Component of the epithelial apical junction complex that may function as an homophilic cell adhesion molecule and is essential for tight junction integrity. Also involved in transepithelial migration of leukocytes through adhesive interactions with AMICA1/JAML a transmembrane protein of the plasma membrane of leukocytes. The interaction between both receptors also mediates the activation of gamma-delta T-cells, a subpopulation of T-cells residing in epithelia and involved in tissue homeostasis and repair. Upon epithelial CXADR-binding, AMICA1 induces downstream cell signaling events in gamma-delta T-cells through PI3-kinase and MAP kinases. Spermatogenesis is the process by which male spermatogonia develop into mature spermatozoa. DAZ (deleted in azoospermia) are RNA-binding proteins that play an essential role in spermatogenesis. DAZ proteins influence the first stages of spermatogenesis and the maintenance of germ cell populations. DAZ proteins (DAZ1, DAZ2, DAZ3, DAZ4 and DAZ5) are encoded by separate genes on chromosome Y, each of which contain an AZFc domain in their coding region. DAZ proteins localize to the nucleus of spermatogonia, but relocate to the cytoplasm during meiosis. DAZ proteins contain an RRM (RNA recognition motif) domain that may regulate mRNA translation by binding to the 3’UTR. Deletions in the genes encoding DAZ proteins may cause azoospermia or oligospermia which can lead to male infertility. DAZ4 (deleted in azoospermia 4), also known as pDP1680 or pDP1681, is a 579 amino acid testis specific protein that contains nine DAZ-like domains and two RNA recognition motifs (RRM). DAZ4 exists as two alternatively spliced isoforms.

Its anti-apoptotic activity is mediated through the inhibition of CASP3, CASP7 and CASP9, as well as by its E3 ubiquitin-protein ligase activity. As it is a weak caspase inhibitor, its anti-apoptotic activity is thought to be due to its ability to ubiquitinate DIABLO/SMAC targeting it for degradation thereby promoting cell survival. May contribute to caspase inhibition, by blocking the ability of DIABLO/SMAC to disrupt XIAP/BIRC4-caspase interactions. Protects against apoptosis induced by TNF or by chemical agents such as adriamycin, etoposide or staurosporine. Suppression of apoptosis is mediated by activation of MAPK8/JNK1, and possibly also of MAPK9/JNK2. This activation depends on TAB1 and NR2C2/TAK1. In vitro, inhibits CASP3 and proteolytic activation of pro-CASP9. Isoform 1 blocks staurosporine-induced apoptosis. Isoform 2 blocks etoposide-induced apoptosis. Isoform 2 protects against natural killer (NK) cell killing whereas isoform 1 augments killing.

High density lipoproteins (HDLs) have been proposed to function jointly with lecithin:cholesterol acyltransferase and CETP to facilitate cholesterol transport from tissues to the liver. This mechanism, referred to as reverse cholesterol transport, is physiologically important because it maintains systemic cholesterol levels. CETP is responsible for neutral lipid transfer activity in plasma in numerous species. Since CETP is able to accelerate specifically the exchange of lipid components between pro- and anti-atherogenic lipoprotein fractions, it may be a key determinant of the global atherogenicity of the plasma lipoprotein profile and arises as a possible target in atherosclerosis prevention. CETP has an important role in reverse cholesterol transport and shaping and affecting the composition of plasma lipoproteins. In general elevated levels of CETP have been associated with increased risk of coronary heart disease.

Olfactory sensory neurons contain olfactory receptors, which are G protein-coupled receptor proteins that localize to the cilia and display affinity for and bind to a variety of odor molecules. Olfactory neurons send their axons from the olfactory epithelium to the olfactory bulb, which is covered by the CNS basal lamina. FEZF1 (Fez family zinc finger protein 1), also known as Forebrain Embryonic Zinc Finger and Zinc finger protein 312B, is a 475 amino acid nuclear protein that is expressed in the olfactory epithelium and hypothalamus of mice. In FEZF1-deficient mice, axons of olfactory neurons do not reach the olfactory bulb, suggesting that FEXF1 is required for the penetration of olfactory axons though the basal lamina before innervation of the olfactory bulb. When FEZF1 translocates to the nucleus, it induces KRAS overexpression, resulting in activation of ERK-signaling. Overexpression of FEZF1 leads to accelerated proliferation in cultured cells and increased tumor mass in mice. There are three isoforms of FEZF1 that are produced as a result of alternative splicing events.
