Three mammalian fringe family members, Manic, Radical and Lunatic Fringe, have been identified as proteins related to Drosophila fringe, a protein involved in development. Fringe proteins act upstream of the Notch signaling pathway and are involved in boundary determination during segmentation. Each mammalian fringe displays different patterns of expression, though all are expressed in the mouse embryo as well as in many adult tissues. Radical fringe plays a key role in the development of the limb bud. Lunatic fringe is required for normal somite segmentation and patterning and is thought to be a target of the molecular clock. Manic fringe, also involved in somatic development, has been shown to render mouse NIH/3T3 cells tumorigenic in SCID mice.

C1orf187, also known as Draxin, Dorsal repulsive axon guidance protein and Neucrin, is a 349 amino acid secreted protein that is required of the development of the spinal cord and forebrain commissures. C1orf187 acts as a chemorepulsive guidance protein and directs commissural axons during development by repelling neurite outgrowth from the spinal cord. During development, C1orf187 modulates neural crest migration by reducing the polarization of these cells, leading to reduced velocity of migration and increased frequency of changing direction, leading to a net decrease in migrational distance. It acts as an antagonist of the Wnt signaling pathway by inhibiting the stabilization of cytosolic _-catenin via its interaction with LRP6. C1orf187 inhibits outgrowth from the olfactory bulb and likely contributes to the formation of the lateral olfactory tract.

The FOX family of transcription factors share a common DIUA binding domain termed a winged-helix or forkhead domain. Many FOX proteins play important roles in development, metabolism, cancer and aging. Development of the vertebrate gut is controlled by paracrine crosstalk between the endodermal epithelium and the associated splanchnic mesoderm. FOXF2 (forkhead box F2), also known as FKHL or FREAC2, is expressed in lung and placenta, and has been shown to transcriptionally activate several lung-specific genes. FOXF2 interacts with transcription factors TFIIB and TBP, and may be involved in regulating transcription in embryogenesis and pattern formation in multicellular organisms. FOXF2-deficient mice develop cleft palate and an abnormal tongue, which suggests that FOXF2 may be critical for palatogenesis.
