Description:   Voltage-gated K+ channels in the plasma membrane control the repolarization and the frequency of action potentials in neurons, muscles and other excitable cells. The KV gene family encodes more than 30 proteins that comprise the subunits of the K+ channels, and they vary in their gating and permeation properties, subcellular distribution and expression patterns. Functional KV channels assemble as tetramers consisting of pore-forming å subunits (KV), which include the KV1, KV2, KV3 and KV4 proteins, and accessory or KV-subunits that modify the gating properties of the coexpressed KV subunits. KV∫, also known as KCNAB1 (potassium voltage-gated channel, shaker-related subfamily, beta member 1), is a 419 amino acid accessory K+ channel protein that exists as three alternatively spliced isoforms and regulates the activity of the pore-forming å subunit. It is expressed in brain, with highest levels detected in caudate nucleus, hippocampus and thalamus.

Description:   Recognizes a 53 kDa protein, which is identified as p53 suppressor gene product. It reacts with the mutant as well as the wild form of p53 under denaturing and non-denaturing conditions. p53 is a tumor suppressor gene expressed in a wide variety of tissue types and is involved in regulating cell growth, replication, and apoptosis. It binds to MDM2, SV40 T antigen and human papilloma virus E6 protein. Positive nuclear staining with p53 antibody has been reported to be a negative prognostic factor in breast carcinoma, lung carcinoma, colorectal, and urothelial carcinoma. Anti-p53 positivity has also been used to differentiate uterine serous carcinoma from endometrioid carcinoma as well as to detect intratubular germ cell neoplasia. Mutations involving p53 are found in a wide variety of malignant tumors, including breast, ovarian, bladder, colon, lung, and melanoma.

Description:   intracellular stimulation of guanylate cyclase (GC) by calcium, a key event in the recovery of the dark state of rod photoreceptors after exposure to light, is mediated by guanylate cyclase-activating protein (GCAP1). GCAPs are calcium-The binding proteins belonging to the calmodulin superfamily. GCAP1 is a calcium-binding protein that stimulates synthesis of c-GMP in photoreceptors. GCAP1 is present in rod and cone photoreceptor outer segments where phototransduction occurs. In contrast to other calcium-binding proteins from the calmodulin superfamily, the calcium-free form of GCAP1 stimulates the effector enzyme. By molecular cloning of human and mouse GCAP cDNA, the known mammalian GCAPs are found to be more than 90% similar, consisting of 201 to 205 amino acids, and containing three identically conserved calcium-binding sites. A related protein, GCAP2, is detectable only in the retina and results from a gene duplication event.

Description:   The chondroitin N-acetylgalactosaminyltransferase family includes Beta-1,4-GalNAc-T, Beta-1,4-GalNAc-T2, Beta-1,4-GalNAc-T3 and Beta-1,4-GalNAc-T4. The Beta-1,4-GalNAc-T protein consists of a short N-terminal residue, a transmembrane region and a long C-terminal residue, which includes a catalytic domain and localizes to the Golgi apparatus. Beta-1,4-GalNAc-T utilizes simple ganglioside GM3 as a substrate for more complex gangliosides GM2, GM1 and GD1a. Beta-1,4-GalNAc-T is expressed in normal brain tissues and in various malignant transformed cells, such as malignant melanoma, neuroblastoma and adult T cell leukemia. Mice lacking the Beta-1,4-GalNAc-T protein develop significant and progressive behavioral neuropathies, including deficits in reflexes, strength, coordination and balance. Beta-1,4-GalNAc-T is a potential molecular marker for detecting melanoma cells and monitoring tumor progression.

Description:   The Oxysterol-binding protein (OSBP) family of proteins consist of OSBP (OSBP1) and OSBP2 (ORP-4), which share a high overall similarity. OSBPs are involved in lipid metabolism and signal transduction, as well as vesicle transport, and can translocate to the periphery of Golgi membranes when they are bound to oxysterols. The OSBP protein transports sterols from lysosomes to the nucleus, where sterols downregulate the genes for HMG synthetase, HMG-CoA reductase and the low density lipoprotein receptor (LDLR). OSBP localizes to the cytosol and is widely expressed, while OSBP2 is mainly detected in testis, retina and fetal liver. The extracellular signal-regulated kinase (ERK) signaling pathway is controlled by OSBP via its cholesterol-binding properties. OSBP binds with a high affinity to 25-hydroxy-cholesterol (25-HC), a suppressor of cholesterol synthesis gene transcription in cultured cells.

Description:   IGSF1 is a highly glycolylated immunoglobulin domain-containing protein. IGSF1 has been shown to act as a coreceptor in inhibin signaling, however, it does not appear to be a high-affinity inhibin receptor by itself. May reduce or inhibit activin A signaling and is believed to be necessary in the mediation of specific effects of inhibin B on activin-stimulated transcription. IGSF1 has been found to interact with several members of the ACVR family and possibly some members of the BMPR group. There are 3 known isoforms of IGSF1, with 1 and 2 likely being multi-pass membrane proteins. Isoform 3 is believed to be expressed as a secreted form. Expression is high in pancreas, testis and fetal liver, while heart, prostate and small intestine show only moderate expression. IGSF1 may be found at very low levels in brain, muscle, thymus, ovary, colon, fetal lung and fetal kidney. Isoform 3 has been detected in pituitary gland.

Description:   Netrin proteins are a family of laminin-related secreted proteins that provide guidance signals for axonal growth and cell migration during development. Netrin signaling is dependent on the concentration of calcium outside the cell and the level of PKA activity. In axonal cells, a reduction in PKA activity converts the responsiveness of the axons to the netrin proteins, as the cells are repelled, rather than attracted, by the netrin gradient. Netrin-4 is related to the Laminin ∫ chains, and is therefore also designated ∫-netrin. It is present in the basement membranes of the vasculature, lateral olfactory tract, kidney and ovary. In humans, the gene encoding for the netrin-4 protein is localized to chromosome 12q22-q23. High levels of netrin-4 mRNA have also been detected in many cells and tissues, including cerebral cortex, hippocampus, amygdaloid nuclei and Purkinje cells. Netrin-4 is important in neural, kidney and vascular development.

Description:   The deduced 257 amino acid protein PACRG (Parkin co-regulated gene) shows potential links to the ubiquitin/proteasome system. PACRG and Parkin are attached in a head-to-head arrangement on opposite DNA strands and share a common 5' flanking promoter region. The PACRG gene maps to chromosome 6q26; Northern blot analysis detects PACRG expression in all tissues examined except placenta. Using a positional cloning strategy in 197 Vietnamese leprosy simplex families (i.e. families with two unaffected parents and one affected child), significant connections between leprosy and 17 markers in the 5' regulatory region that PARK2 and PACRG share were observed. Possession of two or more of the 17 risk alleles is highly predictive of leprosy.PACRG is a gene located very close to parkin, in reverse orientation on the chromosome. It is thought to be co-transcribed with parkin by a bi-directional promoter between the two genes.

Description:   Folate receptor beta is also known as Folate receptor 2, FBP, FOLR2, BETA-HFR, FBP/PL-1, FR-BETA, FR-P3, and is a member of the folate receptor (FOLR) family. and mediate delivery of 5-methyltetrahydrofolate to the interior of cells. This protein has a 68% and 79% sequence homology with the FOLR1 and FOLR3 proteins, respectively. The FOLR2 protein was originally thought to exist only in placenta, but is also detected in spleen, bone marrow, and thymus. FOLR2 is predominantly expressed in placenta, cells of the neutrophilic lineage, and some CD34+ hematopoietic progenitor cells. It is upregulated on myeloid leukemias, head and neck squamous cell carcinomas, and several nonepithelial cancers. It is also upregulated on macrophages and monocytes at chronic inflammatory sites including rheumatoid arthritis synovium and glioblastoma. FOLR2 is a marker for macrophages generated in the presence of M-CSF, but not GM-CSF. Its expression correlates with increased folate uptake ability. Folate conjugates of therapeutic drugs are a potential immunotherapy tool to target tumor-associated macrophages.

Description:   The RA3-6B2 antibody reacts with the human and mouse CD45 isoform known as CD45R, or B220, a protein tyrosine phosphatase of 220 kDa. CD45 is one of the most abundant hematopoietic markers, and is expressed on all leukocytes (the Leukocyte Common Antigen, LCA). Various isoforms are generated and expressed in cell-specific patterns, all critical for leukocyte function. In mouse, the CD45R/B220 isoform is predominantly found on B cells, at varying levels on all stages from pro-B cells to activated B cells, and may also be detected on certain T cell and NK cell subsets. It is of note that B220 is not similarly expressed on human B cells, where it appears to be differentiation-specific and therefore expressed on only some B cell subsets. Other forms of CD45 with restricted cellular expression include CD45RA, CD45RB and CD45RO.

Description:   This antibody recognizes proteins of 80-200 kDa, identified as different members of CEA family. CEA is synthesized during development in the fetal gut and is re-expressed in increased amounts in intestinal carcinomas and several other tumors. This MAb does not react with nonspecific cross-reacting antigen (NCA) and with human polymorphonuclear leucocytes. It shows no reaction with a variety of normal tissues and is suitable for staining of formalin/paraffin tissues. CEA is not found in benign glands, stroma, or malignant prostatic cells. Antibody to CEA is useful in detecting early foci of gastric carcinoma and in distinguishing pulmonary adenocarcinomas (60-70% are CEA ) from pleural mesotheliomas (rarely or weakly CEA ). Anti-CEA positivity is seen in adenocarcinomas from the lung, colon, stomach, esophagus, pancreas, gallbadder, urachus, salivary gland, ovary, and endocervix.
