Description:   There are three SorCS genes that have diverse, partially overlapping functions in the central nervous system. In the developing and mature central nervous system, SorCS1, SorCS2 and SorCS3 genes are expressed in a combinatorial, non-overlapping pattern. SorCS proteins show homology to the mosaic receptor SorLA and the neurotensin receptor sortilin, based on a common VPS10 domain, which is the hallmark of the SorCS receptor family. SorCS2 (sortilin-related VPS10 domain containing receptor 2) is a 1,150 amino acid single-pass type I membrane protein that is highly expressed in brain and kidney. Containing six BNR repeats and a single PKD domain, SorCS2 is encoded by a gene that maps to human chromosome 4, which encodes nearly 6% of the human genome and has the largest gene deserts (regions of the genome with no protein encoding genes) of all of the human chromosomes.

Description:   The glycine cleavage system is comprised of AMT (known as Protein T), GCSH (known as Protein H), DLD (known as Protein L) and GLDC (known as Protein P), all of which work together to catalyze the cleavage and degradation of glycine. GLDC (glycine dehydrogenase ), also known as GCE, GCSP (glycine cleavage system P protein) or HYGN1, is a 1,020 amino acid protein that localizes to the mitochondria and belongs to the gcvP family. GLDC binds to glycine and enables the methylamine group from glycine to be transferred to the Protein T. GLDC exists as a homodimer and utilizes pyridoxal phosphate as a cofactor. Mutations in the gene encoding GLDC leads to nonketotic hyperglycinemia (NKH), also known as glycine encephalopathy (GCE), an autosomal recessive disease characterized by accumulation of a large amount of glycine in body fluid and by severe neurological symptoms.

Description:   Cysteine-rich secretory proteins (CRISPs) represent a family of evolutionarily conserved proteins that may play a role in the innate immune system and are transcriptionally regulated by androgens in several tissues. GAPR-1 (Golgi-associated plant pathogenesis-related protein 1), also known as GLIPR2, is a 154 amino acid lipid anchor protein belonging to the CRISP family. GAPR-1 also shares similarity with the pathogenesis-related protein (PR) superfamily, and may play an important role in the immune system. Existing as a homodimer, GAPR-1 is highly expressed in lung and peripheral leukocytes with minor expression in liver and kidney. Containing a conserved sperm-coating protein (SCP) domain, GAPR-1 binds to negatively charged lipids and may be involved in the differentiation of epithelial cells into mesenchymal cells. Increased expression of GAPR-1 in kidney may contribute to the development of fibrosis.
