Description:
The membranes of myelinating Schwann cells are joined by tight, gap and adherens junctions, all of which are found in regions of noncompact myelin: the paranodal loops, incisures of Schmidt-Lanterman and mesaxons. Tight junctions help establish polarity in mammalian epithelia by forming a physical barrier that separates apical and basolateral membranes. Pals-associated tight junction protein (PATJ), the human homolog of Drosophila Discs Lost, is differentially localized in myelinating Schwann cells. PATJ associates with Claudin-1, CRB1 (a transmembrane protein that plays a role in epithelial cell polarity and photoreceptor development), and Pals1 (a Lin-7 associated protein). The PATJ/Pals1/CRB1 complex can form a tripartite tight junction in epithelial cells crucial to their integrity.

There are three protocadherin gene clusters, designated alpha, beta and gamma, all of which contain multiple tandemly arranged genes. PCDH17 is a 1159 amino acid single-pass type I membrane protein that contains six cadherin domains. Expressed as multiple alternatively spliced isoforms, PCDH17 is thought to function as a calcium-dependent cell adhesion protein that may play a role in establishing cell-cell connections within brain tissue. The gene encoding PCDH17 maps to human chromosome 13, which houses over 400 genes, such as BRCA2 and RB1, and comprises nearly 4% of the human genome.

Recognizes a protein of 210-220 kDa, which is identified as the complement receptor 1 (CR1)/CD35. This MAb is specific for a site in CR1 that is distal from the C3b-binding site, so that it is unable to block CR1 activity. This MAb is highly specific to CR1 and shows no cross-reaction with CR2. The primary function of CR1 is to serve as the cellular receptor for C3b and C4b, the most important components of the complement system leading to clearance of foreign macromolecules. Recognizes the CD45 leukocyte common antigen (LCA) family which is comprised of at least four isoforms of membrane glycoproteins (220, 205, 190, 180 kDa) expressed on hematopoietic cell lines but absent on non-hematopoietic cell lines, normal and malignant non-hematopoietic tissues. The intracellular portions of these molecules have protein phosphatase activity and are involved in regulation of transmembrane signals. Antibody to CD45 is useful in differential diagnosis of lymphoid tumors from non-hematopoietic undifferentiated neoplasms. A positive result with this MAb is highly indicative of lymphoid or myeloid origin. Certain types of lymphoid neoplasms may lack CD45 (Hodgkin lymphoma, some T-cell lymphomas, and some leukemias) so its absence does not rule out a hematolymphoid tumor. This antibody is expressed almost exclusively by cells of hematopoietic lineage and is present in most benign and malignant lymphocytes as well as plasma cell precursors.

Recognizes a protein, which is identified as HGAL. It contains a putative PDZ-interacting domain, an immunoreceptor tyrosine-based activation motif (ITAM), and two putative SH2 binding sites. In B cells, its expression is specifically induced by interleukin-4. HGAL is specifically expressed in germinal center B-cells, but is absent in mantle and marginal zone B-cells and in the inter-follicular and para-cortical regions in normal tonsils and lymph nodes. Its high degree of specificity for germinal center B-cells makes anti-HGAL an ideal marker for the detection of germinal center-derived B-cell lymphomas. HGAL expression has been used to help elucidate nodal marginal zone lymphoma (NMZL) from cases of diffuse follicle center lymphoma. Additionally, HGAL expression was shown to correlate with survival in patients with diffuse large B-cell lymphoma (DLBCL).
