial marker for detecting early metastatic loci of carcinoma in bone marrow or liver.

This product is prepared by labeling highly cross-adsorbed donkey anti-goat IgG (H+L) with CF™405S. CF™405S is a blue fluorescent dye with an absorption peak wavelength that nearly coincides with the 405 nm blue diode laser line. In addition, the emission peak wavelength of the dye well centers within the blue detection window of BD flow cytometers. As a result, flow cytometry analysis using CF™405S dye results in much brighter signal than that using Alexa Fluor® 405. To minimize cross reactivity, the antibody has been adsorbed against chicken, guinea pig, Syrian hamster, horse, human, mouse, rabbit, and rat serum proteins.
