Interferon (IFN) Inducible 16 (IFI16) protein belongs to a family of HIN 200 human and mouse proteins. IFI16 is a nuclear protein comtaining regulatory domains such as DNA binding domain, transcriptional regulatory domain and DAPIN/PAAD domain. IFI16 has three isotypes A, B, and C (85-95 kDa), which arise as a result of mRNA alternative splicing. All are phosphorylated on serine and threonine residues and can homo and heterodimerize. Expression is restricted to the nuclei of hematopoietic cells, fibroblasts and epithelial cells. IFI16 expression in hematopoietic cells of myeloid lineage is tightly regulated and highly induced in the differentiation and proliferation of the cell. Due to its localisation in the nucleus, regulation of protein expression, and ability to bind DNA, it is assumed that IFI16 has a role in transcription regulation of cell differentiation. In addition, it was found that IFI16 can act as a transcriptional repressor and is involved in regulation and activation of p53 in cancer cells.

CYTIP is a cytoplasmic protein that is involved in lymphocytic cell adhesion. Expressed primarily in hematopoetic cells, CYTIP regulates the activity of cytohesin-1 (an integrin-activating protein involved in cell adhesion) by mediating its recruitment to the leukocyte membrane. Through its ability to bind cytohesin-1, CYTIP is able to sequester it to the cytoplasm, thereby preventing cytohesin-1 translocation to lymphocytes and interrupting the flow of information in the cell adhesion pathway. CYTIP can be recruited from the cytoplasm to the membrane by leukocyte integrins which interact with CYTIP through its PDZ domain. After membrane translocation, CYTIP can be re-located to the cytoplasm via exposure to a phorbol ester. Additionally, CYTIP associates with SNX27 (sorting nexin 27) and helps to coordinate trafficking and signaling complexes. Up-regulation of CYTIP is observed in maturing dendritic cells, suggesting a possible role in developmentally-controlled cell adhesion.

DTX2 belongs to the Deltex family. It contains one RING-type zinc finger and two WWE domains. DTX2 is a regulator of Notch signaling, a signaling pathway involved in cell-cell communications that regulates a broad spectrum of cell-fate determinations. It probably acts both as a positive and negative regulator of Notch, depending on the developmental and cell context; mediates the antineural activity of Notch, possibly by inhibiting the transcriptional activation mediated by MATCH1. DTX2 also functions as an ubiquitin ligase protein in vitro, suggesting that it may regulate the Notch pathway via some ubiquitin ligase activity. Protocadherins are a large family of cadherin-like cell adhesion proteins that are involved in the establishment and maintenance of neuronal connections in the brain. There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDH17 is a 1,159 amino acid single-pass type I membrane protein that contains six cadherin domains. Expressed as multiple alternatively spliced isoforms, PCDH17 is thought to function as a calcium-dependent cell adhesion protein that may play a role in establishing cell-cell connections within brain tissue. Protein with hypoxia-induced expression in endothelial cells. May act as a regulator of angiogenesis and modulate tumorgenesis. Inhibits proliferation, migration, and tubule formation of endothelial cells and reduces vascular leakage. May exert a protective function on endothelial cells through an endocrine action. It is directly involved in regulating glucose homeostasis, lipid metabolism, and insulin sensitivity. In response to hypoxia, the unprocessed form of the protein accumulates in the subendothelial extracellular matrix (ECM). The matrix-associated and immobilized unprocessed form limits the formation of actin stress fibers and focal contacts in the adhering endothelial cells and inhibits their adhesion. It also decreases motility of endothelial cells and inhibits the sprouting and tube formation.
