The 17A2 monoclonal antibody specifically reacts with the mouse T lymphocytes receptor (TCR) associated CD3 complex, resulting in cellular activation and proliferation. CD3 is expressed by thymocytes and mature lymphocytes, and contains gamma, delta, and epsilon subunits, involved in the assembly, trafficking, and surface expression of T-cell receptor complex. The interaction between the T lymphocytes and the 17A2 antibody can be blocked by the anti-CD3e antibody, demonstrating that the 17A2 antibody recognizes the CD3 epsilon chain.

The LRRs and NTF2-like domains are required for export activity. NXF1 shuttles between the nucleus and the cytoplasm and binds in vivo to poly (A)+ RNA. Bcl-2-Like Protein 1 (BCL2L1) is a member of the Bcl-2 family. BCL2L1 is expressed at high levels in cells that undergo a high rate of turnover, such as developing lymphocytes. BCL2L1 is a mitochondrial membrane protein. BCL2L1 contains four motifs, BH1, BH2 and BH4. The BH4 motif is required for anti-apoptotic activity. The BH1 and BH2 motifs are required for both heterodimerization with other Bcl-2 family members and for repression of cell death. BCL2L1 regulates cell death by blocking the voltage-dependent anion channnel (VDAC) and preventing the release of the caspase activator, CYC1, from the mitochondrial membrane. In addition, BCL2L1 promotes apoptosis.

The protein arginine methyltransferases (PRMTs) include a family of proteins with related putative methyltransferase domains that modify chromatin and regulate cellular transcription. PRMT2 inhibits NF-kappaB-dependent transcription and promotes apoptosis and it exerts this effect by blocking nuclear export of IkappaB-alpha through a leptomycin-sensitive pathway, increasing nuclear IkappaB-alpha and decreasing NF-kappaB DNA binding. PRMT2 also rendered cells susceptible to apoptosis by cytokines or cytotoxic drugs, likely due to its effects on NF-kappaB.

FLIP is an apoptosis regulator protein which functions as a crucial link between cell survival and cell death pathways in mammalian cells and acts as an inhibitor of TNFRSF6 mediated apoptosis. A proteolytic fragment (p43) is likely retained in the death-inducing signaling complex (DISC) thereby blocking further recruitment and processing of caspase-8 at the complex. Full length and shorter isoforms have been shown either to induce apoptosis or to reduce TNFRSF-triggered apoptosis. FLIP lacks enzymatic (caspase) activity. FLIP is highly expressed in skeletal muscle, pancreas, heart, kidney, placenta and peripheral blood leukocytes.

PGCC1 Antibody: The transcription factor peroxisome proliferator-activated receptor gamma (PPARg) plays essential roles in adipogenesis by regulating adipocyte-specific genes through association of various co-factors. One such co-factor, PGCC1 (also known as FAM120B), is widely expressed in adult tissues and throughout embryonic development. Overexpression of this protein in OP9 pre-adipocytes promoted their differentiation into adipocytes, and knockdown of PGCC1 expression through RNA interference blocked this process. PGCC1 is homologous to C9orf10 (also known as FAM120A) and has been mapped to chromosome Xp11.22. At least two isoforms of PGCC1 are known to exist.

Description:
The correlation of anti-sperm antibodies with cases of unexplained infertility implicates a role for these antibodies in blocking fertilization. Improved diagnosis and treatment of immunologic infertility, as well as identification of proteins for targeted contraception, are dependent on the identification and characterization of relevant sperm antigens. The protein encoded by this gene is recognized by sperm agglutinating antibodies from an infertile woman. This protein is localized in germ cells of the testis at all stages of spermatogenesis and is localized to the acrosomal region of mature spermatozoa. Alternatively spliced variants that encode different protein isoforms have been described but the full-length sequences of only two have been determined.
