This MAb reacts specifically with heat shock protein HSP27 in human and monkey tissues and cell lines such as MCF-7. HSP27, also referred to as the Estrogen-Regulated 24K protein and HSP28, is one of several small heat shock proteins produced by all organisms studied. HSP27 synthesis is induced by elevated temperature, as well as by estrogen in hormone responsive cells. Interestingly, human HSP27 also shares greater than 50% homology with low molecular weight Drosophila HSPs and mammalian α-crystalline lens protein. Because of the estrogen responsive nature of HSP27, this protein has been studied extensively in human estrogen responsive tissues such as cervix, endometrium and breast tissue. Therefore HSP27 may be useful in classifying various hormone sensitive tumors.

The supermix includes electrophoresis tracking dyes that migrate at approximately 4 kb and 50 bp to allow direct loading of PCR product on agarose gels following amplification. AccuStart™ II <i>Taq</i> DNA polymerase in the master mix is inactivated with monoclonal antibodies that bind the polymerase and keep it inactive prior to the initial PCR denaturation step. Upon heat activation (1 minute at 94 °C), the antibodies denature irreversibly, releasing fully active, unmodified <i>Taq</i> DNA polymerase. La comparaison de la couleur de la solution avec l'échelle colorimétrique confirme ou infirme la propreté d'une surface. Il suffit de frotter l'écouvillon humide sur une surface avant de l'insérer dans le tube puis de briser le flacon de verre contenu dans le tube à l'aide d'une pince. À la fin du test, le tube peut être évacué avec les déchets courants à température ambiante.
