Description:
Differentiation of myogenic cells is regulated by multiple positively and negatively acting factors. One well characterized family of helix-loop-helix (HLH) proteins known to play an important role in the regulation of muscle cell development include Myo D, myogenin, Myf-5 and Myf-6 (also designated MRF-4 or herculin). Of interest, most muscle cells express either Myo D or Myf-5 in the committed state, but when induced to differentiate, all turn on expression of myogenin. Myo D transcription factors form heterodimers with products of a more widely expressed family of bHLH genes, the E family, which consists of at least three distinct genes: E2A, IF2 and HEB. Myo D-E heterodimers bind avidly to consensus (CANNTG) E box target sites that are functionally important elements in the upstream regulatory sequences of many muscle-specific terminal differentiation genes.

Thrombopoietin (TPO or THPO), also known as c-Mpl ligand (c-Mpl L), is a cytokine that plays a central role in megakaryopoiesis by influencing the development and maturation of megakaryocytes and platelet production from hematopoietic stem cells. TPO exerts its biological effects through the TPO receptor, c-Mpl. c-Mpl is a member of the cytokine receptor superfamily. Expression of c-Mpl is restricted to hematopoietic tissues and cells, such as bone marrow, spleen, fetal liver and CD34+ cells. Stimulation of c-Mpl with TPO results in the activation of the Janus tyrosine kinase family members, Tyk 2 and JAK2, which in turn phosphorylate Stat5 and Stat3, causing their nuclear translocation and the transcription of Stat responsive genes. Muta-tions in c-Mpl have been implicated as the cause of certain human disorders, including congenital amegakaryocytic thrombocytopenia (CAMT) and thrombocytopenia with absent radii (TAR) syndrome.

GATA1 is a Cys2/Cys2 zinc finger DNA binding protein that is expressed primarily in erythroid, megakaryocytic, mast cells and eosinophilic cells. It belongs to the GATA family of transcription factors. GATA1 is a transcriptional activator which probably serves as a general switch factor for erythroid development. It binds to DNA sites with the consensus sequence [AT]GATA[AG] within regulatory regions of globin genes and of other genes expressed in erythroid cells. The protein also plays an important role in erythroid development by regulating the switch from foetal hemoglobin production to adult hemoglobin.

IL2 Receptor beta (CD122) is a member of the immunoglobulin superfamily that forms the high affinity IL2 receptor with CD25 and CD132. This receptor chain, which is also shared by the IL15 receptor, is constitutively expressed by NK cells and at lower levels by T cells, B cells, monocytes, and macrophages. The IL2 Receptor beta chain can combine with either the common gamma subunit (gamma c) alone or the gamma c subunit and the IL2 Receptor alpha subunit to generate intermediate or high affinity IL2 receptor complexes, respectively. CD122 levels can be upregulated by activation stimuli such as IL2.

DTYMK is a 212 amino acid protein that belongs to the thymidylate kinase family and is involved in pyrimidine metabolism. Specifically, DTYMK catalyzes the ATP-dependent conversion of dTMP (deoxythymidine monophosphate) to dTDP (deoxythymidine diphosphate), which then functions as one of the four nucleotides in DNA. Via its role in the catalytic creation of dTDP, DTYMK plays an important role in the pathway of DNA synthesis and is thought to be involved in cell cycle progression and cell growth. DTYMK expression levels peak during the S phase (synthesis phase) of the cell cycle, further supporting the role of DTYMK in DNA synthesis.

Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is a cell surface glycoprotein and is similar in sequence to its family member CD53 antigen. It is known to complex with integrins and other transmembrane 4 superfamily proteins.
