Description:
Gentamicin is widely used for treating tuberculosis and Gram-negative infections and is particularly useful in neonatal intensive care units. Fluorophore-tagged drugs, such as gentamicin-Texas Red (GTTR) conjugates, have been used to identify drug trafficking routes across the BLB and into hair cells, and specifically in the cochlea - the sensory organ responsible for hearing. Identifying the mechanisms involved in the intracochlear trafficking of ototoxic drugs to hair cells is of fundamental as well as clinical importance. Texas Red-gentamicin conjugate (GTTR) provides researchers a valuable tool to study the cellular uptake, kinetics and distribution of gentamicin through fluorescence techniques, such as fluorescence microscopy, fluorescence correlation spectroscopy, single molecule spectroscopy. The TR fluorophore can be conveniently excited from 500-600 nm, with an emission maximum centered around 610 nm. Acute nephrotoxicity and permanent ototoxicity are serious side-effects that increase patient morbidity, or induce permanent auditory and vestibular deficits. Several approaches have been used to study the intracellular mechanisms induced by ototoxic drugs. However, ototoxic drugs must first cross the blood-labyrinth barrier (BLB) before entering sensory hair cells to exert their cytotoxic effect that leads to hearing loss and deafness. The BLB is similar to the blood-brain barrier (BBB), and is composed of tight junction-coupled endothelial and epithelial barrier layers that transport nutrients and ions from one side of the barrier to the other.

Recognises a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. It does not cross-react with alpha (IgA), gamma (IgG), epsilon (IgE), or delta (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. IgM antibody is prominent in early immune responses to most antigens. Aberrant levels are associated with immune deficiency states, hereditary deficiencies, myeloma, Waldenstrom's macroglobulinemia, chronic infection and hepatocellular disease. This mAb is useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkins lymphomas. The most common feature of these malignancies is the restricted expression of a single heavy chain class. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.

Description:
It recognises an oncofetal glycoprotein with a single chain of 70 kDa, which is identified as alpha fetoprotein (AFP). This mAb is highly specific to AFP and shows no cross-reaction with other oncofetal antigens or serum albumin. The yolk sac and the liver produce AFP during fetal life. AFP expression in adults is often associated with hepatoma or teratoma. However, hereditary persistence of alpha-fetoprotein may also be found in individuals with no obvious pathology. The protein is thought to be the fetal counterpart of serum albumin, and the AFP and albumin genes are present in tandem in the same transcriptional orientation on chromosome 4. AFP is found in monomeric as well as dimeric and trimeric forms, and binds copper, nickel, fatty acids and bilirubin. The level of AFP in amniotic fluid is used to measure renal loss of protein to screen for spinal bifida and anencephaly.

The three mammalian isoforms of TGF-β, TGF-β1, β2, β3, signal through the same receptor and elicit similar biological responses. They are multifunctional cytokines that regulate cell proliferation, growth, differentiation and motility as well as synthesis and deposition of the extracellular matrix. They are involved in various physiological processes including embryogenesis, tissue remodeling and would healing. They are secreted predominantly as latent complexes which are stored at the cell surface and in the extracellular matrix. The release of biologically active TGF-β isoform from a latent complex involves proteolytic processing of the complex and /or induction of conformational changes by proteins such as thrombospondin-1. The physiological role of TGF-β3 is still unknown but its expression pattern suggests a role in the regulation of certain development processes. Recognises a single chain glycoprotein of 70 kDa, identified as CD55 (also known as decay accelerating factor, DAF). This mAb was clustered in Kobe at the Sixth International Workshop on Human Leukocyte Differentiation Antigens as F429D-9 (N-L120). CD55/DAF is widely expressed on cells throughout the body including leukocytes, erythrocytes, epithelium, endothelium, and fibroblasts. It is a Glycosyl phosphatidylinositol anchored (GPI-anchored) member of the membrane bound complement regulatory proteins that inhibit autologous complement cascade activation. It prevents the amplification steps of the complement cascade by interfering with the assembly of the C3-convertases, C4b2a and C3bBb, and the C5-convertase, C4b2a3b and C3bBb3b. CD55 also serves as receptor for CD97 and for echovirus and Coxsackie B virus. Anti-CD55 can be used as marker for paroxysmal nocturnal hemoglobinuria (PNH).

IgM is the first antibody generated in an immune response to an antigen. It is generally a pentamer with each of the five immunoglobulins linked together with disulfide bonds. In its pentamer form, it has a molecular mass of 970 kDa and 10 antigen binding sites (due to the large size of most antigens, not all binding sites can be filled simultaneously. IgM antibodies account for approximately 5%-10% of all the antibody in the body.<p>This mAb is specific for the human IgM heavy chain. It does not cross-react with other immunoglobulin heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. IgM antibody is useful in the identification of certain cancer types. Some tumors express a single heavy chain class. Demonstration of clonality in lymphoid infiltrates indicates that the infiltrate is clonal and therefore malignant.
