Description:   The leucine-rich (LRR) repeat is a 20-30 amino acid motif that forms a hydrophobic å/∫ horseshoe fold, allowing it to accommodate several leucine residues within a tightly packed core. All LRR repeats contain a variable segment and a highly conserved segment, the latter of which accounts for 11 or 12 residues of the entire LRR motif. ISLR2 (immunoglobulin superfamily containing leucine-rich repeat 2), also known as LINX (leucine-rich repeat domain and immunoglobulin domain-containing axon extension protein), is a 745 amino acid single-pass membrane protein that contains five LRR repeats, one Ig-like (immunoglobulin-like) domain, a LRRCT domain and one LRRNT domain. ISLR2 exists as a homomultimer and is essential for axon extension during neural development. The gene encoding ISLR2 maps to human chromosome 15q24.1.

Description:   Cell cycle progression is controlled in part by a family of cyclin proteins and cyclin dependent kinases (Cdks). Cdk proteins work in concert with the cyclins to phosphorylate key substrates involved in each phase of cell cycle progression. Specifically, Cdk2 interacts with Cyclins A, B1, B3, D, or E to control cell cycle progression. The Cyclin-dependent kinase 2-interacting protein (CINP) interacts with components of the replication complex and Cdk2 and Cdc7, thereby providing a functional and physical link between Cdk2 and Cdc7 during firing of the origins of replication. However, CINP is phopshorylated by Cdc7, but not by Cdk2. CINP also interacts with ATR-interacting protein and regulates ATR-dependent signaling, resistance to replication stress and G2 checkpoint integrity.

Description:   Myotubularin and the myotubularin-related proteins (MTMR1-9) belong to a highly conserved family of eukaryotic phosphatases. They are protein tyrosine phosphatases that utilize inositol phospholipids, rather than phosphoproteins, as substrates. MTMR family members hydrolyze both Phosphatidylinositol 3-phosphate (PtdIns3P) and PtdIns(3,5)P2. MTMR2 interacts with MTMR5, an inactive family member that increases the enzymatic activity of MTMR2 and dictates its subcellular localization. Mutations in MTMR2 cause autosomal recessive Charcot-Marie-Tooth type 4B1 (CMT4B1), which is characterized by reduced nerve conduction velocities, focally folded myelin sheaths and demyelination. MTMR3 and MTMR4 can either interact with each other or self associate. MTMR6 regulates the activity of the calcium-activated potassium channel 3.1. MTMR9 regulates the activity of MTMR7 and MTMR8.

Description:   CBG is a monomeric enzyme involved in the absorption and metabolism of flavonoid glucosides. CBG is found predominately in the liver, but is also located in tissues such as spleen, small intestine and kidney. Through its catalytic activity, CBG is able to hydrolyze a variety of glycosides including phytoestrogens, cyanogens, and flavonols. Although its catalytic activity extends to many dietary flavonoids, CBG has increased specificity for hydrophobic aglycones such as beta-D-glucoside and beta-D-galactoside. Hydrolysis is inhibited by sodium taurocholate and glucosyl-sphingosine, both of which regulate CBG enzymatic activity. Deficiencies in CBG have been implicated in Gaucher’s disease, a lysosomal storage disease that causes a build up of fatty material in the spleen, liver, lung and kidneys.

Description:   FSD2 is a 749 amino acid protein containing one B30.2/SPRY domain and two fibronectin type-III domains. The gene encoding FSD2 maps to human chromosome 15q25.2. Encoding more than 700 genes, chromosome 15 is made up of approximately 106 million base pairs and consists of about 3% of the human genome. Angelman and Prader-Willi syndromes are associated with loss of function or deletion of genes in the 15q11-q13 region. In the case of Angelman syndrome, this loss is due to inactivity of the maternal 15q11-q13 encoded UBE3A gene in the brain by either chromosomal deletion or mutation. Prader-Willi syndrome, Tay-Sachs disease and Marfan syndrome are also associated with chromosome 15.

Description:   Fused toes protein homolog (FTS), also known as AKT-interacting protein (AKTIP) and Ft1, is a 292 amino acid protein that localizes to the cytoplasm and the cell membrane. A member of the ubiquitin-conjugating enzyme family, FTS binds directly to AKT1 to regulate apoptosis in a cell population. AKT1 is a protein that plays a critical role in a number of cellular responses, such as cell growth, protein synthesis, and antiapoptotic signaling. The interaction of FTS and AKT1 enhances the phosphorylation and activation of AKT1, which, through an AKT1/GSK-3∫/NFATc1 signaling cascade, results in the increased production of the proapoptotic hormone Fas ligand and thus an increase in apoptosis.

Description:   Functions in the lectin pathway of complement, which performs a key role in innate immunity by recognizing pathogens through patterns of sugar moieties and neutralizing them. The lectin pathway is triggered upon binding of mannan-binding lectin (MBL) and ficolins to sugar moieties which leads to activation of the associated proteases MASP1 and MASP2. Functions as an endopeptidase and may activate MASP2 or C2 or directly activate C3 the key component of complement reaction. Isoform 2 may have an inhibitory effect on the activation of the lectin pathway of complement or may cleave IGFBP5.

Description:   IL15RA is a cytokine receptor that specifically binds IL15 with high affinity. It shares two subunits with the receptor of IL2, the IL2R beta and IL2R gamma chains. This forms the basis of many overlapping biological activities of IL15 and IL2. The IL2 receptor requires an additional IL2-specific alpha subunit for high affinity IL2 binding. This protein is structurally related to IL2R alpha, but is capable of binding IL15 with high affinity independent of other subunits, which suggests the distinct roles between IL15 and IL2. This receptor is reported to enhance cell proliferation and expression of apoptosis inhibitor BCL2L1/BCL2-XL and BCL2. There are nine different isoforms.

Description:   KNDC1 is a 1749 amino acid protein that contains two KIND domains and an N-terminal Ras-GEF domain. Expressed in the cerebral cortex, KNDC1 is a likely guanine nucleotide exchange factor (GEF). Existing as six alternatively spliced isoforms, the gene encoding KNDC1 maps to human chromosome 10q26.3 and mouse chromosome 7 F4. Spanning nearly 135 million base pairs, chromosome 10 makes up approximately 4,5% of total DNA in cells and encodes nearly 1200 genes. Several protein-coding genes, including those that encode for chemokines, cadherins, excision repair proteins, early growth response factors (Egrs) and fibroblast growth receptors (FGFRs), are located on chromosome 10. Defects in some of the genes that map to chromosome 10 are associated with Charcot-Marie Tooth disease, Jackson-Weiss syndrome, Usher syndrome, nonsyndromatic deafness, Wolman's syndrome, Cowden syndrome, multiple endocrine neoplasia type 2 and porphyria.

Description:   Ubiquitination involves at least three classes of enzymes: ubiquitin-activating enzymes (UBE1s), ubiquitin-conjugating enzymes (UBE2s), and ubiquitin-protein ligases (UBE3s). When ubiquitin is activated by a UBE1, it is transferred to the cysteine residue on a UBE2. UBE2 then binds a UBE3, which transfers the ubiquitin from the UBE2 cysteine to a lysine residue on the target protein. Ubiquitin-conjugating enzyme E2 Q1 (UBE2Q1), also known as ubiquitin-protein ligase Q1 or ubiquitin carrier protein Q1, is an 422 amino acid protein belonging to the ubiquitin-conjugating enzyme (UBE2) family. Two named isoforms of UBE2Q1 exist as a result of alternative splicing events.
