CD200 is also known as OX-2 membrane glycoprotein (OX-2), is a type-1 membrane glycoprotein, which contains two immunoglobulin domains (1 Ig-like C2-type (immunoglobulin-like) domain and Ig-like V-type (immunoglobulin-like) domain), and thus belongs to the immunoglobulin superfamily. CD200 / OX-2 is widely expressed in multiple cell types. CD200 interacts with a structurally related receptor (CD200R) expressed mainly on myeloid cells and is involved in regulation of macrophage and mast cell function. OX-2 / CD200 and CD200R associate via their respective N-terminal Ig-like domains. CD200 also plays an important role in prevention of graft rejection, autoimmune diseases and spontaneous abortion.

Programmed cell death protein 1 (PD-1) is also known as CD279 and PDCD1, is a type I membrane protein and is a member of the extended CD28/CTLA-4 family of T cell regulators. PDCD1 is expressed on the surface of activated T cells, B cells, macrophages, myeloid cells and a subset of thymocytes. PD-1 has two ligands, PD-L1 and PD-L2, which are members of the B7 family. PD-L1 is expressed on almost all murine tumor cell lines, including PA1 myeloma, P815 mastocytoma, and B16 melanoma upon treatment with IFN-γ. PD-L2 expression is more restricted and is expressed mainly by DCs and a few tumor lines. PD1 inhibits the T-cell proliferation and production of related cytokines including IL-1, IL-4, IL-10 and IFN-γ by suppressing the activation and transduction of PI3K/AKT pathway. In addition, coligation of PD1 inhibits BCR-mediating signal by dephosphorylating key signal transducer. In vitro, treatment of anti-CD3 stimulated T cells with PD-L1-Ig results in reduced T cell proliferation and IFN-γ secretion. Monoclonal antibodies targeting PD-1 that boost the immune system are being developed for the treatment of cancer.

Osteopontin (OPN) is also known as Secreted phosphoprotein 1 (SPP1), Bone sialoprotein 1, Nephropontin, Urinary stone protein, Uropontin, BNSP, which belongs to the osteopontin family. OPN / SPP1 is a highly negatively charged, extracellular matrix protein that lacks an extensive secondary structure. Full length OPN (OPN-FL) can be modified by thrombin cleavage, which exposes a cryptic sequence, SVVYGLR on the cleaved form of the protein known as OPN-R. Osteopontin / SPP-1 is biosynthesized by a variety of tissue types. OPN is the ligand for integrin alpha-V/beta-3. OPN / SPP1 binds tightly to hydroxyapatite and appears to form an integral part of the mineralized matrix. OPN / SPP1 probably important to cell-matrix interaction. OPN / SPP1 acts as a cytokine involved in enhancing production of interferon-gamma and interleukin-12 and reducing production of interleukin-10 and is essential in the pathway that leads to type I immunity.

Interleukin-19 (IL-19) is a member of the interleukin 10 (IL-10) cytokine family and is produced by B cells and monocytes. IL-19 binds the interleukin 20 receptor complex (IL-20R) to activate STAT3 signaling. Activin receptor type-2B (ACVR2B) is also known as ActR-IIB and MGC116908, ACVR2B is an activin type 2 receptor. Activins are dimeric growth and differentiation factors which belong to the transforming growth factor-beta (TGF-beta) superfamily of structurally related signaling proteins. Activins signal through a heteromeric complex of receptor serine kinases which include at least two type I (I and IB) and two type II (II and IIB) receptors. These receptors are all transmembrane proteins, composed of a ligand-binding extracellular domain with cysteine-rich region, a transmembrane domain, and a cytoplasmic domain with predicted serine/threonine specificity. Type I receptors are essential for signaling; and type II receptors are required for binding ligands and for expression of type I receptors. Type I and II receptors form a stable complex after ligand binding, resulting in phosphorylation of type I receptors by type II receptors. Type II receptors are considered to be constitutively active kinases. This gene encodes activin A type IIB receptor, which displays a 3- to 4-fold higher affinity for the ligand than activin A type II receptor. Stem cell factor (SCF), also known as cKit ligand (KL), mast cell growth factor (MGF), and steel factor (SLF), is a widely expressed 28-40 kDa type I transmembrane glycoprotein. It promotes the survival, differentiation, and mobilisaion of multiple cell types including myeloid, erythroid, megakaryocytic, lymphoid, germ cell, and melanocyte progenitors. SCF is a primary growth and activation factor for mast cells and eosinophils. Noncovalent dimers of transmembrane or soluble SCF interact with the receptor tyrosine kinase SCF R/cKit to trigger receptor dimerisaion and signalling. SCF assists in the recovery of cardiac function following myocardial infarction by increasing the number of cardiomyocytes and vascular channels.

Interleukin-18 (IL-18) is a costimulatory factor for production of interferon-gamma (IFN-gamma) in response to toxic shock and shares functional similarities with IL-12. IL-18 is synthesised as a precursor 24kDa molecule without a signal peptide and must be cleaved to produce an active molecule. IL-1 converting enzyme (ICE; Caspase-1) cleaves pro-IL-18 at aspartic acid in the P1 position, producing the mature, bioactive peptide that is readily released from the cells. It is reported that IL-18 is produced from Kupffer cells, activated macrophages, keratinocytes, intestinal epithelial cells, osteoblasts, adrenal cortex cells and murine diencephalon. IFN-gamma is produced by activated T or NK cells and plays critical roles in the defense against microbiral pathogens. IFN-gamma activates macrophages and enhances NK activity and B cell maturation, proliferation and Ig secretion. IFN-gamma also induces expression of MHC class I and II antigens and inhibits osteoclast activation. IL-18 acts on T helper type-1 (Th1) T cells and in combination with IL-12 strongly induces them to produce IFN-gamma. Pleiotropic effects of IL-18 have also been reported, such as enhancement production of IFN-gamma and GM-CSF in peripheral blood mononuclear cells, production of Th1 cytokines, IL-2, GM-CSF, IFN-gamma in T cells and enhancement of Fas ligand expression by Th1 cells.

Interleukin-35 (IL-35) is a novel IL-12 family cytokine produced by regulatory T cells (Treg) but not by resting or activated effector T cells (Teff). IL-35 is a heterodimeric protein composed of EBI3 (Epstein-Barr-Virus-induced gene 3) and IL-12a (p35). EBI3 is a downstream target of Foxp3, a transcription factor required for Treg-cell development and function, and thus Treg-cell restriction of IL35 occurs. Regulatory T cells are essential for maintaining self tolerance and preventing autoimmunity, and IL-35 is identified as a molecule that mediates the immune suppression function of Treg-cell. As an inhibitory cytokine, IL-35 induces proliferation of Treg-cell populations but suppresses Th17 cell development. Studies in mice show the absence of either IL-35 chain from Treg-cell reduces the cells' ability to suppress inflammation using an experimental model for inflammatory bowel disease. IL-35 is suggested as a potential target of immunotherapy.
