ZAP70 is a 70 kDa protein tyrosine kinase found in T-cells and natural killer cells. Control of this protein translation is via the IgVH gene.In Western blotting of whole cell lysates of normal peripheral blood mononuclear cells, the antibody labels a band corresponding to ZAP70. In Western blotting of whole cell lysates of CD19-positive purified leukemia cells from patients with Ig-unmutated and Ig-mutated CLL, the antibody labels a band corresponding to ZAP70 in the Ig-unmutated CLL samples, whereas no band is observed in the Ig-mutated CLL samples. In Western blotting of cell lysates of Jurkat cells (T-lymphoblastic cell line), the antibody labels a band of 70 kDa protein. In Western blotting of cell lysates of A431 cells (carcinoma cell line), no band is observed. ZAP70 protein is expressed in leukemic cells of approximately 25% of chronic lymphocytic leukemia (CLL) cases as well. Anti-ZAP70 expression is an excellent surrogate marker for the distinction between the Ig-mutated (anti-ZAP70 negative) and Ig-unmutated (anti-ZAP70 positive) CLL subtypes and can identify patient groups with divergent clinical courses. The anti-ZAP70 positive Ig-unmutated CLL cases have been shown to have a poorer prognosis.

Proliferation Marker Ki67 antigen is the prototypic cell cycle related nuclear protein, expressed by proliferating cells in all phases of the active cell cycle (G1, S, G2 and M phase). It is absent in resting (G0) cells. Ki67 are useful in establishing the cell growing fraction in neoplasms (immunohistochemically quantified by determining the number of Ki67 positive cells among the total number of resting cells = Ki67 index). In neoplastic tissues the prognostic value is comparable to the tritiated thymidine labelling index. The correlation between low Ki67 index and histologically low grade tumours is strong. Ki67 is routinely used as a neuronal marker of cell cycling and proliferation.

Induces susceptibility to atherosclerosis (By similarity). Cell adhesion molecule which is required for leukocyte transendothelial migration (TEM) under most inflammatory conditions. Tyr-69 plays a critical role in TEM and is required for efficient trafficking of PECAM1 to and from the lateral border recycling compartment (LBRC) and is also essential for the LBRC membrane to be targeted around migrating leukocytes. Prevents phagocyte ingestion of closely apposed viable cells by transmitting 'detachment' signals, and changes function on apoptosis, promoting tethering of dying cells to phagocytes (the encounter of a viable cell with a phagocyte via the homophilic interaction of PECAM1 on both cell surfaces leads to the viable cell's active repulsion from the phagocyte. During apoptosis, the inside-out signaling of PECAM1 is somehow disabled so that the apoptotic cell does not actively reject the phagocyte anymore. The lack of this repulsion signal together with the interaction of the eat-me signals and their respective receptors causes the attachment of the apoptotic cell to the phagocyte, thus triggering the process of engulfment). Isoform Delta15 is unable to protect against apoptosis. Modulates BDKRB2 activation. Regulates bradykinin- and hyperosmotic shock-induced ERK1/2 activation in human umbilical cord vein cells (HUVEC).

The HB7 antibody is specific for human CD38, a 45 kDa type II transmembrane glycoprotein expressed on thymocytes, plasma cells, and monocytes as well as other non-hematopoietic cells. T and B lymphocyte expression is discontinuous and varies based on differentiation state � present on immature cells, low on intermediate stages, and high on activated mature cells. It is found on most CD34+ cells, but not on pluripotent stem cells. CD38 is an ectoenzyme that functions to catalyze the synthesis and hydrolysis of cyclic ADP-ribose and is involved in cell signaling, regulating cell adhesion, and activation. Additionally, CD38 has been shown to be a prognostic marker for some leukemias and other diseases.

Recognizes a protein of 74 kDa, identified as CD84. It is expressed on mature B cells and on B-cell lines, including pre-B-cell lines, but not on plasma cell lines. Immunohistochemical studies demonstrated that CD84 strongly expressed on tissues macrophages. CD84 is also highly expressed on platelets and, at low levels on peripheral blood T cells. It is a highly N-glycosylated protein and belongs to immunoglobulin superfamily. It may play a role in leukocyte activation.

It can be used to stain the nuclei in cell or tissue preparations and can be used as a nuclear marker in subcellular fractions. It produces a speckled pattern in normal and malignant cells and may be used to stain the nuclei of cells in fixed or frozen tissue sections.
