ILVBL is a 632 amino acid single-pass membrane protein that belongs to the TPP enzyme family. Expressed in the majority of tissues, ILVBL has the highest level of expression in heart, pancreas and placenta. ILVBL is highly homologous to several bacterial enzymes, including the B isozyme of the large catalytic subunit of E. coli acetohydroxy-acid synthase (AHAS) and the oxalyl-coA decarboxylase of O. formigenes, that utilize thiamine pyrophosphate as a cofactor. ILVBL binds one magnesium ion and one thiamine pyrophosphate per subunit, and may catalyze the initial step in branched-chain amino acid biosynthesis. The gene encoding ILVBL maps to human chromosome 19p13.12 and mouse chromosome 10 C1.

Actin filament associated protein (AFAP-110) interacts directly with actin filaments through its C-terminal actin-binding domain. AFAP-110 contains additional protein-binding domains as well, and serves as an adaptor protein. By linking signaling molecules to actin filaments, AFAP-110 provides a platform for the preparation of larger signaling complexes, activates Src kinases in response to cellular signals and also directly affects Actin organization as an Actin filament cross-linking protein. AFAP-1L2 (Actin filament-associated protein 1-like 2), also known as XB130, is a 818 amino acid cytoplasmic protein that contains two Pleckstrin homology (PH) domains, which are normally found in proteins involved in intracellular signaling. Like its relative AFAP110, AFAP-1L2 interacts with Src kinase and may play a role in Src-regulated transcription activation. AFAP-1L2 is expressed in thyroid and spleen and can also be detected at lower levels in lung, brain, pancreas and kidney. There are four isoforms of AFAP-1L2 that are produced as a result of alternative splicing events.

Glypican-1 (GPC1), glypican-2 (GPC2) and glypican-3 (GPC3) are members of the glypican family of heparan sulfate proteoglycans, which attach to the cell membrane via a glycosylphosphatidylinositol (GPI) anchor. Cell-surface heparan sulfate proteoglycans participate in molecular events that regulate cell adhesion, migration, and proliferation. Glypican-2, a cell surface proteoglycan bearing heparan sulfate, may have a function related to the motile behaviors of developing neurons. Ligation of cell-surface glypican-2 with midkine (MK) or an; against epitope-tagged glypican-2 induces cell adhesion and promotes neurite outgrowth. MK binds to heparan sulfate chains of glypican-2 in a manner similar to syndecan-3, but different localization of epitope-tagged glypican-2 and syndecan-3 on the surface of N2?cells suggests that they may play different roles in MK-mediated neural function.

The immunophilins are a highly conserved family of cis-trans peptidyl-prolyl isomerases that bind to and mediate the effects of immunosuppressive drugs, such as cyclosporin, FK506 and rapamycin. Immunophilins have also been implicated in protein folding and trafficking within the endoplasmic reticulum (ER). FKBP11 (FK506-binding protein 11), also known as FKBP19 or peptidyl-prolyl cis-trans isomerase FKBP11, is a 201 amino acid single-pass membrane protein belonging to the FKBP-type PPIase family, a group of proteins known to catalyze the folding of proline-containing polypeptides. Containing one PPIase FKBP-type domain, FKBP11 is expressed in secretory tissues such as pancreas, pituitary, stomach, lymph node and salivary gland, and is encoded by a gene that maps to human chromosome 12q13.12. FK506 and rapamycin are known to inhibit FKBP11’s peptidyl-prolyl isomerase activity.
