GLT8D1 is a 371 amino acid single-pass type II transmembrane protein that is expressed by a gene residing on human chromosome 3. Chromosome 3 is made up of about 214 million bases encoding over 1,100 genes, including a chemokine receptor (CKR) gene cluster and a variety of human cancer-related gene loci. Key tumor suppressing genes on chromosome 3 include those that encode the apoptosis mediator RASSF1, the cell migration regulator HYAL1 and the angiogenesis suppressor SEMA3B. Marfan Syndrome, porphyria, von Hippel-Lindau syndrome, osteogenesis imperfecta and Charcot-Marie-Tooth Disease are a few of the numerous genetic diseases associated with chromosome 3. There are two isoforms of GLT8D1 that are produced as a result of alternative splicing events.

Shootin1 is a 631 amino acid protein that belongs to the shootin family. The shootin1 protein contains three coiled-coil domains, a proline-rich region and interacts with RUFY3. Shootin1 is involved in the generation of internal asymmetric signals required for neuronal polarization. The shootin1 protein acts upstream of PI3K (phosphoinositide 3-kinase), by being required for spatially localized PI3K activity. By accumulating asymmetrically in a single neurite before polarization, shootin1 leads to axon induction for polarization, additionally the absence of shootin1 from the nascent axon's siblings by competition prevents the formation of surplus axons. Existing as seven alternatively spliced isoforms, the shootin1 gene is conserved in chimpanzee, dog, mouse, rat, chicken and zebrafish, and maps to human chromosome 10q25.3.

Hephaestin is a single-pass type I membrane protein that belongs to the multicopper oxidase family of proteins. Hephaestin, a copper-dependant ferroxidase protein, is crucial for iron exiting intestinal enterocytes into the circulation. It mediates the movement of iron across the basolateral membrane in conjunction with ferroportin 1. This is an important link between iron and copper metabolism in mammalian systems, as copper deficiency leads to reduced hephaestin and reduced iron absorption resulting in anemia. Hephaestin can bind six copper ions per monomer and is regulated by the homeobox transcription factor CDX2. Increased levels of iron leads to an increase in CDX2 expression and thus Hephaestin. Hephaestin is primarily detected in the intestine, but is also expressed in colon, breast, bone trabecural cells and fibroblasts.

The MAC forms upon complement system activation by invading pathogenic bacteria and consists of the four major complement proteins: C5b, C6, C7 and C8. These complement proteins bind to the outer surface of the plasma membrane of the invading cell. C9 binds to the membrane associated C5b-8 protein, which leads to the circular polymerisation of 12-18 C9 molecules. These polymerised C9 molecules form a ring structure in the membrane. Molecules can then diffuse freely through this transmembrane channel, causing cell lysis and destruction of the invading bacterial cell.

Involved in a variety of functions, protocadherins help to regulate neural development and synapse formation. PCDH1 is a 1,026 amino acid single-pass type I membrane protein that contains seven cadherin domains and is a member of the protocadherin family. Localized to cell-cell and cell-matrix boundaries and expressed at high levels in brain and neuro-glial cells, PCDH1 is thought to be involved in cell adhesion and cell-cell interactions and may play a role in neuronal development. PCDH1 contains a C-terminal cytoplasmic region, an extracellular region and a transmembrane region, and is expressed as two isoforms due to alternative splicing events.

ADCK2 (aarF domain containing kinase 2), also known as AARF, is a 626 amino acid single-pass membrane protein belonging to the protein kinase superfamily and the ADCK protein kinase family. The ADCK family consists of five paralogs in human (ADCK1-5). Encoded by a gene that maps to human chromosome 7q34, ADCK2 contains one protein kinase domain. ADCK2 participates in ATP and nucleotide binding, transferase functions and protein serine/threonine kinase activities. Expression of ADCK2 inversely correlates with cellular viability, suggesting elevated expression of ADCK2 may be essential for tumour survival. Bloom’s syndrome is an autosomal recessive disorder characterized by pre- and post-natal growth deficiencies, sun sensitivity, immunodeficiency and a predisposition to various cancers. The gene responsible for Bloom’s syndrome, BLM, encodes a protein homologous to the RecQ helicase of E. coli and is mutated in most Bloom’s syndrome patients. One characteristic of Bloom’s syndrome is an increased frequency of sister chromatid exchange (SCE). BLM has been shown to unwind G4 DNA, and a failure of this function is thought to be responsible for the increased rate of SCE. BLM is known to be translocated to the nucleus, where its ATPase activity is stimulated by both single- and double-stranded DNA. Mutations in the yeast SGS1, a homolog of BLM, are known to cause mitotic hyperrecombination similiar to that observed in Bloom’s cells.

Members of the RTP (receptor transporter proteins) family have recently been discovered to influence bitter taste receptor expression in addition to inducing the expression of mammalian odorant receptors. RTP3 is a 232 amino acid single-pass type III membrane protein belonging to the TMEM7 family. Unlike other RTP proteins, RTP3 is not expressed in olfactory neurons but is expressed predominantly in liver. RTP3 is involved in the functional expression of bitter taste receptors and suppresses cell proliferation, and is also found in human circumvallate papillae and testis (regions where bitter taste receptors are expressed). The gene encoding RTP3 maps to human chromosome 3p21.31 within C3CER1 (chromosome 3 common eliminated region 1), which is frequently eliminated in chromosomal deletions of solid tumors.
