Early B-cell factor 2 is a 575 amino acid protein belonging to the COE family of proteins, whose members are all helix-loop-helix transcription factors. EBF2 is a transcription factor which, in synergy with the Wnt-responsive LEF1/CTNNB1 pathway, activates the decoy receptor for RANKL, OPG, in osteoblasts. OPG, in turn, regulates osteoclast differentiation. Lack of EBF2 has been found to cause a small defect in the terminal differentiation of osteoblasts, along with reduced bone mass and an increase in osteoclasts. Localized to the nucleus, EBF2 forms a homodimer or a heterodimer with a related family member.

Description:
Matrix Metalloproteinase 28, also known as epilysin, was first cloned from human skin keratinocytes and described to reflect its role in the remodeling of the epidermis. It was found in testis, as well as heart, brain, placenta, lung, prostate, intestine, and colon. MMP28 was later cloned from human lung and found in lung, kidney, brain, skeletal muscle, and several tumor cell lines. At least three MMP28 transcripts of 2.6, 2.0, and 1.2 kb have been reported possibly representing alternatively spliced forms of MMP28. There are two human sequences (isoform1 and isoform 2) which encode proteins of 520 and 393 amino acids with predicted masses of 58.9 and 44.5 kDa respectively. The mouse sequence contains two inserts of 41 and 39 amino acids respectively, relative to the human sequence. Mouse epilysin has a predicted molecular mass of approx. 70 kDa. Mouse and human epilysin are highly conserved and share 97% identical residues. Epilysin (MMP-28) contains the key domains of the other MMPs: a signal peptide, conserved cysteine-containing prodomain (with a furin cleavage site), conserved histidine-containing catalytic domain, hinge domain, and hemopexin domain. MMP28 has a furin cleavage site, similar to MMP11, and is cleaved by the prohormone convertase family of enzymes. Sequence identity between MMP28 and the other MMPs is low overall. MMP28 is most closely related to MMP19 (39% sequence identity). Recombinant MMP28 degrades casein.

GM130, a cis-Golgi matrix protein, interacts specifically with p115 and provides a membrane docking site. Both GM130 and p115 are involved in vesicle tethering to Golgi membranes. The protein p115 also binds p400, alternatively called giantin. Giantin, the majority of whose mass projects into the cytoplasm, is involved in the docking of COPI vesicles via p115 to the Golgi membrane. Giantin, which also is known as macrogolgin or Golgi complex-associated protein, is involved in cross-bridge formation in the Golgi complex. Giantin, which can form a homodimer, is a single-pass type I membrane protein that is an antigen in Sjoegren syndrome and in chronic rheumatoid arthritis.

The full-length cDNA encodes a deduced 414-amino acid protein with a molecular mass of 46.56 kD. DRAKs contain an N-terminal kinase domain and a C-terminal regulation domain. DRAK1 messenger RNA appears to be ubiquitously expressed in human tissues. Overexpression of DRAK1 induces apoptosis. It has been shown in vitro that DRAK1 is capable of autophosphorylation and of phosphorylating the myosin light chain as an exogenous substrate, and that the noncatalytic C terminus is crucial for full kinase activity.
