Description:
Bcl-6, a transcriptional repressor, binds Stat recognition-like DNA elements and influences germinal center development and cell differentiation. Additionally, Bcl-6 negatively regulates NFâ…¹B expression, thereby inhibiting NFâ…¹B-mediated cellular functions. Bcl-6b (B-cell CLL/lymphoma 6, member B), also known as ZNF62, BAZF or ZBTB28, is a 480 amino acid nuclear protein that contains one BTB (POZ) domain and five C2H2-type zinc fingers. Expressed ubiquitously with highest expression in placenta and heart, Bcl-6b associates with Bcl-6 and functions as a sequence-specific transcriptional repressor that is thought to be necessary for early B-cell development. The gene encoding Bcl-6b maps to human chromosome 17, which comprises over 2.5% of the human genome and encodes over 1,200 genes.

Description:
GARNL1 is expressed during embryogenesis with E12. During development, GARNL1 expression decreases, persisting at high levels only in neurons of the adult brain. GARNL1 localizes to the cytoplasm where it may play a role regulating GTP hydrolysis of proteins such as Ran and Rap. GARNL1 is imported to the nucleus via dimerization with E12. GARNL1 interacts with the HLH region of E12 and may function to negatively regulate the transcription of E12-dependent downstream target genes. This suggests that at least a portion of the function of GARNL1 is dependent upon its association with E12. GARNL1 may also associate with other HLH proteins and influence a variety of HLH signaling cascades. In adult brain, GARNL1 activity does not involve E12 and therefore it may serve a different function in developed neural tissue.

Description:
The X and Y chromosomes are the human sex chromosomes. Chromosome X consists of about 153 million base pairs and nearly 1000 genes. The combination of an X and Y chromosome lead to normal male development while two copies of X lead to normal female development. There are a number of conditions related to an unusual number and combination of sex chromosomes being inherited. More than one copy of the X chromosome with a Y chromosome causes Klinefelter's syndrome. A single copy of X alone leads to Turner's syndrome. More than 2 copies of the X chromosome, in the absence of a Y chromosome, is known as Triple X syndrome. Color blindness, hemophilia, and Duchenne muscular dystrophy are well known X chromosome-linked conditions which affect males more frequently as males carry a single X chromosome. The FAM70A gene product has been provisionally designated FAM70A pending further characterisation.

Description:
Growth hormone-releasing hormone (GHRH) stimulates secretion and synthesis of growth hormone (GH), causes somatotroph proliferation and may have direct actions in fetal/placental development, reproduction and immune function. It exerts its action through high-affinity GHRH receptors present in the anterior pituitary. GSH-1 (GS homeobox 1) is a 264 amino acid hypothalamic nuclear protein that functions as a transcription factor responsible for maintaining GHRH expression as well as playing an important role in pituitary development. Coexpression of CBP leads to significantly enhanced GSH-1-induced GHRH expression, which suggest that CBP may function as a co-activator. Knockdown of GSH-1 mRNA in mice causes a dwarf phenotype, which suggests that certain cases of familial dwarfism may be caused by a mutation of the GSH-1 gene.

Description:
Tyrosine-protein kinase that acts as a cell-surface receptor for VEGFC and VEGFD, and plays an essential role in adult lymphangiogenesis and in the development of the vascular network and the cardiovascular system during embryonic development. Promotes proliferation, survival and migration of endothelial cells, and regulates angiogenic sprouting. Signaling by activated FLT4 leads to enhanced production of VEGFC, and to a lesser degree VEGFA, thereby creating a positive feedback loop that enhances FLT4 signaling. Modulates KDR signaling by forming heterodimers. Mediates activation of the MAPK1/ERK2, MAPK3/ERK1 signaling pathway, of MAPK8 and the JUN signaling pathway, and of the AKT1 signaling pathway. Phosphorylates SHC1. Mediates phosphorylation of PIK3R1, the regulatory subunit of phosphatidylinositol 3-kinase. Promotes phosphorylation of MAPK8 at 'Thr-183' and 'Tyr-185', and of AKT1 at 'Ser-473'.

Description:
Receptor tyrosine kinase which binds promiscuously membrane-bound ephrin family ligands residing on adjacent cells, leading to contact-dependent bidirectional signaling into neighboring cells. The signaling pathway downstream of the receptor is referred to as forward signaling while the signaling pathway downstream of the ephrin ligand is referred to as reverse signaling. Highly promiscuous for ephrin-A ligands it binds preferentially EFNA5. Upon activation by EFNA5 regulates cell-cell adhesion, cytoskeletal organization and cell migration. Plays a role in cardiac cells migration and differentiation and regulates the formation of the atrioventricular canal and septum during development probably through activation by EFNA1. Involved in the retinotectal mapping of neurons. May also control the segregation but not the guidance of motor and sensory axons during neuromuscular circuit development.

Description:
Contactin 2 is a neuronal cell adhesion molecule (CAM) that influences the formation of axon connections in the developing nervous system. Contactin 2 is a member of the immunoglobulin superfamily (IgSF) and contains a glycosylphosphatidylinositol-anchor, six immunogobulin (Ig)-like and four Fibronectin type III (FNIII)-like domains. Contactin 2 is expressed predominantly during neural development on the cell membrane of axons in nerve fiber tracts in order to guide commissural axons without promoting their growth. Contactin 2 binds with NgCAM in the plane of the same membrane (cis-binding). The Contactin 2 heterophilic (Contactin 2/NgCAM and Contactin 2/NrCAM) binding sites are localized to the first four Ig domains. The Contactin 2 homophilic (Contactin 2/Contactin 2) binding site is localized to the FNIII domain.

Description:
Functions as a ubiquitin ligase protein in vivo, mediating ubiquitination and promoting degradation of MEKK1, suggesting that it may regulate the Notch pathway via some ubiquitin ligase activity (By similarity). Regulator of Notch signaling, a signaling pathway involved in cell-cell communications that regulates a broad spectrum of cell-fate determinations. Mainly acts as a positive regulator of Notch, but it also acts as a negative regulator, depending on the developmental and cell context. Mediates the antineural activity of Notch, possibly by inhibiting the transcriptional activation mediated by MATCH1. Involved in neurogenesis, lymphogenesis and myogenesis, and may also be involved in MZB (Marginal zone B) cell differentiation. Promotes B-cell development at the expense of T-cell development, suggesting that it can antagonize NOTCH1.
