Applications:
Urea is used for the denaturation of proteins and as a mild solubilization agent for insoluble or denatured proteins. Useful for renaturing proteins from samples already denatured with 6 M guanidine chloride such as inclusion bodies and in the extraction of the mitochondrial complex. It is commonly used to solubilize and denature proteins for denaturing isoelectric focusing and two-dimensional electrophoresis and in acetic acid-urea PAGE gels. May be used with guanidine hydrochloride and dithiothreitrol (DTT) in the refolding of denatured proteins into their native or active form. Urea is used in cell or tissue culture media to increase the osmolality. Urea has also been used as fertilizer because of the easy availability of nitrogen; in animal feeds; it is reacted with aldehydes to make resins and plastics; condensed with malonic ester to form barbituric acid; used in the paper industry to soften cellulose. Biochem/physiol Actions: Urea has been shown to act as an aldosterone antagonist in the development of peanut agglutinin binding in cultured embryonic renal collecting duct epithelial cells. Urea is the principal end product of nitrogen metabolism in most mammals, formed by the enzymatic reactions of the Kreb's cycle. Typical Working Concentration: The use of 2 g/L urea in the culture of Kluyveromyces marxianus to produce a thermostable extracellular lipase has been described. Urea is typically used at a concentration of 8 M for protein denaturation or solubilization. A final concentration of 5 M urea is commonly used in molecular biology for sequencing gels. To prevent carbamylation, do not heat urea containing buffers above 37 °C

This MAb recognizes a 21 kDa protein, identified as the p21WAF1 tumor suppressor protein. This MAb is highly specific to p21 and shows no cross-reaction with other closely related mitotic inhibitors. p21WAF1 is a specific inhibitor of cdk's and a tumor suppressor involved in the pathogenesis of a variety of malignancies. The expression of this gene acts as an inhibitor of the cell cycle during G1 phase and is tightly controlled by the tumor suppressor protein p53. Its expression is induced by the wild type, but not mutant, p53 suppressor protein. Normal cells generally display a rather intense nuclear p21 expression. Loss of p21 expression has been reported in many carcinomas (gastric carcinoma, non-small cell lung carcinoma, thyroid carcinoma).

Cell cycle progression is controlled in part by a family of cyclin proteins and cyclin-dependent kinases (Cdks). Cdk proteins work in concert with the cyclins to phosphorylate key substrates involved in each phase of cell cycle progression. Another family of proteins, Cdk inhibitors, also plays a role in regulating the cell cycle by binding to cyclin-Cdk complexes and modulating their activity. CDKL5 (cyclin-dependent kinase-like 5) is a 1030 amino acid protein that belongs to the CMGC Ser/Thr protein kinase family. Expressed in brain, lung, kidney, prostate, ovary, placenta, pancreas and testis, CDKL5 is thought to play a role in cell cycle regulation. Defects in CDKL5 are a cause of several disorders, such as X-linked infantile spasm syndrome and Rett syndrome.

Lipocalin-1 is a secretory protein that is highly expressed in fluids covering epithelial surfaces such as tears and respiratory secretions. This major lipid-binding protein in tears is also called tear lipocalin (TL) and von Ebner’s gland protein (VEG), as it is also a major secretion of these lingual salivary glands. In addition to lacrimal glands and lingual glands, Lipocalin-1 is secreted by nasal mucosal glands, secretory glands of the tracheobronchial tract, sweat glands, mammary glands, adrenal gland, prostate, thymus, testis and corticotrophs of the pituitary gland. Specifically, Lipocalin-1 functions to stabilize the lipid film of human tear fluid by removing harmful lipids from the human corneal surface and delivering them to the aqueous phase of tears. Lipocalin-1 may also function as a transporter of hydrophobic molecules such as bitter substances on the tongue.

Fibulin-1 expression is stimulated by estrogen in ovarian cancer cell lines and has been suggested as both an agent of metastasis in ovarian cancer cells and as an indicator for predicting cancer risk or aggressiveness in ovarian carcinomas. The mobility of cancer cells may be inhibited with increasing exposure to Fibulin-1. Fibulin-2 binds to the lectin domains of extracellular matrix proteins aggrecan, versican and brevican. Fibulin-2 is abundantly expressed in heart, placenta and ovarian tissue, where it localizes to basement membranes and connective tissue compartments. In mice, differential Fibulin-2 gene expression correlates with the early phase of diabetic kidneys and glomerulosclerosis. The gene encoding human Fibulin-2 maps to chromosome 3p25.1.

Transcription regulator involved in inner cell mass and embryonic stem (ES) cells proliferation and self-renewal. Imposes pluripotency on ES cells and prevents their differentiation towards extraembryonic endoderm and trophectoderm lineages. Blocks bone morphogenetic protein-induced mesoderm differentiation of ES cells by physically interacting with SMAD1 and interfering with the recruitment of coactivators to the active SMAD transcriptional complexes. Acts as a transcriptional activator or repressor. Binds optimally to the DNA consensus sequence 5'-TAAT[GT][GT]-3' or 5'-[CG][GA][CG]C[GC]ATTAN[GC]-3'. Able to autorepress its expression in differentiating (ES) cells: binds to its own promoter following interaction with ZNF281/ZFP281, leading to recruitment of the NuRD complex and subsequent repression of expression. When overexpressed, promotes cells to enter into S phase and proliferation.
