Description:
BAMBI is a membrane spanning glycoprotein that acts as a negative regulator of TGF-beta signaling during development. The BAMBI family of proteins are related to type I TGF-beta receptor family, however, BAMBI is a pseudoreceptor that lacks an intracellular serine/threonine kinase domain. BAMBI transcription regulation is under the influence of beta-catenin, BMP, smad3 and smad4. BAMBI expression can increase in colorectal and hepatocellular carcinomas relative to non-cancerous tissues. BAMBI is expressed at high levels during odontogenesis. BAMBI is coexpressed with Bmp-4 during early Xenopus embryogenesis and can be detected in poorly metastatic human melanoma cell lines.

Members of the bactericidal/permeability-increasing protein family have antimicrobial properties and bind lipophilic substances, therefore targeting gram-negative bacteria. The bactericidal permeability increasing protein (BPI) is an antibacterial and endotoxin-neutralizing molecule that is abundant in the granules of polymorphonuclear leukocytes (neutrophil granules). Sharing structural and sequence homologies with BPI, BPIL2 (Bactericidal/permeability-increasing protein-like 2) is a 507 amino acid secreted protein that contains the family’s common conserved feature of two cysteine residues that are critical for protein function. While BPIL2 is expressed in the basal layer of inflamed epidermis from psoriasis patients, it is not detected in normal skin.

TRITC (Tetramethylrhodamine isothiocyanate) is a popular rhodamine derivative pink in color but emits a red-orange color upon Emission at 580nm. TRITC is frequently paired with FITC in double labeling experiments and conjugated antibodies are often used in immunofluorescence microscopy, flow cytometry, FLISA (fluorescent ELISA) and high throughput screening assays. These conjugates are best excited with the 532 nm spectral line of the He-Ne laser and are also recommended as a second color detection reagent in in situ hybridization applications. Recognizes a phosphor-protein of 45 kDa, identified as MyoD1. The epitope of this MAb maps between amino acid 180-189 in the C-terminal of mouse MyoD1 protein. It does not cross react with myogenin, Myf5, or Myf6. Antibody to MyoD1 labels the nuclei of myoblasts in developing muscle tissues. MyoD1 is not detected in normal adult tissue, but is highly expressed in the tumor cell nuclei of rhabdomyosarcomas. Occasionally nuclear expression of MyoD1 is seen in ectomesenchymoma and a subset of Wilm s tumors. Weak cytoplasmic staining is observed in several non-muscle tissues, including glandular epithelium and also in rhabdomyosarcomas, neuroblastomas, Ewing s sarcomas and alveolar soft part sarcomas.

The PAX5 gene is a member of the paired box (PAX) family of transcription factors. The central feature of this gene family is a novel, highly conserved DNA-binding motif, known as the paired box. The PAX proteins are important regulators in early development, and alterations in the expression of their genes are thought to contribute to neoplastic transformation. The PAX5 gene encodes the B-cell lineage specific activator protein (BSAP) that is expressed at early, but not late stages of B-cell differentiation. Its expression has also been detected in developing CNS and testis, therefore, PAX5 gene product may not only play an important role in B-cell differentiation, but also in neural development and spermatogenesis.

Description:
Capases are a highly conserved family of cysteine proteases and are synthesized as inactive zymogens, which are cleaved at Asp-x sites during apoptosis, generating large and small subunits that heterodimerize to form the active enzyme. Caspase-1 is a member of the ICE/CED-3 family of caspases and is detected in the pro-form as a protein of 45 kDa.
Caspase-1 generates the bioactive form of the pro-inflammatory cytokine IL-1β from its biologically inactive precursor. Overexpression of caspase-1 in cells induces apoptosis, which can be inhibited by cytokine response modifier A (CrmA). Caspase-1 mRNA is predominantly expressed in the spleen and lung and to a lesser extent in skeletal muscle, kidney, testis and heart and within these tissues macrophages preferentially express caspase-1. Caspase-1 deficient mice have no gross abnormalities, however, macrophages from caspase-1-deficient mice are defective in IL-β processing and release.

Recognizes a protein of 40 kDa, identified as cytokeratin-19 (CK19), which is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Anti-CK19 is a useful marker for detection of tumor cells in lymph nodes, peripheral blood, bone marrow and breast cancer.

In Western blotting, this antibody recognizes proteins in MW range of 265-400 kDa, identified as different glycoforms of EMA. The α subunit has cell adhesive properties. It can act both as an adhesion and an anti-adhesion protein. EMA may provide a protective layer on epithelial cells against bacterial and enzyme attack. The β subunit contains a C-terminal domain, which is involved in cell signaling, through phosphorylations and protein-protein interactions. In immunohistochemical assays, it superbly stains routine formalin/paraffin carcinoma tissues. Antibody to EMA is useful as a pan-epithelial marker for detecting early metastatic loci of carcinoma in bone marrow or liver.
