Vascular wall-linked protein, or Vasculin, is primarily expressed in the arterial wall and in plasma. It is also differentially expressed in human atherogenesis. Alternative splicing of exon 3 of the Vasculin gene produces three variants. Vasculin binds to and activates the minimal self-sufficient promoter element (MSPE) of the mouse Ada gene promoter and binds to and partially suppresses the GC-rich promoter of the nonhomologous human TOP2A gene promoter. It acts as a nuclear factor that can form complexes with TATA-binding proteins, transcription factors TFIIB and TFIIF, RNA polymerase II and p300. The regulated expression of Vasculin in plaques suggests that it may be involved in atherogenesis, and its presence in plasma may implicate Vasculin as a marker for atherosclerosis.

Description:
The complement pathway is an important host defense system that contributes to both innate and acquired immunity. There are three pathways of complement activation: the classical pathway, lectin pathway and alternative pathway. Complement protein Factor I is a key serine protease that modulates the complement cascade by regulating the levels of C3 convertases. It circulates in plasma as a heavily N-glycosylated heterodimer made up of two disulfide linked chains, each carrying three N-linked oligosaccharide chains that may have both structural and functional roles in the interactions with the natural substrate and the cofactor during catalysis. Factor I is a serine protease with a high degree of specificity for C3b and C4b. It requires protein cofactors for cleavage of these complement proteins; Factor H, CR1 or MCP are required for C3b cleavage, and C4bp or CR1 are required for C4b cleavage.

CNTD2 (cyclin N-terminal domain containing 2) is a 155 amino acid protein that contains one cyclin N-terminal domain and is encoded by a gene that maps to human chromosome 19q13.2. Chromosome 19 consists of approximately 63 million bases and makes up over 2% of human genomic DNA. Chromosome 19 is recognized for having the greatest gene density of the human chromosomes. It is the genetic home for a number of immunoglobulin superfamily members, including killer cell and leukocyte Ig-like receptors, various ICAMs, the CEACAM and PSG families, and Fc?receptors. Key genes for eye color and hair color also map to chromosome 19. Peutz-Jeghers syndrome, spinocerebellar ataxia type 6, the stroke disorder CADASIL, hypercholesterolemia and insulin-dependent diabetes have been linked to chromosome 19. The β-Amyloid protein precursor (AbPP) is a widely expressed transmembrane protein that is processed into the b-Amyloid (Ab) peptide, which accumulates in insoluble plaques in the brain of Alzheimer’s disease patients and AbPP intracellular domain (AID). AID may function as a pro-apoptotic peptide, a regulator of calcium homeostasis and a molecule involved in transcriptional regulation. The AID associated protein 1 (AIDA-1) is highly expressed in the brain and is regulated by AbPP. It interacts with AbPP to play a role in brain development. AIDA-1 also interacts with coilin in Cajal bodies to regulate pre-mRNA splicing.
