Belonging to the glycosyltransferase 14 family, GCNT7, also known as Beta-1,3-galactosyl-O-glycosyl-glycoprotein beta-1,6-N-acetylglucosaminyltransferase 7, is a 430 amino acid glycosyltransferase that is localized to the Golgi apparatus. Other members of this family include GCNT1, GCNT2, GCNT3, GCNT4 and GCNT6. GCNT1 has been shown to play an important regulatory role in the biosynthesis of mucin-type O-glycans, which serve as ligands in cell adhesion. Specifically, GCNT1 expression in leukocytes regulates the synthesis of core 2 O-glycans that carry sialyl-Lewis x (sLex) oligosaccharides, which confer high affinity binding to Selectin proteins. Since downregulation of Selectin ligand expression has been shown to inhibit tissue infiltration, glycosyltransferase 14 family members represent potential drug targets for the treatment of inflammatory disorders and other pathologies involving Selectin proteins.

Contactin 2 is a neuronal cell adhesion molecule (CAM) that influences the formation of axon connections in the developing nervous system. Contactin 2 is a member of the immunoglobulin superfamily (IgSF) and contains a glycosylphosphatidylinositol-anchor, six immunogobulin (Ig)-like and four Fibronectin type III (FNIII)-like domains. Contactin 2 is expressed predominantly during neural development on the cell membrane of axons in nerve fiber tracts in order to guide commissural axons without promoting their growth. Contactin 2 binds with NgCAM in the plane of the same membrane (cis-binding). The Contactin 2 heterophilic (Contactin 2/NgCAM and Contactin 2/NrCAM) binding sites are localized to the first four Ig domains. The Contactin 2 homophilic (Contactin 2/Contactin 2) binding site is localized to the FNIII domain.

Fibrinogen-like 1 is a member of the fibrinogen family. This protein is homologous to the carboxy terminus of the fibrinogen beta- and gamma- subunits which contains the four conserved cysteines of fibrinogens and fibrinogen related proteins. Overexpression of the gene may confer a growth advantage to cancer cells and favor tumor cell proliferation. E6-associating protein is a component of the ubiquitin-mediated proteolytic pathway, which selectively targets proteins for degradation by the 26S proteasome. Ubiquitin (Ub) is directly conjugated to protein substrates by the transfer of Ub from an E2 ubiquitin conjugating enzyme to the target protein. This conjugation is facilitated by the enzymatic activity of E3 ubiquitin ligase family members such as E6-AP. Several substrates of E6-AP have been identified and include the tumour suppressor protein p53 and the mammalian homolog of Rad23, HHR23A. Previous studies have indicated that E6-AP associates with the human papillomavirus E6 oncogene, which complexes with p53 and thereby potentiates E6-AP mediated ubiquitination of p53. Genetic mutations that impair E6-AP activity result in the accumulation of p53 in the cytoplasm, and, in many instances, these mutations are associated with the development of the rare neurodevelopmental disorder Angelman syndrome (AS), which is characterised by severe motor dysfunction and mental retardation.

CatSpers (cation channel, sperm associated proteins) are ion transport proteins located on the surface of sperm cells in the principal piece of the sperm tail. CatSpers are vital to sperm motility, fertilization and cAMP-mediated calcium influx in sperm. There are four CatSper proteins in mammalian sperm, namely CatSper (or CatSper1), CatSper2, CatSper3 and CatSper4. CatSper proteins contain a single, six-transmembrane-spanning segment and exhibit the voltage-dependent Ca2+ channel four-repeat structure. CatSper proteins are believed to assemble into a heterotetrameric complex, forming an alkalinization-activated Ca2+-selective channel. Mutations in any of the genes encoding CatSper family proteins can result in male infertility. CatSper3 plays an important role in the hyperactivated motility of sperm cells, a process that is required in the preparation of sperm for fertilization.
