Description:
Mahogany (MG), originally identified as a protein involved in pigmentation, acts in conjunction with melanocortin receptors to suppress diet-induced obesity. Mahogany contains a single transmembrane domain, and it is expressed in a broad range of tissues, including the hypothalamus and pigment cells. Mutations within the mahogany gene were shown to rescue agouti-lethal-yellow mutant mice from obesity. The extracellular domain of mouse mahogany is the ortholog of the human protein attractin. Attractin (also designated DPPT-L) is a human serum glycoprotein and is a member of the CUB family of cell adhesion and guidance proteins. Attractin is expressed on activated T cells and is released from the cells 48 to 72 hours after activation.

This antibody recognizes a transcription factor of 64-67 kDa, identified as c-myc. This MAb shows no cross-reaction with v-myc. c-myc is involved in the control of cell proliferation and differentiation and is amplified and/or over-expressed in a variety of tumors. Over-expression of c-myc protein occurs frequently in luminal cells of prostate intraepithelial neoplasia as well as in most primary carcinomas and metastatic disease. Rearrangement of the MYC gene is found in 3% to 16% of diffuse large B-cell lymphoma (DLBCL's) and in nearly 100% of Burkitt lymphomas (BL). Identifying MYC status is important in establishing final diagnosis of DLBCL, BL, or B-cell lymphoma, with features intermediate between DLBCL and BL as well as in differential diagnoses of the lymphomas.

NHE-3 are integral membrane proteins that are expressed in most mammalian tissues, where they regulate intracellular pH and cell volume. NHEs mediate the transport of hydrogen (H+) ions out of cells in exchange for extracellular sodium (Na+) ions. While NHE-1 is ubiquitously expressed, the NHE isoforms 2-8 have distinct tissue- and cell type-dependent expression and inhibitory characteristics. NHE-3 localizes to the apical membrane of renal proximal tubules where it is responsible for most of the sodium transport and fluid reabsorption. NHE-3 translocates to internal pools where it mediates natriuresis when blood pressure increases abruptly. NHE-3 is also expressed in the stomach and functions to protect the mucosa by secreting protons that diffuse into the mucous cells.

PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This gene is preferentially expressed in a variety of hematopoietic cells, and is an early response gene in lymphokine stimulated cells. The non-catalytic N-terminus of this PTP can interact with MAP kinases and suppress the MAP kinase activities. This PTP was shown to be involved in the regulation of T cell antigen receptor (TCR) signaling, which was thought to function through dephosphorylating the molecules related to MAP kinase pathway. Recognizes a protein of 74 kDa, identified as CD84 (Workshop V; Code A085, C057). It is expressed on mature B cells and on B-cell lines, including pre-B-cell lines, but not on plasma cell lines. Immunohistochemical studies demonstrated that CD84 strongly expressed on tissues macrophages. CD84 is also highly expressed on platelets and, at low levels on peripheral blood T cells. The DTA-1 antibody reacts with mouse CD357, also known as GITR or AITR (in humans), a 66-70 kDa member of the Tumor Necrosis Factor superfamily (TNFRSF18). GITR is primarily found on T cells, and its function may vary depending on the T cell type where it is expressed. GITR is upregulated on activated T cells where it provides co-stimulation, yet GITR may promote the inhibition of CD4+ CD25+ Treg cells, where it is expressed at high levels. GITR ligand (GITRL) is found on B cells, macrophages, dendritic and endothelial cells, and is implicated in regulating both innate and adaptive immune responses.

This MAb reacts with an extracellular domain (close to transmembrane) of CD56/NCAM. Three isoforms of neural cell adhesion molecule (NCAM) are produced by differential splicing of the RNA transcript from a single gene. The 135 kDa isoform is the basic molecule, which is glycosylated or sialylated to produce the mature species. Anti-CD56 recognizes two proteins of the neural cell adhesion molecule, the basic molecule expressed on most neuroectodermally derived tissues and neoplasms (e.g. retinoblastoma, medulloblastomas, astrocytomas, neuroblastomas, and small cell carcinomas). It is also expressed on some mesodermally derived tumors (rhabdomyosarcoma). Anti-CD56 plays an important role in the diagnosis of nodal and nasal NK/T-cell lymphomas.
