This MAb stains the cytoplasm of macrophages and histiocytes in hematopoietic organs, Kupffer's cells of the liver and Langerhan's cells of the skin. It also stains the mantle zone B-lymphocytes of the lymph node and spleen, spermatogonia, and chief cells of the stomach. S100A9 is expressed by macrophages in acutely inflamed tissues and in chronic inflammation. It is detected in peripheral blood leukocytes, in neutrophils and granulocytes. It is present at sites of vascular inflammation. S100A9 is also expressed in epithelial cells constitutively or induced during dermatoses. S100A9 is a Calcium-binding protein. It has antimicrobial activity towards bacteria and fungi. It is important for resistance to invasion by pathogenic bacteria. It up-regulates transcription of genes that are under the control of NF-kappa-B. S100A9 plays a role in the development of endotoxic shock in response to bacterial lipopolysaccharide (LPS). It promotes tubulin polymerization when unphosphorylated. It also promotes phagocyte migration and infiltration of granulocytes at sites of wounding. It plays a role as a pro-inflammatory mediator in acute and chronic inflammation and up-regulates the release of IL8 and cell-surface expression of ICAM1.

Recognizes a single protein of 33-34 kDa, identified as the prostate specific antigen (PSA). This MAb is highly specific to PSA and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. PSA is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign prostate hyperplasia. This MAb makes an excellent pair with MAb 1A7G6B6 for PSA tests.

Voltage-gated potassium (Kv) channels represent the most complex class of voltage-gated ion channels from both functional and structural standpoints. Their diverse functions include regulating neurotransmitter release, heart rate, insulin secretion, neuronal excitability, epithelial electrolyte transport, smooth muscle contraction, and cell volume. This gene encodes a member of the potassium channel, voltage-gated, isk-related subfamily. This member is a type I membrane protein, and a beta subunit that assembles with a potassium channel alpha-subunit to modulate the gating kinetics and enhance stability of the multimeric complex. ANKRD5 is a 776 amino acid protein that contains one calcium-binding EF-hand domain and 8 ankyrin repeats. The gene encoding ANKRD5 maps to chromosome 20. Comprising approximately 2% of the human genome, chromosome 20 contains nearly 63 million bases that encode over 600 genes, some of which are associated with Creutzfeldt-Jakob disease, amyotrophic lateral sclerosis, spinal muscular atrophy, ring chromosome 20 epilepsy syndrome and Alagille syndrome. Additionally, chromosome 20 contains a region with numerous genes which are thought important for seminal production and may be potential targets for male contraception.

Description:
This MAb is specific to kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. It recognizes human Ig kappa light chains of both secreted and cell surface immunoglobulin. It detects also free kappa light chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.
