Description:   PMP22 is a 22 kDa glycoprotein expressed in the compact myelin of the peripheral nervous system. In the peripheral nervous system, PMP 22 is produced by myelinating Schwann cells and is coexpressed with the genes for myelin basic protein (MBP) during nerve development and regeneration. Alterations in the level of this protein cause several genetic human diseases. If the protein is duplicated, patients develop Charcot Marie Tooth disease. If one copy of the gene is deleted, they suffer from the inherited tendency to pressure palsies.

Description:   Catalyzes the transfer of sulfate to position 6 of galactose residues of keratan. It has a preference for sulfating keratan sulfate, but it also transfers sulfate to the unsulfated polymer. The sulfotransferase activity on sialyl LacNAc structures is much higher than the corresponding desialylated substrate, and only internal galactose residues are sulfated. It also may function in the sulfation of sialyl N-acetyllactosamine oligosaccharide chains attached to glycoproteins. It participates in biosynthesis of selectin ligands. Selectin ligands are present in high endothelial cells (HEVs) and play a central role in lymphocyte homing at sites of inflammation. Aggrecan is the major proteoglycan of human articular cartilage. The core protein is substituted by a number of keratan sulfate and chondroitin sulfate glycosaminoglycan chains. Whereas chondroitin sulfate is widely distributed throughout the body, keratan sulfate is primarily expressed in cartilage (joints, trachea, intervertebral discs) and cornea.

Description:   GAD-65 and GAD-67, glutamate decarboxylases, function to catalyse the production of GABA (gamma-aminobutyric acid). In the central nervous system GABA functions as the main inhibitory transmitter by increasing a Cl- conductance that inhibits neuronal firing. GABA has been shown to activate both ionotropic (GABAA) and metabotropic (GABAB) receptors as well as a third class of receptors called GABAC. Both GABAA and GABAC are ligand-gated ion channels, however, they are structurally and functionally distinct. Members of the GABAA receptor family include GABAA R Alpha 1-6, GABAA R Beta 1-3, GABAA R Gamma 1-3, GABAA R Delta, GABAA R Epsilon, GABAA R Zeta 1 and GABAA R Zeta 2. The GABAB family is composed of GABAB R1 Alpha and GABAB R1 Beta. GABA transporters have also been identified and include GABA T-1, GABA T-2 and GABA T-3 (also designated GAT-1, -2, and -3). The GABA transporters function to terminate GABA action.

Description:   In the central nervous system GABA functions as the main inhibitory transmitter by increasing a Cl-conductance that inhibits neuronal firing. GABA has been shown to activate both ionotropic (GABAA ) and metabotropic (GABAB ) receptors as well as a third class of receptors called GABAC. In addition to GABA receptors, several proteins have been identified as regulators of GABA function, including GAD65, GAD67, GABA transporters and GABARAP (GABAA receptor-associated protein). GABARAP associates with GABAA Rg2 to link GABAA receptors to the cytoskeleton. The GABARAP protein sequence is similar to light chain-3 of microtubule-associated proteins (MAPs) suggesting that it may be a type of MAP or a component of a MAP complex.

Description:   Members of the bactericidal/permeability-increasing protein family have antimicrobial properties and bind lipophilic substances, therefore targeting gram-negative bacteria. The bactericidal permeability increasing protein (BPI) is an antibacterial and endotoxin-neutralizing molecule that is abundant in the granules of polymorphonuclear leukocytes (neutrophil granules). Sharing structural and sequence homologies with BPI, BPIL3 (Bactericidal/permeability-increasing protein-like 3) is a 453 amino acid secreted protein that contains the family’s common conserved feature of two cysteine residues that are critical for protein function. While BPIL3 is primarily expressed at low levels in tonsils, it has been found to be upregulated in hypertrophic tonsil tissue, suggesting that it may play a role in the pathogenesis of inflamed disease tissue.

Description:   SH3KBP1 belongs to the CIN85/CMS family of adaptor molecules, characterized by containing three N-terminal Src homology domains, a proline rich region and a C-terminal coiled-coil domain. The different members of the family orchestrate a network involved in dowregulation and degradation of recpetor tyrosine kinases. SH3KBP1 is involved in regulating diverse signal transduction pathways. Involved in the regulation of endocytosis and lysosomal degradation of ligand-induced receptor tyrosine kinases, including EGFR and MET/hepatocyte growth factor receptor, through a association with CBL and endophilins. In the case of EGF receptor turnover, its activation involves recruitment of SH3KBP1- endophilin complexes to mediate internalization. Once internalized, RTKs are delivered into the endosomal compartment where receptors get sorted for either recycling back to the cell surface or are targeted to lysosomes for degradation. Alternate splicing results in multiple transcript variants.

Description:   Adenosine deaminase is an enzyme that is present in most tissues and exists predominantly as a monomer, although in some tissues it is associated with adenosine deaminase-binding protein. Adenosine deaminase degrades extracellular adenosine, which is toxic for lymphocytes. A novel family of growth factors that share sequence similarity to adenosine deaminase has been identified. The cat eye syndrome critical region protein (CECR) family includes CECR1, CECR2, CECR3, CECR4, CECR5, CECR6, CECR7, CECR8 and CECR9. The genes encoding CECR proteins are candidates for Cat Eye Syndrome (CES), a developmental disorder associated with the duplication of a 2 Mb region of 22q11.2. CES is characterized by the combination of coloboma of the iris and anal atresia with fistula, downslanting palpebral fissures, preauricular tags and/or pits, frequent occurrence of heart and renal malformations, and normal or near-normal mental development. CECR family members are widely expressed. Specifically, CECR1 has the highest expression in adult heart, lung, lymphoblasts and placenta. CECR2 is also involved in neurulation and chromatin remodeling. Mutations in the CECR2 gene result in neural tube defects.

Description:   Bad is a member of the Bcl2 family and acts to promote apoptosis by forming heterodimers with the survival proteins Bcl2 and BclxL, thus preventing them from binding with BAX. Bad is found on the outer mitochondrial membrane and, once phosphorylated in response to growth stimuli, translocates to the cytoplasm. The phosphorylation status of Bad represents a key checkpoint for death or cell survival. JNK-induced phosphorylation of BAD serine 128 promotes the apoptotic role of Bad by opposing the inhibitory effect of growth factor on Bad-mediated apoptosis. Cdc2-induced phosphorylation of Bad serine 128 has an inhibitory effect on its interaction with 14-3-3 proteins. The latter interaction is critical for Bad phosphorylation at serine 155, a site within the BH3 domain that leads to the release of BclxL and the promotion of cell survival. Alternative splicing of this gene results in two transcript variants which encode the same isoform.

Description:   The calcium-activated chloride channel (CLCA) protein family, which includes the human homologs CLCA1 and CLCA2, display distinct tissue distribution patterns. CLCA1 is expressed as a precursor protein that is processed into two cell surface associated subunits and a group of proteins. CLCA1 is upregulated by interleukin-9 and regulates the expression of mucins. CLCA1 may provide a therapeutic target to control mucus overproduction in airway disease patients with cystic fibrosis. CLCA2 expression is downregulated in breast cancer, therefore CLCA2 is thought to act as a tumor suppressor in normal cells. CLCA3 (known as Clca3 in mouse) is a structurally divergent member of the CLCA family that does not function as a channel protein. CLCA4 is a CLCA member that is expressed in human rectal mucosa, CLCA5 shows strong expression in eye and spleen, and CLCA6 is primarily expressed in intestine and stomach.
