SLAMF7 contains one Ig-like C2-type (immunoglobulin-like) domain. Isoform 1 mediates NK cell activation through a SAP-independent extracellular signal-regulated ERK-mediated pathway. It may play a role in lymphocyte adhesion. Isoform 3 does not mediate any activation. SAP can bind the cytoplasmic tail of isoform 1 when phosphorylated in the presence of Fyn (in vitro). SLAMF7 is expressed in spleen, lymph node, peripheral blood leukocytes, bone marrow, small intestine, stomach, appendix, lung and trachea. Expression was detected in NK cells, activated B-cells, NK-cell line but not in promyelocytic, B-, or T-cell lines. The isoform 3 is expressed at much lower level than isoform 1. There are three named isoforms.

No reactivity has been observed with reactive B-cells. So a B-lineage population that co-expresses CD43 is highly likely to be a malignant lymphoma, especially a low-grade lymphoma, rather than a reactive B-cell population. When CD43 antibody is used in combination with anti-CD20, effective immunophenotyping of the lymphomas in formalin-fixed tissues can be obtained. Co-staining of a lymphoid infiltrate with anti-CD20 and anti-CD43 argues against a reactive process and favors a diagnosis of lymphoma.

Recognizes a protein of 55 kDa, identified as CD25. It is expressed on activated T- and B-cells and activated monocytes/macrophages. With respect to lymphomas, CD25 is present on malignant cells of Hodgkin's disease, HTLV-1 adult T-cell leukemia, cutaneous T-cell lymphoma, and hair cell leukemia. Increased levels of soluble CD25 are observed in the leukemias/lymphomas and inflammatory/ autoimmune diseases. CD25 molecule alone appears to function as a low affinity receptor and associates with CD122 (IL-2R chain, p75) and CD132 (common chain) to form the high affinity IL-2 receptor complex. CD25 antibodies detect three epitope regions, A, B and C. This MAb recognizes the epitope B, which is located at residue 3-104 of CD25 and doe not block IL-2 binding to CD25.

TrkA is a member of the neurotrophic tyrosine kinase receptor family. It is a membrane-bound receptor that upon neurotrophin binding, phosphorylates itself and members of the MAPK pathway. TrkA is required for high-affinity binding to nerve growth factor (NGF), neurotrophin-3 and neurotrophin-4/5 but not brain-derived neurotrophic factor (BDNF). TrkA leads to cell differentiations and may play a role in specifying sensory neuron subtypes. It has a crucial role in the development and function of the nociceptive reception system as well as establishment of thermal regulation via sweating. SUBUNIT: Exists in a dynamic equilibrium between monomeric (low affinity) and dimeric (high affinity) structures. The 53-6.7 antibody reacts with the 32-34 kDa alpha subunit of mouse CD8, known as CD8a or CD8 alpha. CD8a can form a homodimer (CD8 alpha-alpha), but is more commonly expressed as a heterodimer with a second chain known as CD8b or CD8 beta. CD8 acts as a co-receptor in antigen recognition and subsequent T cell activation that is initiated upon binding of the T cell receptor (TCR) to antigen-bearing MHC Class I molecules. The cytoplasmic domains of CD8 provide binding sites for the tyrosine kinase lck, facilitating intracellular signaling events that lead to T cell activation, development, and cytotoxic effector functions. CD8+ cytotoxic T cells (CTLs) play an important role in inducing cell death of tumor cells, as well as cells infected by virus, bacteria or parasites.

CD109 is a a novel member of the alpha2-macroglobulin (alpha2M)/complement superfamily. It is a glycosylphosphatidylinositol (GPI)-linked cell surface glycoprotein of approximately 170 kd found onendothelial cells, activated platelets and T-cells and on a subset of hematopoietic stem and progenitor cells. Although it has been suggested that T-cell CD109 may play a role in antibody-inducing T-helper function and it is known that platelet CD109 carries the Gov alloantigen system, the role of CD109 in hematopoietic cells remains largely unknown.CD109 has been identified as part of the TGF-beta receptor system in human keratinocytes and upregulation of CD109 expression has been observed in several different types of tumour.

SP17 is a protein present at the cell surface. The N-terminus has sequence similarity to human cAMP-dependent protein kinase A (PKA) type II alpha regulatory subunit (RIIa) while the C-terminus has an IQ calmodulin-binding motif. The central portion of the protein has carbohydrate binding motifs and likely functions in cell-cell adhesion. The protein was initially characterized by its involvement in the binding of sperm to the zona pellucida of the oocyte. Recent studies indicate that it is also involved in additional cell-cell adhesion functions such as immune cell migration and metastasis. A retrotransposed pseudogene is present on chromosome 10q22.[provided by RefSeq, Jan 2009].
