Description:   Serine/threonine kinase which plays an important role in the regulation of electrolyte homeostasis, cell signaling, survival and proliferation. Acts as an activator and inhibitor of sodium-coupled chloride cotransporters and potassium-coupled chloride cotransporters respectively. Phosphorylates WNK4. Regulates the phosphorylation of SLC12A1 and SLC12A2. Increases Ca(2+) influx mediated by TRPV5 and TRPV6 by enhancing their membrane expression level via a kinase-dependent pathway. Inhibits the activity of KCNJ1 by decreasing its expression at the cell membrane in a non-catalytic manner. Isoform 1, isoform 2, isoform 3 and isoform 4 stimulate the activity of SLC12A1, SLC12A2 and SLC12A3 and inhibit the activity of SLC12A4, SLC12A5, SLC12A6 and SLC12A7. According to PubMed:19470686, isoform 1 inhibits the activity of SLC12A3.

Description:   MTA1 is a component of the NURD (nucleosome remodeling and histone deacetylation) complex, which is associated with ATP-dependent chromatin-remodeling and histone deacetylase activity. MTA1 functions in conjunction with other components of NURD to mediate transcriptional repression as it facilitates the association of repressor molecules with the chromatin. Structurally, MTA1 contains a single SH3-binding motif and a zinc finger domain, along with a region similar to the co-repressor protein N-Cor. MTA1 is normally expressed at low levels in various tissues and is more highly expressed in testis. Overexpression of MTA1 correlates with tumour invasion and metastasis in various carcinomas including colorectal, gastrointestinal and breast carcinomas. Elevation of MTA1 levels in these tumours appears to enhance the metastases to lymph nodes, increase mammary cell motility and potentiate growth, and therefore may be an indicator for assessing the potential malignancies of various tumours. A similar protein, MTA2, also designated MTA1-L1 (MTA1-like protein 1), shares more than 55% sequence homology with MTA1 and is ubiquitously expressed.

Description:   The Alpha-, Beta-, Gamma-, and Delta -catenins are proteins that bind to the highly conserved, intracellular cytoplasmic tail of E-cadherin. Together, the catenin/cadherin complexes play an important role mediating cellular adhesion. Alpha-catenin interacts with E-cadherin associated protein and also associates with other members of the cadherin family, such as N-cadherin and P-cadherin. Beta-catenin associates with the cytoplasmic portion of E-cadherin, which is necessary for the function of E-cadherin as an adhesion molecule. Beta-catenin also complexes with the tumor suppressor protein APC. Gamma-catenin, also known as plakoglobin, is a protein that binds ?catenin and N-cadherin. Gamma-catenin interacts with presenilin 1 and is expressed in the brain. The gene encoding Gamma-catenin maps to human chromosome 5p15.2. A hemizygous loss of the gene encoding Gamma-catenin leads to the mental retardation associated with Cri-du-Chat syndrome. In addition, the transmembrane phosphatase PTPm associates with catenin/ cadherin complexes and may regulate complex signaling.

Description:   NMDA receptor subtype of glutamate-gated ion channels with high calcium permeability and voltage-dependent sensitivity to magnesium. Mediated by glycine. In concert with DAPK1 at extrasynaptic sites, acts as a central mediator for stroke damage. Its phosphorylation at Ser-1303 by DAPK1 enhances synaptic NMDA receptor channel activity inducing injurious Ca2+ influx through them, resulting in an irreversible neuronal death (By similarity).

Description:   CBLL1, also known as HAKAI (meaning ‘destruction’ in Japanese), or RNF188 (RING finger protein 188), is a 491 amino acid protein that contains one C2H2-type zinc finger and one RING-type zinc finger. CBLL1 is believed to function as an E3 ubiquitin-protein ligase that accepts a ubiquitin residue from an E2 ubiquitin-conjugating enzyme and immediately transfers that residue to a protein that is targeted for degradation. More specifically, upon activation of c-Src, CBLL1 interacts with and ubiquitinates tyrosine-phosphorylated E-cadherin, thereby targeting the E-cadherin complex for endocytosis and disrupting epithelial cell-cell contacts. Via its role as an E-cadherin regulator, CBLL1 participates in cell adhesion and may also be involved in the regulation of epithelial-mesenchymal transitions.

Description:   Gross cystic disease is a common premenopausal disorder in which gross cysts are the predominant pathologic lesion. It is characterized by production of a fluid secretion which accumulates in the breast cysts. Gross cystic disease fluid is a pathologic secretion from breast composed of several glycoproteins, including a unique 15 kDa monomer protein, GCDFP 15. The cells within the body that produce GCDFP 15 appear to be restricted primarily to those with apocrine function. Studies have found GCDFP 15 to be a highly specific and sensitive marker for breast cancer.
