Description:   Gas2 is a 313 amino acid protein encoded by the human gene GAS2. Gas2 is thought to play a role in apoptosis by acting as a cell death substrate for caspases. Gas2, a component of the microfilament system, is cleaved by a caspase (caspase-3 and caspase-7) at Asparagine 278 during apoptosis. The cleaved form resulting from this dramatically induces the rearrangement of the Actin cytoskeleton and causes potent changes in the shape of the affected cells. Gas2 is believed to also be involved in the membrane ruffling process. During the G0-G1 transition phase Gas2 can be found phosphorylated on its serine residues. Gas2 is a cytoskeleton and peripheral membrane protein that co-localizes with Actin fibers at the cell border and along the stress fibers in growth-arrested fibroblasts. Gas2 is mainly membrane-associated but when hyperphosphorylated it will accumulate at membrane ruffles. Gas2 is specifically expressed at growth arrest and is ubiquitously expressed with highest levels found in liver, lung and kidney. There is no evidence, however, of Gas2 expression in spleen.

Description:   In eukaryotic cells, selective breakdown of cellular proteins is ensured by two distinct pathways, ubiquitination and degradation by the 26S proteasome. At specific stages of development, embryo- and tissue-specific components of the 26S proteasome are formed by developmentally regulated alternative splicing, including Rpn10a through Rpn10e (also designated pUb-R2 through pUb-R5). The pUb-R2 subunit, originally identified as S5a, is ubiquitously expressed and may perform proteolysis constitutively in a wide variety of cells. p44S10 is a highly conserved proteasome regulatory subunit that is expressed in heart, liver, skeletal muscle and pancreas. In addition to normal tissue expression, p44S10 is also expressed in several melanoma cell lines, such as MCF-7, 451Lu and WM164. Since forced expression of p44S10 in radial growth phase melanoma cells results in an increase in cellular proliferation, p44S10 may represent a potential link between regulation of proteasome activity and tumor cell proliferation <i>in vivo</i>.

Chromosome 9 consists of about 145 million bases, represents 4% of the human genome and encodes nearly 900 genes. Description:   The protein encoded by this gene is a member of the INK4 family of cyclin-dependent kinase inhibitors. This protein has been shown to form a stable complex with CDK4 or CDK6, and prevent the activation of the CDK kinases, thus function as a cell growth regulator that controls cell cycle G1 progression. The abundance of the transcript of this gene was found to oscillate in a cell-cycle dependent manner with the lowest expression at mid G1 and a maximal expression during S phase. Description:   This antibody recognizes a protein of 55-62 kDa, identified as cyclin B1. In mammals, cyclin B associates with inactive p34cdc2, which facilitates phosphorylation of p34cdc2 at aa 14Thr and 15Tyr. This maintains the inactive state until the end of G2-phase. The inactive cyclin B-p34cdc2 complex continues to accumulate in the cytoplasm until the completion of DNA synthesis, when Cdc25, a specific protein phosphatase, dephosphorylates aa 14Thr and 15Tyr of p34cdc2 rendering the complex active at the G2/M boundary. This mitotic kinase complex remains active until the metaphase/anaphase transition when cyclin B is degraded. This degradation process is ubiquitin-dependent and is necessary for the cell to exit mitosis. So, cyclin B-p34cdc2 plays a critical role in G2 to M transition.

Description:   CREB3L3 is a 461 amino acid single-pass type II membrane protein that localizes to the endoplasmic reticulum (ER) and, in response to ER stress, is cleaved and translocated to the nucleus. Expressed exclusively in liver, CREB3L3 functions as a transcription factor that, during ER stress, is thought to activate genes that are involved in both the unfolded protein response and the acute phase response (APR). Additionally, CREB3L3 is underexpressed in hepatocellular carcinoma, suggesting a possible role as a tumour suppressor. CREB3L3 functions as a dimer and contains one leucine zipper domain, a KDEL-like sequence and a bZIP domain, through which it conveys its DNA binding ability. Three isoforms of CREB3L3 exist due to alternative splicing events.
