Description:   Recognizes the epsilon-chain of CD3 (Workshop V; Code: CD03.09), which consists of five different polypeptide chains (designated as γ, δ, ε, ζ, and η) with MW ranging from 16-28 kDa. The CD3 complex is closely associated at the lymphocyte cell surface with the T cell antigen receptor (TCR). Reportedly, CD3 complex is involved in signal transduction to the T cell interior following antigen recognition. The CD3 antigen is first detectable in early thymocytes and probably represents one of the earliest signs of commitment to the T cell lineage. In cortical thymocytes, CD3 is predominantly intra-cytoplasmic. However, in medullary thymocytes, it appears on the T cell surface. CD3 antigen is a highly specific marker for T cells, and is present in majority of T cell neoplasms.

Description:   Recognizes a protein of 12 kDa, identified as microglobulin. Major histocompatibility complex (MHC) class 1 molecules bind to antigens for presentation on the surface of cells. The proteasome is responsible for producing these antigens from the components of foreign pathogens. MHC class 1 molecules consist of an α heavy chain that contains three subdomains (α1, α2, α3) and a non-covalent associating light chain, known as β-2-Microglobulin. β-2-Microglobulin associates with the α3 subdomain of the α heavy chain and forms an immunoglobulin domain-like structure that mediates proper folding and expression of MHC class 1 molecules. The α1 and α2 domains of the α heavy chain form the peptide antigen-binding cleft. Mutations in the β-2-Microglobulin gene can enhance the progression of malignant melanoma phenotypes.

Description:   Recognizes a polypeptide which is identified as insulin, a 51-amino acid polypeptide composed of A and B chains connected through the C-peptide. Proinsulin, which has very little biological activity, is cleaved by proteases within its cell of origin into the insulin molecule and the C-terminal basic residue. Insulin enhances membrane transport of glucose, amino acids, and certain ions. It also promotes glycogen storage, formation of triglycerides, and synthesis of proteins and nucleic acids. Deficiency of insulin results in diabetes mellitus. The main storage site for insulin is the pancreatic islets. Antibodies to insulin are important as beta-cell and insulinoma marker.

Description:   This MAb reacts with a reduction-resistant epitope present in both free and SIgA bound Secretory Component. It does not react with the cell lines lacking secretory component. The antibody is useful for studying the distribution and level of both free and bound secretory component. Secretory component is differentially expressed in epithelium, and the antibody is a popular marker for identifying subpopulations of epithelial cells and epithelial differentiation. The Secretory component antibody is a useful research tool for studying mucosal immunity, inflammation, remodeling, differentiation and tumorigenesis, all processes associated with differential secretory component expression.

Description:   This MAb reacts with an extracellular domain (close to transmembrane) of CD56/NCAM. Three isoforms of neural cell adhesion molecule (NCAM) are produced by differential splicing of the RNA transcript from a single gene. The 135 kDa isoform is the basic molecule, which is glycosylated or sialylated to produce the mature species. Anti-CD56 recognizes two proteins of the neural cell adhesion molecule, the basic molecule expressed on most neuroectodermally derived tissues and neoplasms (e.g. retinoblastoma, medulloblastomas, astrocytomas, neuroblastomas, and small cell carcinomas). It is also expressed on some mesodermally derived tumors (rhabdomyosarcoma). Anti-CD56 plays an important role in the diagnosis of nodal and nasal NK/T-cell lymphomas.

Description:   Myogenin is a member of the MyoD family of myogenic basic helix-loop-helix (bHLH) transcription factors that also includes MyoD, Myf-5, and MRF4 (also known as herculinor Myf-6). MyoD family members are expressed exclusively in skeletal muscle and play a key role in activating myogenesis by binding to enhancer sequences of muscle-specific genes. The regulatory domain of MyoD is approximately 70 amino acids in length and includes both a basic DNA binding motif and a bHLH dimerization motif. MyoD family members share about 80% amino acid homology in their bHLH motifs. Anti-myogenin labels the nuclei of myoblasts in developing muscle tissue, and is expressed in tumor cell nuclei of rhabdomyosarcoma and some leiomyosarcomas. Positive nuclear staining may occur in Wilms' tumor.

Description:   This MAb recognizes the peptide core of gastric mucin M1 (recently identified as Mucin 5AC). Its epitope is destroyed by beta-mercaptoethanol but not by periodate treatment. This mucin is present in primary ovarian mucinous cancer but usually absent in colorectal adenocarcinoma, thus showing an expression pattern opposite to MUC2. Together with a panel of antibodies, Anti-MUC5AC may be useful for differential identification of primary mucinous ovarian tumors from colon adenocarcinoma metastatic to the ovary. MUC5AC antibodies may also be useful for identification of intestinal metaplasia as well as in the identification of pancreatic carcinoma and pre-cancerous changes vs. normal pancreas.

Description:   Recognizes a protein of 104 kDa-110 kDa, characterized as major vault protein (MVP). Vaults are large ribonucleoprotein particles (RNPs) present in all eukaryotic cells. They have a complex morphology, including several small molecules of RNA, but a single protein species. The MVP accounts for >70% of their mass. Their shape is reminiscent of the nucleopore central plug. Treatment of cells with estradiol increases the amount of MVP in nuclear extract. The hormone-dependent interaction of vaults with ER is prevented in vitro by sodium molybdate. Antibodies to estrogen, progesterone and glucocorticoid receptors are able to co-immunoprecipitate the MVP. MVP is overexpressed in many neoplastic tissues and cell lines. Expression of MVP predicts a poor response to chemotherapy.

Description:   This MAb is specific to lambda light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the lambda light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is malignant.

Description:   Recognizes a 180-185 kDa protein, identified as isoform of leukocyte common antigen (CD45RO) (4th Leucocyte Typing Workshop: Code No. N31). The epitope recognized by this antibody is sensitive to neuraminidase digestion. This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. It rarely stains NK cells or B-cell lymphomas.

Description:   Recognizes an N-glycosylated glycoprotein of 120 kDa with intra-chain disulfide bonds, identified as CD50 or ICAM-3. CD50 is the major ligand for LFA-1 (CD11a/CD18) and may have signaling role to increase adhesion. It is expressed on thymocytes and T lymphocytes and is resistant to treatment with phosphatidylinositol (PI) phospholipase C. This MAb is excellent for staining of formalin/paraffin tissues.

Description:   Recognizes a protein of 27 kDa, identified as the Bcl-X protein. This MAb shows no cross-reaction with Bcl-2 or Bax protein. Bcl-X has two isoforms, Bcl-XL (long), a 241 amino acid protein which suppresses cell death. And Bcl-XS (short), a 178 amino acid protein lacking a 63 amino acid domain which functions as a dominant inhibitor of Bcl-2. This MAb reacts with both Bcl-XS and Bcl-XL proteins.

HMFG is present on normal human breast epithelial cells and cell lines derived from breast carcinomas, as well as to the outer surface of the human milk fat globule. HMFG is considered as a differentiation marker. It is useful as specific breast epithelial marker and can also provide a tool to study the role of the cell surface in normal and neoplastic mammary development.
