The BCR(KLHL21) E3 ubiquitin ligase complex regulates localisation of the chromosomal passenger complex (CPC) from chromosomes to the spindle midzone in anaphase and mediates the ubiquitination of AURKB. The DnaJ family is one of the largest of all the chaperone families and has evolved with diverse cellular localization and functions. The presence of the J domain defines a protein as a member of the DnaJ family. DnaJ heat shock induced proteins are from the bacterium Escherichia coli and are under the control of the htpR regulatory protein. The DnaJ proteins play a critical role in the HSP 70 chaperone machine by interacting with HSP 70 to stimulate ATP hydrolysis. The proteins contain cysteine rich regions that are composed of zinc fingers that form a peptide binding domain responsible for the chaperone function. DnaJ proteins are important mediators of proteolysis and are involved in the regulation of protein degradation, exocytosis and endocytosis. DnaJB2 (DnaJ homolog subfamily B member 2), also known as HSJ1 or HSPF3, is expressed almost exclusively in the brain, with the highest levels in the frontal cortex and hippocampus. Two isoforms are produced due to alternative splicing.

BAF53b is a member of a family of actin-related proteins (ARPs), which shares significant sequence identity with conventional actins. Both actins and ARPs comprise an actin fold, which forms an ATP binding cleft. BAF53b is a component of the BAF (BRG/brm-associated factor) complex in mammals, which is functionally related to the SWI/SNF complex in S. cerevisiae and Drosophila, and is associated with chromatin remodelling to facilitate transcriptional activation. As neural progenitors exit the cell cycle, the highly homologous BAF53a subunit of the BAF complex is replaced by BAF53b. This subunit switch is essential for the transition from from neural stem cells / progenitors to postmitotic neurons. BAF53b also plays a role in dendritic outgrowth, during which it is required to recruit the BAF complex and the dendritic regulator CREST to specific gene promoters. This function can not be performed by BAF53a.

Has an inhibitory effect on wound healing probably by modulating both growth and migration of primary keratinocytes and by altering the TGF-mediated chemotaxis of monocytes. This effect on wound healing appears to be hormone-sensitive. Regulator of chondrogenesis and osteogenesis and inhibits early healing of bone fractures. Positively regulates PDPK1 kinase activity by stimulating its dissociation from the 14-3-3 protein YWHAQ which acts as a negative regulator.
