CD31 (PECAM-1) is a transmembrane glycoprotein member of the immunoglobulin supergene family of adhesion molecules. CD31 is expressed by stem cells of the hematopoietic system and is primarily used to identify and concentrate these cells for experimental studies as well as for bone marrow transplantation. Anti-CD31 has shown to be highly specific and sensitive for vascular endothelial cells. Staining of nonvascular tumors (excluding hematopoietic neoplasms) is rare. CD31 MAb reacts with normal, benign, and malignant endothelial cells which make up blood vessel lining. The level of CD31 expression can help to determine the degree of tumor angiogenesis, and a high level of CD31 expression may imply a rapidly growing tumor and potentially a predictor of tumor recurrence.

Recognizes a glycoprotein of 44-88 kDa, which is identified as CD28. It is the critical T-cell co-stimulatory receptor which provides to the cell the important second activation signal by binding CD80 and CD86 that are expressed by antigen presenting cells. Besides its co-stimulation role, CD28 functions in preventing T-cells from anergic hyporesponsive state or from undergoing premature apoptotic cell death. CD28 is also expressed on human fetal NK cells and some NK cell lines, whereas on murine NK cells the CD28 expression is much broader.

Recognizes a protein of a disulfide-linked 120 kDa dimer, identified as CD27 (Workshop VI; Code 6T-028). CD27 is expressed on the majority of peripheral T cells, medullary thymocytes, memory-type B cells, and natural killer cells. It is a transmembrane phosphoglycoprotein that belongs to the tumor necrosis factor receptor (TNFR) superfamily. CD27 binds to its ligand CD70, a member of the TNF family, and induces T-cell co-stimulation and B-cell activation. It also interacts with TRAFs and is involved in activation of NFB and SAPK/JNK and induces apoptosis.

This MAb reacts with the beta-chain of HLA-DRB1 antigen, a member of MHC class II molecules. It does not cross react with HLA-DP and HLA-DQ. Its epitope is different from that of MAb L243. HLA-DR is a heterodimeric cell surface glycoprotein comprised of a 36 kDa alpha (heavy) chain and a 28 kDa beta (light) chain. It is expressed on B-cells, activated T-cells, monocytes/macrophages, dendritic cells and other non-professional APCs. In conjunction with the CD3/TCR complex and CD4 molecules, HLA-DR is critical for efficient peptide presentation to CD4 T cells. It is an excellent histiocytic marker in paraffin sections producing intense cytoplasmic staining. True histiocytic neoplasms are similarly positive. HLA-DR antigens also occur on a variety of epithelial cells and their corresponding neoplastic counterparts. Loss of HLA-DR expression is related to tumor microenvironment and predicts adverse outcome in diffuse large B-cell lymphoma.

No reactivity has been observed with reactive B-cells. So a B-lineage population that co-expresses CD43 is highly likely to be a malignant lymphoma, especially a low-grade lymphoma, rather than a reactive B-cell population. When CD43 antibody is used in combination with anti-CD20, effective immunophenotyping of the lymphomas in formalin-fixed tissues can be obtained. Co-staining of a lymphoid infiltrate with anti-CD20 and anti-CD43 argues against a reactive process and favors a diagnosis of lymphoma.

Hepatocyte Paraffin 1 or HepPar1 localizes to the mitochondria of hepatocytes. It is a sensitive marker for distinguishing hepatocellular carcinomas (HCC) from other metastatic carcinomas as well as cholangio-carcinomas. HCC s occur primarily in the stomach, but they are also found in many other organs. The Hepatocyte Specific Antigen may also be a useful marker for intestinal metaplasia. Reportedly, strong expression of the Hepatocyte Specific Antigen correlates with smaller tumor size and longer patient survival. Occasionally, Hepatocyte Specific Antigen is also found in gastric carcinomas as well as in a few other non-hepatic tumors.

Recognizes a protein of 80 kDa-90 kDa, identified as CD36 (Workshop IV; Code P-26). Its epitope maps between aa155-183. It is expressed on platelets, monocytes and macrophages, microvascular endothelial cells, erythrocyte precursors, mammary epithelial cells, and some macrophage derived dendritic cells. CD36 acts as a receptor for thrombospondin (TSP), collagen types I, IV and V, P. falciparum malaria-infected erythrocytes, and sickle erythrocytes. It also functions as a scavenger receptor, mediating macrophage uptake of oxidized low-density lipoprotein (LDL) and recognition of apoptotic polymorphonuclear leukocytes (PMN). CD36 plays a role in platelet aggregation, macrophage foam cell development, inflammation, and the tissue ischemia observed in sickle cell disease and cerebral malaria. Note that 1-4% of Japanese and East Asia population lack CD36. This MAb blocks adhesion of P. falciparum parasitized red blood cells to CD36 and strongly inhibits collagen-induced platelet aggregation.

The antigen recognized by PNL2 is different from Melan A and gp100. Its epitope is not destroyed by digestion with neuraminidase i.e. its epitope id not glycosylated. Anti-PNL2 may be most useful because of its high sensitivity for metastatic melanoma (87%), as opposed to 76% for anti-HMB45 and 82% for anti-MART-1. Anti-PNL2 labels intra-epidermal nevi while the dermal component of compound nevi are largely non-reactive with anti-PNL2. Antibodies against PNL2, MART-1 (Melan A) and HMB45 stain most clear cell sarcoma cells and a few cells in angio-myolipomas and lymphangioleiomyomatosis. Anti-PNL2 is a useful antibody for the identification of melanomas and clear cell sarcomas. Differential diagnosis is aided by the results from a panel of antibodies, including antibodies against HMB45, MART-1, tyrosinase, and MiTF.

Recognizes a protein of 104 kDa-110 kDa, characterized as major vault protein (MVP). Vaults are large ribonucleoprotein particles (RNPs) present in all eukaryotic cells. They have a complex morphology, including several small molecules of RNA, but a single protein species. The MVP accounts for >70% of their mass. Their shape is reminiscent of the nucleopore central plug. Treatment of cells with estradiol increases the amount of MVP in nuclear extract. The hormone-dependent interaction of vaults with ER is prevented in vitro by sodium molybdate. Antibodies to estrogen, progesterone and glucocorticoid receptors are able to co-immunoprecipitate the MVP. MVP is overexpressed in many neoplastic tissues and cell lines. Expression of MVP predicts a poor response to chemotherapy.

This MAb recognizes a 60 kDa antigen associated with the mitochondria in human cells. It is a part of a new panel of reagents, which recognizes subcellular organelles or compartments of human cells. These markers may be useful in identification of these organelles in cells, tissues, and biochemical preparations. It recognizes an antigen associated with the mitochondria in human cells only. It can be used to stain the mitochondria in cell or tissue preparations and can be used as a mitochondrial marker in subcellular fractions. It produces a spaghetti-like pattern in normal and malignant cells and may be used to stain mitochondria of cells in fixed or frozen tissue sections. It can also be used with paraformaldehyde fixed frozen tissue or cell preparations. This MAb is an excellent marker for human cells in xenographic model research. It reacts specifically with human cells, including neurons and embryonic stem cells.

This MAb recognizes protein of 26 kDa-60 kDa, which is identified as CD63. Its epitope is different from that of MAb LAMP3/529. The tetraspanins are integral membrane proteins expressed on cell surface and granular membranes of hematopoietic cells and are components of multi-molecular complexes with specific integrins. The tetraspanin CD63 is a lysosomal membrane glycoprotein that translocates to the plasma membrane after platelet activation. CD63 is expressed on activated platelets, monocytes and macrophages, and is weakly expressed on granulocytes, T cell and B cells. It is located on the basophilic granule membranes and on the plasma membranes of lymphocytes and granulocytes. CD63 is a member of the TM4 superfamily of leukocyte glycoproteins that includes CD9, CD37 and CD53, which contain four transmembrane regions. CD63 may play a role in phagocytic and intracellular lysosome-phagosome fusion events. CD63 deficiency is associated with Hermansky-Pudlak syndrome and is strongly expressed during the early stages of melanoma progression.
