Non-catalytic subunit of AMP-activated protein kinase (AMPK), an energy sensor protein kinase that plays a key role in regulating cellular energy metabolism. In response to reduction of intracellular ATP levels, AMPK activates energy-producing pathways and inhibits energy-consuming processes: inhibits protein, carbohydrate and lipid biosynthesis, as well as cell growth and proliferation. AMPK acts via direct phosphorylation of metabolic enzymes, and by longer-term effects via phosphorylation of transcription regulators. Also acts as a regulator of cellular polarity by remodelling the actin cytoskeleton; probably by indirectly activating myosin. Beta non-catalytic subunit acts as a scaffold on which the AMPK complex assembles, via its C-terminus that bridges alpha (PRKAA1 or PRKAA2) and gamma subunits (PRKAG1, PRKAG2 or PRKAG3).

DnaJ-like proteins interact with HSP 70 molecular chaperones and function to facilitate protein folding and mitochondrial protein import. HSP 40-4, also known as HDJ2, is the human DnaJ homolog that functions as a co-chaperone with a cysteine-rich zinc finger domain. The cellular redox enzyme thioredoxin interacts with HSP 40-4, and oxidation and reduction reversibly regulate HSP 40-4 function in response to the changing redox states of the cell. The zinc finger domain of HSP 40-4 may act as a redox sensor of chaperone-mediated protein-folding machinery, since HSP 40-4 inactivation leads to the oxidation of cysteine thiols and a simultaneous release of coordinated zinc. Loss of the HSP 40-4 protein may be linked to severe defects in spermatogenesis that involve aberrant androgen signaling.

NEEP21, also known as Brain neuron cytoplasmic protein 1, NSG1 (neuron-specific proteins family member 1), P21 or D4S234E, is a single pass type II membrane protein belonging to the NSG family. It is highly expressed during neuronal maturation but its expression is downregulated in adult tissues. NEEP21 predominantly localizes to Rab 4-positive early endosomes in the somatodendritic neuronal compartment and is essential for proper receptor sorting and recycling in neurons. It associates with GRIP1 and GluR-2 and mediates the surface expression of GluR-2. When this interaction is interrupted, GluR-2 accumulates in early endosomes and leads to changes in evoked synaptic current properties. In addition, NEEP21 forms a complex with the SNARE protein, Syntaxin 13 (also known as Syntaxin 12), and participates in the recycling of transferrin receptors (TFRs) and NTR2 (neurotensin receptor 2).

Utilising magnesium, manganese or nickel as cofactors, HEAB participates in the phosphorylation of the 5'-hydroxyl groups of double- and single- stranded RNA and DNA. HEAB is a member of the tRNA splicing endonuclease complex, in conjunction with TSEN2, TSEN15, TSEN34 and TSEN54, and is also a member of the pre-mRNA cleavage complex II. The gene encoding HEAB maps to human chromosome 11q12.1 and mouse chromosome 2 D; mutations to this gene may lead to a reduced pre-mRNA cleavage activity. HEAB exists as two isoforms due to alternative splicing events.

The functional form of BMP2 is a 26 kDa protein composed of two identical 114 amino acid polypeptide chains linked by a single disulfide bond. Recognizes a single glycoprotein of 520 kDa, identified as mucin 2 (MUC2). This MAb shows no cross-reaction with human milk fat globule membranes, MUC1, or MUC3. Its epitope has been defined as GTQTP (GlyThrGlnThrPro). Mucins are high molecular weight glycoproteins, which constitute the major component of the mucus layer that protects the gastric epithelium. MUC2 is specifically expressed in goblet cells of the small intestine & colon; in about 65% of colonic carcinomas and about 40% of gastric carcinomas. MUC2 is rarely expressed outside of the GI tract with the exceptions of mucinous carcinoma of breast and clear cell-type carcinomas of the ovary.

Human chromosome 4 represents approximately 6% of the human genome and contains nearly 900 genes. Notably, the Huntingtin gene, which is found to encode an expanded glutamine tract in cases of Huntington's disease, is on chromosome 4. FGFR-3 is also encoded by a gene that is located on chromosome 4, and has been associated with thanatophoric dwarfism, achondroplasia, Muenke syndrome and bladder cancer.
