Description:   Biotin, also known as vitamin B7, is an essential water-soluble vitamin that is a cofactor in glucogenesis and in the metabolism of fatty acids and leucine. Biotinidase is a 523 amino acid enzyme that catalyzes the hydrolysis of biocytin to biotin and lysine. Secreted into extracellular space, biotinidase is expressed in liver, heart, placenta, brain, skeletal muscle, pancreas and kidney. Biotinidase contains one carbon-nitrogen hydrolase domain, which is involved in the reduction of organic nitrogen compounds and ammonia production. Defects in the gene encoding biotinidase are the cause of biotinidase deficiency, which is characterized by skin rash, ataxia, seizures, hearing loss, hypotonia and optic atrophy. These symptoms are due to the individual’s inability to reutilize biotin and can, therefore, typically be treated with the addition of free biotin.

Description:   Beta-defensins (also designated BD, and hBD in human) are small cationic peptides with broad-spectrum antimicrobial activity. Produced in mucosal epithelia and neutrophils of several species, Beta-defensins are developmentally regulated. Human b-defensin 2 is locally regulated by inflammation and is the first member of the b-defensin family that is locally inducible by inflammation. The murine homolog of human b-defensin 2, which is called b-defensin 3, is present in the respiratory system and in low levels in the epithelial cells of the intestine and lung. The unique murine b-defensin 2 (Defb2) is not expressed in airways of untreated mice, but is upregulated in the airways by lipopolysaccharide and may contribute to host defense at the mucosal surface of the airways

Description:   DCAKD belongs to the coaE family. It contains one DPCK (dephospho CoA kinase) domain. There are two isoforms.Coenzyme A (CoA) is an essential cofactor used in numerous biochemical pathways. It plays a critical role in the synthesis and oxidation of fatty acids and is vital to the citric acid cycle. The biosynthesis pathway of CoA from pantothenic acid (also known as vitamin B5) is essential and universal in prokaryotes and eukaryotes. In humans, the final steps of the biosynthesis pathway are carried out by the bifunctional enzyme COASY. The sequence of these enzymes are highly conserved between different bacterial species. The phosphopantetheine adenylyltransferase and decoenzyme A kinase activities of COASY are evolutionarily conserved activities. DCAKD (deCoA kinase domain containing protein) is a 231 amino acid protein that consists of a deCoA kinase domain and an ATP nucleotide binding motif. Localizing to mitochondria and the cytosol, DCAKD belongs to the coaE family which suggests that it may play a role in the biosynthesis of CoA.

Description:   MEK5 is a dual specificity protein kinase belonging to thr Ser/Thr protein kinase family, (MAP kinase kinase family). It is activated by phosphorylation on Ser/Thr by MAP kinase kinases and interacts specifically with ERK5, and not with another MAP kinase like P38. This kinase specifically interacts with and activates MAPK7/ERK5. This kinase itself can be phosphorylated and activated by MAP3K3/MEKK3, as well as by atypical protein kinase C isoforms (aPKCs). It is not phosphorylated by RAFA, RAFB or RAFC and it may interact with GTPases such as CDC42. The signal cascade mediated by this kinase is involved in growth factor stimulated cell proliferation and muscle cell differentiation. MEK5 is expressed in many adult tissues and is most abundant in heart and skeletal muscle.

Description:   The FAH family contains two highly homologous 314 amino acid proteins, designated FAHD2A (fumarylacetoacetate hydrolase domain-containing protein 2A) and FAHD2B (fumarylacetoacetate hydrolase domain-containing protein 2A). FAHD2A and B utilize calcium and magnesium as cofactors, and may possess hydrolase activity. The genes encoding FAHD2A/B map to human chromosome 2, the second largest human chromosome, which consists of 237 million bases, encodes over 1,400 genes and makes up approximately 8% of the human genome. A number of genetic diseases are linked to genes on chromosome 2. Harlequin icthyosis, a rare and morbid skin deformity, is associated with mutations in the ABCA12 gene. The lipid metabolic disorder sitosterolemia is associated with ABCG5 and ABCG8. An extremely rare recessive genetic disorder, Alstré°‰ syndrome, is also associated with mutations to chromosome 2.

Description:   May be involved in neurite outgrowth.G protein-coupled receptors (GPCRs) represent a large superfamily of cell-surface receptors that are involved in a multitude of physiological processes such as perception of sensory information, modulation of synaptic transmission, hormone release/action, regulation of cell contraction/migration and cell growth/differentiation. GPCRs interact with G proteins (heterotrimeric GTPases) to synthesize intracellular second messengers, such as diacylglycerol, cyclic AMP, inositol phosphates and calcium ions. Their diverse biological functions range from vision and olfaction to neuronal and endocrine signaling, and are involved in many pathological conditions. GRIN3 (G protein-regulated inducer of neurite outgrowth 3), also known as GPRIN3, is a 776 amino acid protein that contains a C-terminal region which shares a high homology with GRIN2 and GRIN1, and may function in neurite outgrowth.

Description:   Transmembrane serine/threonine kinase activin type-2 receptor forming an activin receptor complex with activin type-1 serine/threonine kinase receptors (ACVR1, ACVR1B or ACVR1c). Transduces the activin signal from the cell surface to the cytoplasm and is thus regulating many physiological and pathological processes including neuronal differentiation and neuronal survival, hair follicle development and cycling, FSH production by the pituitary gland, wound healing, extracellular matrix production, immunosuppression and carcinogenesis. Activin is also thought to have a paracrine or autocrine role in follicular development in the ovary. Within the receptor complex, the type-2 receptors act as a primary activin receptors (binds activin-A/INHBA, activin-B/INHBB as well as inhibin-A/INHA-INHBA). The type-1 receptors like ACVR1B act as downstream transducers of activin signals. Activin binds to type-2 receptor at the plasma membrane and activates its serine-threonine kinase. The activated receptor type-2 then phosphorylates and activates the type-1 receptor. Once activated, the type-1 receptor binds and phosphorylates the SMAD proteins SMAD2 and SMAD3, on serine residues of the C-terminal tail. Soon after their association with the activin receptor and subsequent phosphorylation, SMAD2 and SMAD3 are released into the cytoplasm where they interact with the common partner SMAD4. This SMAD complex translocates into the nucleus where it mediates activin-induced transcription. Inhibitory SMAD7, which is recruited to ACVR1B through FKBP1A, can prevent the association of SMAD2 and SMAD3 with the activin receptor complex, thereby blocking the activin signal. Activin signal transduction is also antagonized by the binding to the receptor of inhibin-B via the IGSF1 inhibin coreceptor (By similarity).
