Description:
Cell cycle progression is subject to arrest at G1 and G2 checkpoints in response to DNA damage, presumably to allow time for DNA repair prior to entry into S and M phase, respectively. The p53 tumor suppressor is required for one such G1 checkpoint and functions to upregulate expression of GADD 45 and the mitotic inhibitory protein p21. GADD 45 stimulates DNA excision repair in vitro and inhibits entry of cells into S phase, and it apparently acts in concert with GADD 153 in inducing growth arrest. A related DNA-damage inducible gene, GADD 34 synergizes with GADD 45 or GADD 153 in suppressing cell growth. PEG-3 (progression elevated gene-3) shares significant homology with GADD 34 and is inducible by DNA damage. An additional GADD related gene, PA26, is a possible target of p53. Three isoforms of PA26 have been identified as PA26-T1, PA26-T2 and PA26-T3.

CPXM (carboxypeptidase X, member 1) belongs to the peptidase M14 family. However, no carboxypeptidase activity has yet been detected. It may be involved in cell-cell interactions.Members of the M14 metallocarboxypeptidase protein family serve many diverse functions and are divided into three subfamilies based on structure, function and amino acid sequence similarity. Belonging to the N/E subfamily, CPXM (metallocarboxypeptidase CPX-1) is a 734 amino acid protein that contains a F5/8 type C domain and likely binds one zinc ion per subunit. Most members of the N/E subfamily contain several domains, including an active carboxypeptidase domain and signal peptide, and are thought to function mostly in protein-protein interactions and/or protein-membrane interactions, thereby targeting the protein to specific locations within the secretory pathway. CPXM is a unique member of this subfamily in that it does not appear to exhibit any enzymatic activity due to lack of several active-site residues that are present in the catalytic domain of other members of the N/E subfamily. Studies showing that CPXM expression is regulated during osteoclastogenesis suggest that CPXM may play a role in osteoclast differentiation. There are two isoforms of CPXM which are a result of alternative splicing events.

FGFR1OP2 belongs to the SIKE family. The FGFR1OP2 (FGFR1 oncogene partner 2) gene was identified through its involvement in a fusion with the FGFR1 gene. FGFR1OP2 may be involved in the wound healing pathway. It is expressed in bone marrow, spleen and thymus. A chromosomal aberration involving FGFR1OP2 may be a cause of stem cell myeloproliferative disorder (MPD). Insertion ins(12;8)(p11;p11p22) with FGFR1. MPD is characterized by myeloid hyperplasia, eosinophilia and T cell or B cell lymphoblastic lymphoma. In general it progresses to acute myeloid leukemia. The fusion protein FGFR1OP2-FGFR1 may exhibit constitutive kinase activity and be responsible for the transforming activity.

Chromosome 20 contains a region with numerous genes expressed in the epididymis which are thought important for seminal production and some viewed as potential targets for male contraception. The PRNP gene encoding the prion protein associated with spongiform encephalopathies, like Creutzfeldt-Jakob disease, is found on chromosome 20. Amyotrophic lateral sclerosis, spinal muscular atrophy, ring chromosome 20 epilepsy syndrome and Alagille syndrome are also associated with chromosome 20.

Recognizes a protein of 21 kDa, identified as the Bax protein. This MAb is highly specific to Bax and shows no cross-reaction with Bcl-2 or Bcl-X protein. Bcl-2 blocks cell death following a variety of stimuli. Bax has extensive amino acid homology with Bcl-2 and it homodimerizes and forms heterodimers with Bcl-2. Overexpression of Bax accelerates apoptotic death induced by cytokine deprivation in an IL-3 dependent cell line, and Bax also counters the death repressor activity of Bcl-2.

ACTH (same as Corticotropin) is a 39 amino acid active peptide produced by the anterior pituitary. This MAb is specific to Synacthen (aa1-24 of ACTH); does not react with CLIP (aa17-39 of ACTH). POMC (pro-opiomelanocortin or corticotropin-lipotropin) is a 267 amino acid polypeptide hormone precursor that goes through extensive, tissue-specific posttranslational processing by convertases. POMC is cleaved into ten hormone chains named NPP, ACTH, alpha-MSH (Melanocyte Stimulating Hormone), beta-MSH, gamma-MSH, CLIP (corticotropin-like intermediary peptide), Lipotropin-beta, Lipotropin-gamma, beta-endorphin and Met-enkephalin. ACTH is also produced by cells of immune system (T-cells, B-cells, and macrophages) in response to stimuli associated with stress. Anti-ACTH is a useful marker in classification of pituitary tumors and the study of pituitary disease. It reacts with ACTH-producing cells (corticotrophs).It also may react with other tumors (e.g. some small cell carcinomas of the lung) causing paraneoplastic syndromes by secreting ACTH.
