Description:   This antibody reacts with both SUMO2 and SUMO3. The small ubiquitin-related modifier (SUMO) proteins, which include SUMO1, 2 and 3, belong to the ubiquitin-like protein family. Like ubiquitin, the SUMO proteins are synthesised as precursors that undergo processing before conjugation to target proteins. Also, both utilize the E1, E2 and E3 cascade enzymes for conjugation. However, SUMO and ubiquitin differ with respect to targeting. Ubiquitination predominantly targets proteins for degradation, whereas sumoylation targets for a variety of cellular processing, including nuclear transport, transcriptional regulation, apoptosis and protein stability. The unconjugated SUMO1/2/3 proteins localize to the nuclear membrane, nuclear bodies and cytoplasm, respectively. SUMO1 utilizes Ubc9 for conjugation to several targets, which include MDM2, p53, Pml and RanGap1. SUMO2/3 contribute to a greater percentage of protein modification than does SUMO1 and they can form polymeric chains. In addition, SUMO3 regulates beta-Amyloid generation and may be critical in the onset or progression of Alzheimer’s disease.

Description:   PAX genes contain paired domains with strong homology to genes in Drosophila, which are involved in programming early development. Lesions in the PAX6 gene account for most cases of aniridia, a congenital malformation of the eye, chiefly characterised by iris hypoplasia, which can cause blindness. PAX6 is involved in other anterior segment malformations besides aniridia, such as Peters’ anomaly, a major error in the embryonic development of the eye with corneal clouding with variable iridolenticulocorneal adhesions. The PAX6 gene encodes a transcriptional regulator that recognises target genes through its paired-type DNA-binding domain. The paired domain is composed of two distinct DNA-binding subdomains, the amino-terminal subdomain and the carboxy-terminal subdomain, which bind respective consensus DNA sequences. The human PAX6 gene produces two alternatively spliced isoforms that have the distinct structure of the paired domain.

Description:   p40 (p63 delta) is a marker recently determined to be highly specific for squamous basal cells in the immunohistochemistry (IHC) application. The current more routinely recommended marker, p63, appears to have less specificity compared to p40, especially on squamous cell tumors. The ability to differentiate between lung adenocarcinoma vs. squamous cell carcinoma is difficult and has bearing on the different therapeutic avenues for each subtype treatment. p40 antibody’s ability to distinguish between the tumor types appears to be superior when compared to p63. The ability to utilize a p40 antibody probe as a squamous cell marker bolsters its use for future sub-classification of lung cancers, especially by immunohistochemical techniques.

Description:   Peripheral blood granulocytes are classified into neutrophils, basophils and eosinophils according to the staining characteristics of their cytoplasmic granules. Granule proteins are released by physiologic and pharmacologic stimuli and play important roles in both normal and pathological host immune responses. Eosinophil major basic protein and eosinophil peroxidase (EPX) are granule proteins specific to the eosinophil. AHE-1 recognises human EPX, a granule protein specific to eosinophils. It does not cross-react with eosinophil major basic protein, elastase, cathepsin G, esterase N, thrombin, plasmin, kallikrein, lactoferrin, or transferrin. This MAb stains eosinophils only and does not stain other peripheral blood cells, including platelets, neutrophils, monocytes, lymphocytes or red blood cells. Human EPX gene product can form a tetramer of two light chains and two heavy chains. Other peroxidase family members include myeloperoxidase (MPO), lactoperoxidase (LPO), and thyroid peroxidase (TPO).

Description:   This antibody specifically reacts with a protein of 20-30 kDa, identified as PGP9.5, also known as ubiquitin carboxyl-terminal hydrolase-1 (UchL1). Initially, expression in normal tissues was reported in neurons and neuroendocrine cells but later it was found in distal renal tubular epithelium, spermatogonia, Leydig cells, oocytes, melanocytes, prostatic secretory epithelium, ejaculatory duct cells, epididymis, mammary epithelial cells, Merkel cells, and dermal fibroblasts. PGP9.5 antibody has been used to show the protein to be a useful prognostic indicator of colorectal, pancreatic and gall bladder cancer. A mutation in the gene is believed to cause a form of Parkinson's disease.
