SNAP 29 is a 258 amino acid protein that localizes to the membrane and the cytoplasm, as well as to the cell junction, and contains one t-SNARE coiled-coil homology domain. Expressed in liver, heart, brain, kidney, placenta, lung, spleen, pancreas and skeletal muscle, SNAP 29 binds tightly to Syntaxins and, via this binding, is involved in membrane trafficking events. Defects in the gene encoding SNAP 29 are the cause of CEDNIK syndrome, a neurocutaneous syndrome that is associated with cerebral dysgenesis, neuropathy, ichthyosis and palmoplantar keratoderma. The gene encoding SNAP 29 maps to human chromosome 22, which houses over 500 genes and is the second smallest human chromosome. Mutations in several of the genes that map to chromosome 22 are involved in the development of Phelan-McDermid syndrome, Neurofibromatosis type 2, autism and schizophrenia.

MAPK10 (JNK3) is a member of the MAP kinase family. MAP kinases act as an integration point for multiple biochemical signals, and are involved in a wide variety of cellular processes such as proliferation, differentiation, transcription regulation and development. This protein is a neuron-specific form of c-Jun N-terminal kinases (JNKs). Through its phosphorylation and nuclear localization, this kinase plays regulatory roles in the signaling pathways of neuronal apoptosis. Beta-arrestin 2, a receptor-regulated MAP kinase scaffold protein, is found to interact with and stimulate the phosphorylation of this kinase by MAP kinase kinase 4 (MKK4). Cyclin-dependent kianse 5 (CDK5) can phosphorylate and inhibit the activity of this kinase, which may be important in preventing neuronal apoptosis. Four alternatively spliced transcript variants encoding distinct isoforms have been reported.

EphB1, previously known as Elk (eph like kinase), is a receptor tyrosine kinase of the highly tissue restricted family of eph proteins. EphB1 and other ephB family members are type 1 membrane spanning proteins, comprised of immunoglobulin, fibronectin type III, and cysteine rich subdomains in the ecto domain, and the single uninterrupted cytoplasmic tyrosine kinase domain upstream of a carboxyterminal sterile alpha motif (SAM) domain. EphB family proteins bind ephrins of the B class. EphB1 is expressed predominately in developing neural structures in embryos, and in vascular epithelium of kidney, and other tissues. Upon binding to alternatively oligomerized ephrin B1, EphB1 signals regulation of cell attachment and cell to cell assembly. Members of this protein family are implicated in neuronal and vascular cell targeting.

Notably, the PARK2 gene, which is associated with Parkinson's disease, and the genes encoding the major histocompatiblity complex proteins, which are key molecular components of the immune system and determine predisposition to rheumatic diseases, are also located on chromosome 6. Stickler syndrome, 21-hydroxylase deficiency and maple syrup urine disease are also associated with genes on chromosome 6. A bipolar disorder susceptibility locus has been identified on the q arm of chromosome 6.

FRAT1 and FRAT2 were originally characterized as proteins frequently rearranged in advanced T cell lymphoma, and they have since been identified as proto-oncogenes involved in tumorigenesis. These proteins share significant homology with the Xenopus glycogen synthase kinase-3 (xGSK-3) binding protein, which is designated GBP and is essential for the formation of the dorsal-ventral axis during embryonic development. Establishment of these embryonic axes is mediated by the Wnt intracellular signaling pathway. Wnt signaling is regulated in part by the activity of GSK-3, which phosphorylates and thereby facilitates the degradation of ?catenin. GBP binds to GSK-3 and inhibits this phosphorylation, resulting in the accumulation of ?catenin and the subsequent transcription of Wnt target genes. DNA fragmentation factor (DFF) is a heterodimeric protein of 40-kD (DFFB) and 45-kD (DFFA) subunits. DFFA is the substrate for caspase-3 and triggers DNA fragmentation during apoptosis. DFF becomes activated when DFFA is cleaved by caspase-3. The cleaved fragments of DFFA dissociate from DFFB, the active component of DFF. DFFB has been found to trigger both DNA fragmentation and chromatin condensation during apoptosis. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene but the biological validity of these variants has not been determined.

CDK5 (Cyclin Dependent Kinase 5) is serine/threonine kinase involved in synaptic regulation and neuronal development; phosphorylates synaptic protein Pctaire1; regulates acetylcholine receptor expression. CDK5 is a member of the cyclindependent kinase family of serine/threonine kinases. It is present in numerous mammalian tissues including kidney, testes, and ovary. Its activity is detected almost exclusively in brain extracts. Neuronal and muscle cells contain the highest amount of this protein. Similar to other Cdks, monomeric Cdk5 displays no enzymatic activity, but Cdk5 is not activated by cyclins. Instead, the p35 protein, which is expressed solely in the brain, activates Cdk5. Cdk5 interacts with D1 and D3 type G1 cyclins and can phosphorylate histone H1, TAU, MAP2 and NF-H and NF-M. Cdk5 activity is involved in terminal differentiation of neurons and muscle cells.
