Description:   The PAX5 gene is a member of the paired box (PAX) family of transcription factors. The central feature of this gene family is a novel, highly conserved DNA-binding motif, known as the paired box. The PAX proteins are important regulators in early development, and alterations in the expression of their genes are thought to contribute to neoplastic transformation. The PAX5 gene encodes the B-cell lineage specific activator protein (BSAP) that is expressed at early, but not late stages of B-cell differentiation. Its expression has also been detected in developing CNS and testis, therefore, PAX5 gene product may not only play an important role in B-cell differentiation, but also in neural development and spermatogenesis.

Description:   ATP-binding cassette (ABC) transporters are an evolutionarily conserved family of widely-expressed proteins that use ATP hydrolysis to catalyse the transport of various molecules across extracellular and intracellular membranes. As the largest family of transmembrane proteins, ABC genes comprise several subfamilies. Eukaryotic ABC transporters are largely responsible for trafficking hydrophobic compounds either within the cell, as part of a metabolic process, or outside the cell, for transport to other organs or for secretion from the body. ABCF2 in particular plays a putative role in tumour suppression at metastatic sites and in the endocrine pathway for breast cancer and may be a prognostic marker for clear cell ovarian adenocarcinoma.

Description:   RAI3 is an transmembrane G-protein-coupled receptor that affects many essential biological processes including embryogenesis, cell growth, differentiation, and apoptosis. RAI3 may also be involved in maintaining homeostasis of epithelial cells. Retinoic acid receptors directly regulate RAI3 during its transcription in embryonal carcinoma differentiation. RAI3 expression is upregulated in most tumor cell lines that express mutant p53, suggesting that p53 interacts with the promoter of RAI3 and represses its expression at the beginning of apoptosis. RAI3 is a potential molecular target for diagnosing breast cancer, and selective suppression of signals from RAI3 may have a place in breast cancer treatments.

Description:   This gene encodes a member of the transforming growth factor beta (TGFB) family of cytokines, which are multifunctional peptides that regulate proliferation, differentiation, adhesion, migration, and other functions in many cell types. Many cells have TGFB receptors, and the protein positively and negatively regulates many other growth factors. The secreted protein is cleaved into a latency-associated peptide (LAP) and a mature TGFB1 peptide, and is found in either a latent form composed of a TGFB1 homodimer, a LAP homodimer, and a latent TGFB1-binding protein, or in an active form composed of a TGFB1 homodimer. The mature peptide may also form heterodimers with other TGFB family members. This gene is frequently upregulated in tumor cells, and mutations in this gene result in Camurati-Engelmann disease.

Description:   Leupaxin is a 386 amino acid cytoplasmic protein and member of the paxillin family. Leupaxin is highly expressed in lymphoid tissues such as spleen, lymph node, thymus and appendix, with low expression in bone marrow and fetal liver. Consisting of four leucine-rich LD-motifs at the N-terminus and four LIM domains at the C-terminus, leupaxin associates with a member of the focal adhesion kinase family, PYK2, in lymphoid cells. The leupaxin and PYK2 complex is involved in cell type-specific signaling in which it regulates signaling at sites of adhesion. Leupaxin is a substrate for tyrosine kinase in lymphoid cells and is suggested to participate in and be regulated by tyrosine kinase activity. Leupaxin may be a potential progression marker for a subset of prostate cancer and may act as a novel coactivator of the androgen receptor.

Description:   Cell cycle progression is controlled in part by a family of cyclin proteins and cyclin dependent kinases (Cdks). Cdk proteins work in concert with cyclins to phosphorylate key substrates involved in cell cycle progression. Another family of proteins, Cdk inhibitors, also play a role in regulating the cell cycle by binding to cyclin-Cdk complexes and modulating their activity. Members of the Cdk family include Cdk2–Cdk8, PCTAIRE-1–3, PITALRE and PITSLRE. PCTAIRE-1, PCTAIRE-2 and PCTAIRE-3 comprise a subfamily of cdc2-related serine/threonine kinases. PCTAIRE-1, which is expressed primarily in mammalian brain, interacts with a variety of proteins and is thought to be part of a multiple signal transduction cascade. PCTAIRE-2, also expressed in brain, may be important in terminally differentiated neurons.

Description:   CBLL1, also known as HAKAI (meaning ‘destruction’ in Japanese), or RNF188 (RING finger protein 188), is a 491 amino acid protein that contains one C2H2-type zinc finger and one RING-type zinc finger. CBLL1 is believed to function as an E3 ubiquitin-protein ligase that accepts a ubiquitin residue from an E2 ubiquitin-conjugating enzyme and immediately transfers that residue to a protein that is targeted for degradation. More specifically, upon activation of c-Src, CBLL1 interacts with and ubiquitinates tyrosine-phosphorylated E-cadherin, thereby targeting the E-cadherin complex for endocytosis and disrupting epithelial cell-cell contacts. Via its role as an E-cadherin regulator, CBLL1 participates in cell adhesion and may also be involved in the regulation of epithelial-mesenchymal transitions.

Description:   The claudin superfamily consists of structurally related proteins that are important structural and functional components of tight junctions. Claudin-23, also known as CLDN23 or CLDNL, is a 292 amino acid multi-pass membrane protein that localizes to cell junctions and belongs to the claudin family. Expressed in stomach and placenta, as well as in germinal center B-cells, claudin-23 is thought to exhibit calcium-dependent cell-adhesion activity through which it plays an essential role in tight junction-specific obliteration of the intercellular space. Human claudin-23 shares 80% sequence similarity with its mouse counterpart, suggesting a conserved role between species. Overexpression of claudin-23 is associated with colon tumors, implicating claudin-23 as a possible metastasis factor.

Description:   The protein tyrosine phosphatase (PTP) family of proteins are signaling molecules that regulate processes such as cell growth, cell differentiation, oncogenic transformation and the mitotic cycle. PCPTP1, also known as PTPRR (Receptor-type tyrosine-protein phosphatase R), ECPTP, PTPBR7 or PTPRQ, is a 657 amino acid protein that functions to sequester inactive mitogen-activated protein kinases (MAPKs) to the cytoplasm. Expressed primarily in the brain with weaker expression in other parts of the body, PCPTP1 is a receptor-like molecule that is able to dephosphorylate MAPKs, thereby rendering them inactive. Three isoforms of PCPTP1 exist and are designated alpha, beta and gamma. The alpha form is localized to the cell membrane, while the beta and gamma forms are localized to the perinuclear areas within the cytoplasm.
