Description:
Anti-PNL2 is a novel monoclonal antibody, which has recently been introduced as an immunohistochemical reagent to stain melanocytes and tumors derived therefrom. The antigen recognized by PNL2 is different from Melan A and gp100. Its epitope is not destroyed by digestion with neuraminidase i.e. its epitope id not glycosylated. Anti-PNL2 may be most useful because of its high sensitivity for metastatic melanoma (87%), as opposed to 76% for anti-HMB45 and 82% for anti-MART-1. Anti-PNL2 labels intra-epidermal nevi while the dermal component of compound nevi are largely non-reactive with anti-PNL2. Antibodies against PNL2, MART-1 (Melan A) and HMB45 stain most clear cell sarcoma cells and a few cells in angio-myolipomas and lymphangioleiomyomatosis. Anti-PNL2 is a useful antibody for the identification of melanomas and clear cell sarcomas. Differential diagnosis is aided by the results from a panel of antibodies, including antibodies against HMB45, MART-1, tyrosinase, and MiTF.

Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). CK8 is primarily found in the non-squamous epithelia and is present in majority of adenocarcinomas and ductal carcinomas. It is absent in squamous cell carcinomas. Hepatocellular carcinomas are defined by the use of antibodies that recognize only cytokeratin 8 and 18. CK8 exists on several types of normal and neoplastic epithelia, including many ductal and glandular epithelia such as colon, stomach, small intestine, trachea, and esophagus as well as in transitional epithelium. Anti-CK8 does not react with skeletal muscle or nerve cells. Epithelioid sarcoma, chordoma, and adamantinoma show strong positivity corresponding to that of simple epithelia (with antibodies against CK8, CK18 and CK19). The RM4-5 antibody reacts with mouse CD4, a 55 kDa protein which acts as a co-receptor for the T cell receptor (TCR) in its interaction with MHC Class II molecules on antigen-presenting cells. The extracellular domain of CD4 binds to the beta2-domain of MHC Class II, while its cytoplasmic tail provides a binding site for the tyrosine kinase lck, facilitating the signaling cascade that initiates T cell activation. CD4 is typically expressed on thymocytes, certain mature T cell populations such as Th17 and T regulatory (Treg) cells, as well as on dendritic cells. The RM4-5 antibody is widely used as a phenotypic marker for CD4 expression. If used together, the RM4-5 antibody and an alternative antibody, Anti-Mouse CD4 clone GK1.5, will compete for binding, i.e. RM4-5 antibody is able to block GK1.5 antibody binding to cells. In contrast, RM4-5 antibody does not block the binding of Anti-Mouse CD4 clone RM4-4 to cells.

Penicillin refers to any member of beta-lactam antibiotics group. These agents are identified by a beta-lactam ring within their molecular structure. As the most widely used group of antibiotics available, beta-lactams are used for the treatment of bacterial infections usually caused by gram-positive organisms. Beta-lactam antibiotics are bactericidal, functioning to inhibit the synthesis of the peptidoglycan layer of bacterial cell walls. Bacterial penicillin-binding proteins and beta-lactamases constitute a large family of serine proteases that perform essential functions in the synthesis and maintenance of peptidoglycan cell wall. Notably, beta-lactamases cleave beta-lactams, therefore providing the bacteria with resistance to the antibiotic. Homologues of beta-lactamases occur in many species, including human, rat, cow, rabbit, pig, xenopus, zebrafish, and C. elegans. The human homologues, LACTB and LACTB2, are active-site-serine enzymes thought to be involved in metabolism.
