Description:
MP1 (MEK partner 1) functions as a scaffolding protein in the mitogen activated protein (MAP) kinase signaling pathway. Growth factor induced MAP kinase activation is selectively mediated by the extracellular signal-regulated kinase (ERK) cascade. MAPBPIP (mitogen-activated protein-binding protein-interacting protein), also known as p14 and late endosomal/lysosomal MP1-interacting protein, functions as an adaptor protein augmenting the regulation of the MAP kinase cascade. Partner proteins MAPBPIP and MP1 are structurally almost identical each with a five-stranded -sheet flanked between a two-helix and one-helix layer. MAPBPIP compels the recruitment of MP1 to late endosomes where they form a very stable heterodimeric complex required for ERK activation on endosomes. Knockdown of the individual proteins in the MP1/MAPBPIP complex resulted in decreased expression of the partner proteins which implies greater stability of the heterodimeric complex than either MP1 or MAPBPIP individually. Early research suggests the MP1-MAPBPIP-MEK-1 signaling complex may be critical in the regulation of tissue homeostasis.

Encoded by a gene that maps to human chromosome 5q23.2, C5orf48 is linked to Autosomal dominant leukodystrophy (ADLD). Chromosome 5 makes up approximately 6% of the human genome and contains 181 million base pairs, which encode 1,000 genes. Chromosome 5 is associated with Cockayne syndrome through the ERCC8 gene and familial adenomatous polyposis through the adenomatous polyposis coli (APC) tumor suppressor gene. Treacher Collins syndrome is caused by insertions or deletions within the TCOF1 gene and is also associated with chromosome 5. Deletion of 5q or chromosome 5 altogether is common in therapy-related acute myelogenous leukemias and myelodysplastic syndrome.

There are three protocadherin gene clusters, designated a, b and g, all of which contain multiple tandemly-arranged genes. PCDHB10 (protocadherin b10), also known as PCHB10 or PCDH-b10, is an 800 amino acid protein that is one of 16 proteins in the protocadherin b cluster. Unlike the a and g gene clusters, whose genes are spliced to downstream constant-region exons during transcription, members of the b cluster (such as PCDHB10) do not use constant-region exons to produce mRNAs. As a result, each protocadherin b gene encodes the transmembrane, extracellular and short cytoplasmic domains of the protein. Glycogen phosphorylase (also known as GP) is an allosteric enzyme important in carbohydrate metabolism. Its activity is regulated through either noncovalent binding of metabolites or by covalent modification. Glycogen phosphorylase catalyzes the phosphorylation of glycogen to Glc-1-P. There are three genes which encode the brain, liver and muscle forms of glycogen phosphorylase, PYGB, PYGL and PYGM. Because of its fundamental role in the metabolism of glycogen, glycogen phosphorylase has been a target for the design of inhibitory compounds, which could be valuable in the therapeutic treatment of type 2 diabetes mellitus.
