APS (adapter protein with Pleckstrin homology and Src homology 2 domains) is a member of the Lnk family, an adaptor protein that is involved in B cell signaling, insulin signaling and cytoskeletal reorganisation. A PH and an SH2 domain containing adaptor protein that links activated tyrosine kinases to signaling pathways. It is tyrosine phosphorylated by JAK2, KIT and other kinases during B cell receptor stimulation of many different cytokines, chemokines and leukokines. APS has been shown to inhibit the JAK STAT pathway in collaboration with cCbl.

The RAF family of signaling proteins has three isoforms: A RAF, RAF 1 and B RAF. These proteins are instrumental in forwarding the stimulus generated by mitogens from monomeric G proteins to the MAP kinase pathway. RAF 1 is known to be downstream of Ras and binds to it by an N terminal noncatalytic domain. This domain is conserved in A RAF and B RAF. 14-3-3 is also known to be an activator of RAFs. Activated RAF directly phosphorylates and activates MEK. A RAF is expressed at a very high concentration in urogenital and kidney tissues.

Human Rad1 is a component of a heterotrimeric PCNA like complex that also contains the Rad9 and Hus1 proteins. This complex is believed to be involved in cellular responses to DNA damage, possibly by associating with Rad17 and several components of the PCNA-loading heteropentamer, replication factor C. Human Rad1 exhibits significant homology to Rad1 from S. pombe, and its expression in yeast rad1 mutants has been shown to partially restore radiation resistance and G2 checkpoint activity. CDK5 (Cyclin Dependent Kinase 5) is serine/threonine kinase involved in synaptic regulation and neuronal development; phosphorylates synaptic protein Pctaire1; regulates acetylcholine receptor expression. CDK5 is a member of the cyclindependent kinase family of serine/threonine kinases. It is present in numerous mammalian tissues including kidney, testes, and ovary. Its activity is detected almost exclusively in brain extracts. Neuronal and muscle cells contain the highest amount of this protein. Similar to other Cdks, monomeric Cdk5 displays no enzymatic activity, but Cdk5 is not activated by cyclins. Instead, the p35 protein, which is expressed solely in the brain, activates Cdk5. Cdk5 interacts with D1 and D3 type G1 cyclins and can phosphorylate histone H1, TAU, MAP2 and NF-H and NF-M. Cdk5 activity is involved in terminal differentiation of neurons and muscle cells.

BIVM is highly charged and localizes to the cytoplasm and nucleus where it may bind to either DNA or RNA or associate with other cellular proteins. There are two forms of glutamic acid decarboxylases (GADs) that exist in brain: GAD-65 (also known as GAD2) and GAD-67 (also known as GAD1, GAD or SCP). GAD-65 and GAD-67 are members of the group II decarboxylase family of proteins and are responsible for catalyzing the rate limiting step in the production of GABA (gamma-aminobutyric acid) from L-glutamic acid. Although both GADs are found in brain, GAD-65 localizes to synaptic vesicle membranes in nerve terminals, while GAD-67 is distributed throughout the cell. GAD-67 is responsible for the basal levels of GABA synthesis. In the case of a heightened demand for GABA in neurotransmission, GAD-65 transiently activates to assist in GABA production. As a member of the group II decarboxylase family, GADL1 (Glutamate decarboxylase-like protein 1) is a 521 amino acid protein that utilizes pyridoxal phosphate as a cofactor for its carboxylase activity. There are two isoforms of GADL1 that exist as a result of alternative splicing events.
